Bis(thiosemicarbazonato) complexes Cu(II)(Btsc) have attracted interest as promising metallodrugs and, in particular, as copper radiopharmaceuticals. Prototypes Cu(Atsm) and Cu(Gtsm) are membrane-permeable, but their metabolisms in cells are distinctly different: copper that is delivered by Cu(Gtsm) is trapped nonselectively in all cells, whereas copper that is delivered by Cu(Atsm) is retained selectively in hypoxic cells but is "washed out" readily in normal cells. We have studied copper-transfer reactions of these two complexes under various conditions, aiming to model their cellular chemistry. In Me2SO, both complexes exhibited reversible one-electron-reduction processes with Cu(Atsm) being more difficult to reduce than Cu(Gtsm) (E(1/2)'=-0.60 and -0.44 V, respectively, vs AgCl/Ag). Upon introduction of an aqueous buffer into Me2SO, the electrochemical reduction remained chemically reversible for Cu(Atsm) but became irreversible for Cu(Gtsm). However, the estimated difference in their reduction potentials did not change. Chromophoric ligand anions bicinchonate (Bca) and bathocuproine disulfonate (Bcs) were used as Cu(I) indicators to trace the destinations of copper in the reactions and to mimic cellular Cu(I)-binding components ("sinks"). While both BtscH2 ligands have high affinities for Cu(I) (KD in the picomolar range), they cannot compete with Cu(I) sinks such as the copper-binding proteins Atx1 and Ctr1c (or a mimic such as Bcs). In the presence of these proteins, reduction of Cu(II)(Btsc) leads to irreversible transfer of copper to the protein ligands. Endogenous reductants ascorbate and glutathione can reduce Cu(II)(Gtsm) in the presence of such protein ligands but cannot reduce Cu(II)(Atsm). These properties establish a strong correlation between the contrasting cellular retention properties of these complexes and their different reduction potentials. The endogenous reductants in normal cells appear to be able to reduce Cu(II)(Gtsm) but not Cu(II)(Atsm), allowing the latter to be washed out. The more reducing environment of hypoxic cells leads to reduction of Cu(II)(Atsm) and retention of its copper.

HMA2, HMA4, and HMA7 are three of the eight heavy metal transporting P(1B)-type ATPases in the simple plant Arabidopsis thaliana. The first two transport Zn(2+), and the third transports Cu(+). Each protein contains soluble N-terminal metal-binding domains (MBDs) that are essential for metal transport. While the MBD of HMA7 features a CxxC sequence motif characteristic of Cu(I) binding sites, those of HMA2 and HMA4 contain a CCxxE motif, unique for plant Zn(2+)-ATPases. The three MBDs HMA2n (residues 1-79), HMA4n (residues 1-96), and HMA7n (residues 56-127) and an HMA7/4n chimera were expressed in Escherichia coli. The chimera features the ICCTSE motif from HMA4n inserted in place of the native MTCAAC motif of HMA7n. Binding affinities for Zn(II) and Cu(I) of each MBD were determined by ligand competition with a number of chromophoric probes. The challenges of using these probes reliably were evaluated, and the relative affinities of the MBDs were verified by independent cross-checks. The affinities of HMA2n and HMA4n for Zn(II) are higher than that of HMA7n by a factor of 20-30, but the relative affinities for Cu(I) are inverted by a factor of 30-50. These relativities are consistent with their respective roles in metal selection and transportation. Chimera HMA7/4n binds Cu(I) with an affinity between those of HMA4n and HMA7n but binds Zn(II) more weakly than either parent protein does. The four MBDs bind Cu(I) more strongly than Zn(II) by factors of >10(6). It is apparent that the individual MBDs are not able to overcome the large thermodynamic preference for Cu(+) over Zn(2+). This information highlights the potential toxicity of Cu(+) in vivo and why copper sensor proteins are approximately 6 orders of magnitude more sensitive than zinc sensor proteins. Metal speciation must be controlled by multiple factors, including thermodynamics (affinity), kinetics (including protein-protein interactions), and compartmentalization. The structure of Zn(II)-bound HMA4n defined by NMR confirmed the predicted ferredoxin betaalphabetabetaalphabeta fold. A single Zn atom was modeled onto a metal-binding site with protein ligands comprising the two thiolates and the carboxylate of the CCxxE motif. The observed (113)Cd chemical shift in [(113)Cd]HMA4n was consistent with a Cd(II)S(2)OX (X = O or N) coordination sphere. The Zn(II) form of the Cu(I) transporter HMA7n is a monomer in solution but crystallized as a polymeric chain [(Zn(II)-HMA7n)(m)]. Each Zn(II) ion occupied a distorted tetrahedral site formed from two Cys ligands of the CxxC motif of one HMA7n molecule and the amino N and carbonyl O atoms of the N-terminal methionine of another.

We present a unique matter-wave interferometer whose phase scales with the cube of the time the atom spends in the interferometer. Our scheme is based on a full-loop Stern-Gerlach interferometer incorporating four magnetic field gradient pulses to create a state-dependent force. In contrast to typical atom interferometers which make use of laser light for the splitting and recombination of the wave packets, this realization uses no light and can therefore serve as a high-precision surface probe at very close distances.

The reaction of K28Li5H7[P8W48O184].92H2O with early lanthanides under hydrothermal and conventional conditions yields novel structures of the molecular formula Ln4(H2O)28K6Li7[K subsetP8W48O184(H4W4O12)2Ln2(H2O)10] congruent with 57H2O, Ln = La (1), Ce (2, 2a), Pr (3), Nd (4), in which the central cavity of the precursor anion is occupied by lanthanide cations and H4W4O12 moieties. The new heteropolyanions were characterized by elemental analysis, infrared spectroscopy, 31P NMR, and X-ray crystallography. We study a heavy-heavy-light three-body system confined to one space dimension. Both binding energies and corresponding wave functions are obtained for (i) the zero-range, and (ii) two finiterange attractive heavy-light interaction potentials. In case of the zero-range potential, we apply the method of Skorniakov and Ter-Martirosian to explore the accuracy of the Born-Oppenheimer approach. For the finite-range potentials, we solve the Schrödinger equation numerically using a pseudospectral method. We demonstrate that when the two-body ground state energy approaches zero, the three-body bound states display a universal behavior, independent of the shape of the interaction potential.

The quantum mechanical propagator of a massive particle in a linear gravitational potential derived already in 1927 by Earle H. Kennard [2, 3] contains a phase that scales with the third power of the time T during which the particle experiences the corresponding force. Since in conventional atom interferometers the internal atomic states are all exposed to the same acceleration a, this T 3 -phase cancels out and the interferometer phase scales as T 2 . In contrast, by applying an external magnetic field we prepare two different accelerations a 1 and a 2 for two internal states of the atom, which translate themselves into two different cubic phases and the resulting interferometer phase scales as T 3 . We present the theoretical background for, and summarize our progress towards experimentally realizing such a novel atom interferometer.
