Marfan syndrome is an extracellular matrix disorder with cardinal manifestations in the eye, skeleton and cardiovascular systems associated with defects in the gene encoding fibrillin (FBN1) at 15q21.1 (ref. 1). A second type of the disorder (Marfan syndrome type 2; OMIM 154705) is associated with a second locus, MFS2, at 3p25-p24.2 in a large French family (family MS1). Identification of a 3p24.1 chromosomal breakpoint disrupting the gene encoding TGF-beta receptor 2 (TGFBR2) in a Japanese individual with Marfan syndrome led us to consider TGFBR2 as the gene underlying association with Marfan syndrome at the MSF2 locus. The mutation 1524G-->A in TGFBR2 (causing the synonymous amino acid substitution Q508Q) resulted in abnormal splicing and segregated with MFS2 in family MS1. We identified three other missense mutations in four unrelated probands, which led to loss of function of TGF-beta signaling activity on extracellular matrix formation. These results show that heterozygous mutations in TGFBR2, a putative tumor-suppressor gene implicated in several malignancies, are also associated with inherited connective-tissue disorders.

Six new monoclonal antibodies (TDM-2, TDM-3, 64M-2, 64M-3, 64M-4 and 64M-5) specific for ultraviolet (UV) induced DNA damage have been established. In the antibody characterization experiments, two TDM antibodies were found to show a dose-dependent binding to UV-irradiated DNA (UV-DNA), decrease of binding to UV-DNA after cyclobutane pyrimidine dimer photoreactivation, binding to DNA containing cyclobutane thymine dimers, and unchanged binding to UV-DNA after photoisomerization of (6-4)photoproducts to Dewar photoproducts. These results indicated that the epitope of TDM monoclonal antibodies was the cyclobutane pyrimidine dimer in DNA. On the other hand, four 64M antibodies were found to show a dose-dependent binding to UV-DNA, unchanged binding to UV-DNA after cyclobutane pyrimidine dimer photoreactivation, undetectable binding to DNA containing thymine dimers, and decrease of binding to UV-DNA after photoisomerization of (6-4)photoproducts. These results indicated that the epitope of 64M antibodies was the (6-4)photoproduct in DNA. This is the first report of the simultaneous establishment of monoclonal antibodies against the two different types of photolesions from the same mouse. By using these monoclonal antibodies, we have succeeded in measuring both cyclobutane pyrimidine dimers and (6-4)photoproducts in the DNA from human primary cells irradiated with physiological UV doses.

Human earwax consists of wet and dry types. Dry earwax is frequent in East Asians, whereas wet earwax is common in other populations. Here we show that a SNP, 538G --> A (rs17822931), in the ABCC11 gene is responsible for determination of earwax type. The AA genotype corresponds to dry earwax, and GA and GG to wet type. A 27-bp deletion in ABCC11 exon 29 was also found in a few individuals of Asian ancestry. A functional assay demonstrated that cells with allele A show a lower excretory activity for cGMP than those with allele G. The allele A frequency shows a north-south and east-west downward geographical gradient; worldwide, it is highest in Chinese and Koreans, and a common dry-type haplotype is retained among various ethnic populations. These suggest that the allele A arose in northeast Asia and thereafter spread through the world. The 538G --> A SNP is the first example of DNA polymorphism determining a visible genetic trait.

Cyclobutane pyrimidine dimers (CPDs) and pyrimidine (6-4) pyrimidone photoproducts ((6-4)photoproducts) are the two major classes of cytotoxic, mutagenic and carcinogenic DNA photoproducts produced by ultraviolet light irradiation of cells. The phenomenon of photoreactivation, the reduction of the lethal and mutagenic effects of ultraviolet radiation by simultaneous or subsequent irradiation with near ultraviolet or visible light, has been identified in several organisms and in some cases the enzymes that catalyse this process have been characterized in sufficient detail. CPDs are the only known substrate for the photoreactivating enzymes so far analysed and enzymatic photoreactivation of (6-4)photoproducts has not yet been reported. We report here that an enzyme that catalyses the light-dependent repair of (6-4)photoproduct exists in Drosophila melanogaster. This is, to our knowledge, the first report of such photoreactivating activity specific for (6-4)photoproducts in any organism.

Sumoylation is involved in mediating protein±protein interactions, subcellular compartmentalization and protein stability. Our analysis of various Wnt signaling molecules revealed that one of them, Tcf-4, is sumoylated at the endogenous level. At least one sumoylation site, Lys297, of Tcf-4 was identi®ed. The sumoylation of Tcf-4 was enhanced by PIASy, a SUMO E3 enzyme, and inhibited by Axam, a desumoylation enzyme. Although PIASy did not affect the interaction of Tcf-4 with b-catenin or DNA, Tcf-4, SUMO-1 and PIASy were co-localized in the nucleus and present in a complex in the PML body. PIASy enhanced b-catenin-dependent transcriptional activity of Tcf-4, whereas Axam inhibited it. Reduction of the protein level of Axam by RNA interference led to an increase in sumoylation of Tcf-4 and activation of Tcf-4. Furthermore, b-catenin and PIASy activated Tcf-4 K297R , in which Lys297 was changed to arginine, less than wild-type Tcf-4. These results suggest that sumoylation of Tcf-4 is involved in b-catenin-dependent and Tcf-4-mediated gene expression in the Wnt signaling pathway.

Cyclic carbonates are valuable compounds that have applications in a variety of chemical fields. Methodologies for the synthesis of cyclic carbonates are well investigated in recent years, and the most successful and popular procedure is the utilization of CO(2). This paper presents recent progress in the synthesis of cyclic carbonates by a CO(2)-fixation process, which involves novel palladium-catalyzed CO(2)-recycling reactions.

The reaction mechanism of the most commonly used anode material, Ni/yttria-stabilized zirconia ͑YSZ͒ cermets, in H 2 -H 2 O solid oxide fuel cells ͑SOFCs͒ was investigated. Because the reaction mechanism for the Ni/YSZ anode in H 2 has not been conclusively determined, we investigated the detailed dependence of dc polarization and interfacial conductivity of Ni/YSZ cermet anode on the partial pressure of hydrogen (p H 2 ). Based on our experimental results, we developed a model that links the chemical reactions on the anode with the electrical characteristics of the anode such as the dc polarization and the interfacial conductivity. In our model, we assumed competitive adsorption equilibrium of H 2 , H 2 O, and O on Ni surfaces at the three-phase boundary, and assumed the rate-determining step to be Langmuir-type reactions of H with O. In SOFCs with Ni/YSZ anodes, reported dependencies of the current and interfacial conductivity on p H and p H 2 O differ among previous studies. Both our measured dependencies and previously published dependencies were successfully reproduced by our Langmuir reaction model. Furthermore, possible reasons for these different observed dependencies include the different adsorption equilibrium constants.

The absorption coefficient of the dye used in dye-sensitized solar cells is a major factor in the total energy efficiency of the cell. In this work, we increased the absorption coefficient of the dye cis-(NCS) 2 bis(2,2′-bipyridyl-4,4′-dicarboxylate)ruthenium(II) used in such cells by having the dye adsorbed on silver islands. We studied the effect of the surface area of silver (per surface area of substrate) and the effect of dye concentration on this enhancement. This technique significantly enhanced the absorption coefficient of the Ru-dye for the metal-ligand charge-transfer transition, which generates electrons and holes in dye-sensitized solar cells. The enhancement ratio of the absorption was controlled by the amount of Ru-dye directly adsorbed on the silver islands. The absorption was enhanced by a factor of 149, which was achieved for a dye concentration of 3 × 10 -10 mol/cm 2 and an average surface area of silver island of 0.63 cm 2 /cm 2 of substrate. Such enhancement is expected to be useful in improving the electric energy conversion efficiency of dyesensitized solar cells.
