MicroRNAs (miRNAs) are noncoding RNAs that play an important role in the regulation of the genetic expression in animals and plants by targeting mRNAs for cleavage or translational repression. Several miRNAs regulate the plant development, the metabolism, and the responses to biotic and abiotic stresses. Characterization of an miRNA has helped to show its role in fine tuning the mechanisms of posttranscriptional gene regulation. Although there is a lot of information related to miRNA regulation of some processes, the role of miRNA involved in the regulation of biosynthesis of secondary plant product is still poorly understood. In this chapter, we summarize the identification and characterization of miRNAs that participate in the regulation of the biosynthesis of secondary metabolites in plants and their use in the strategies to manipulate a controlled manipulation.
Los trigos harineros y cristalinos son importantes en México por su consumo e impacto económico. Actualmente el país tiene un déficit en la producción de trigo harinero y un superávit del cristalino. Para lograr su autosuficiencia alimentaria y aumentar la productividad es necesario desarrollar variedades de mayor rendimiento e identificar sus mejores condiciones de producción. Con el propósito de determinar la especie más productiva y mejor adaptada a las regiones productoras de México bajo diferentes condiciones de riego y fechas de siembra, se diseñó un experimento con ocho genotipos cristalinos (Triticum durum Desf.) y ocho harineros (Triticum aestivum L.) sembrados en los ciclos otoño invierno 2012-2013 y 2013-2014 en 30 ambientes del Noroeste, Norte y El Bajío, como parte de los Ensayos Nacionales de Trigo de Riego (ENTRI) del Instituto Nacional de Investigaciones Forestales, Agrícolas y Pecuarias. En un primer análisis se utilizaron sólo los ambientes con fecha normal, un total de 22, y se agruparon en: 1) todos los ambientes, 2) ambientes de riego completo, 3) riego reducido, 4, 5 y 6) por región y 7, 8, 9 y 10) nivel de riego por región en el Noroeste y El Bajío. Los ocho ambientes restantes permitieron evaluar el efecto fecha de siembra. Los resultados indicaron que el rendimiento de grano varió significativamente entre regiones obteniéndose los más altos en el Noroeste, seguidos por el Norte y El Bajío. El rendimiento de trigos cristalinos fue significativamente superior al de los harineros en la mitad de los grupos de ambientes. Las mayores diferencias en rendimiento se observaron en condiciones de riego completo, donde los cristalinos fueron superiores. En riego reducido, los rendimientos de ambas especies fueron similares. El efecto fecha de siembra, de normal a tardía, afectó negativamente a ambas especies, aunque en mayor magnitud a los trigos cristalinos. Las variedades de reciente liberación tienen mayor potencial productivo que las liberadas anteriormente.
RESUMENLos objetivos de este estudio fueron identificar las especies de Fusarium causantes de pudrición de raíz en trigos producidos en el Bajío de México, además de determinar la respuesta de 28 variedades de trigo a la enfermedad. En 2014 y 2015 se obtuvieron aislados de Fusarium spp. a partir de muestras de trigo con síntomas de fusariosis. Los aislados fúngicos se identificaron mediante caracterización morfológica y análisis de secuencias ITS y TEF-1α, así como el uso de iniciadores específicos para la detección de especies de Fusarium. Un aislado representativo de cada especie de Fusarium obtenida se incrementó para verificar su patogenicidad a través de la inmersión de semillas de 28 variedades de trigo en una suspensión de conidios. Los resultados de la caracterización morfológica y análisis de secuencias, así como las pruebas de patogenicidad, indicaron que Fusarium proliferatum y F. graminearum son las principales especies causantes de la pudrición de raíz en trigo cultivado en el Bajío de México. Entretanto, la respuesta a la inoculación indicó que las variedades Gálvez M87, Castrejón F97 y Maya S2007 fueron las más tolerantes a la enfermedad, mientras que Monarca F2007 y Batán F96 fueron las variedades más susceptibles.Palabras clave: Fusarium, trigo, Triticum aestivum, Fusarium proliferatum, Fusarium graminearum, microscopía electrónica de barrido.
ABSTRACTThe objectives of this study were to identify Fusarium species causing root rot in wheat produced in the Bajío of Mexico, and to determine the response of 28 wheat varieties to this disease. Fusarium spp. isolates were obtained from samples with root rot symptoms in 2014 and 2015. The fungal isolates were identified by morphological characterization and sequence analysis of ITS and TEF-1α, and also using specific primers for the detection of Fusarium species. Pathogenicity of a representative isolate of each Fusarium species was verified by immersion of seeds of 28 wheat varieties in a conidial suspension. Tobacco mosaic virus is the etiological agent of one of the most critical diseases limiting chili pepper production. Various practices have been used to manage the disease, e.g., the use of resistant varieties and interference with the vector through chemical control. However, these practices are not helpful once the virus has been established in the plant. There is still no effective method for the sustainable management of the disease; therefore, exploring new options is required. Currently, some studies have reported the activity of TiO2 NPs against viruses in plants, although not against TMV in chili pepper. The present work aims to determine a possible direct action of TiO2 NPs against TMV and if there is a relationship between the amount of virus and symptoms. The application of TiO2 NPs at 150 μg/mL in infected pepper plants reduced symptoms and viral load and improved the morphological characteristics compared to the control. Incubation of 150 µg/mL TiO2 NPs with the virus for 6 and 8 h before infection decreased viral concentration significantly after infection compared to the control. In this work, it is reported, for the first time, that the use of TiO2 NPs is a novel practice for the control of TMV in chili pepper.
