Highly sensitive and selective chloride liquid/polymeric membrane electrodes are described that employ [9]-mercuracarborand-3 (MC3), a neutral preorganized macrocyclic Lewis acid, as the anion carrier. MC3-based chloride-sensitive membrane electrodes, doped with different mole percentages of cationic additives (5, 10, and 60 mol % tridodecylmethylammonium chloride) relative to the amount of the carrier, exhibit enhanced potentiometric selectivity for chloride over other anions, including more lipophilic anions such as perchlorate, nitrate, and thiocyanate. In addition, the selectivity coefficients obtained are shown to meet the requirement for clinical applications. The obtained selectivity pattern is shown to correlate very well with 199Hg NMR titrations of MC3 with various anions, performed in organic solvents. Optimized membrane electrodes show a near-Nernstian response toward chloride over a wide concentration range and have micromolar detection limits. MC3-based chloride sensors show a fast response time (in the order of few seconds), as well as short recovery time. The developed mercuracarborand-based sensors do not practically respond to pH changes over the pH range of 2.5-7.0. Response characteristics (e.g., detection limit, linear range, response slope, and selectivity) of the [9]mercuracarborand-3 based chloride sensors remain essentially the same over a period of approximately 2 months, reflecting remarkable stability and well-defined chemistry of the macrocyclic Lewis acid ionophore.

A highly selective optical sensor for chloride, based on the multidentate Lewis acid ionophore [9]mercuracarborand-3, is described herein. This sensor is constructed by embedding the mercuracarborand ionophore, a suitable pH-sensitive lipophilic dye, and lipophilic cationic sites in a plasticized polymeric membrane. The multiple complementary interactions offered by the preorganized complexing cavity of [9]mercuracarborand-3 is shown to control the anion selectivity pattern of the optical film. The film exhibits a significantly enhanced selectivity for chloride over a variety of lipophilic anions such as perchlorate, nitrate, salicylate, and thiocyanate. Furthermore, the optical selectivity coefficients obtained for chloride over other biologically relevant anions are shown to meet the selectivity requirements for the determination of chloride in physiological fluids, unlike previously reported chloride optical sensors. In addition, the optical film responds to chloride reversibly over a wide dynamic range (16 microM-136 mM) with fast response and recovery times.

The measured 13 C chemical shifts of over forty carborane compounds correlate very well with ab-initio/ IGLO/NMR calculated values at both the DZ//3-21G and DZ//6-31G* (IGLO-NMR//Gaussian-geometry-optimized) levels of theory as well as with the ab-initio/GIAO/NMR values at the 6-31G*//6-31G* level of theory. For the carboranes in this study, the linear relationships δ( 13 C-IGLO-DZ//6-31G*) ) 0.941δ( 13 C-exp) -1.897 (r 2) 0.990) and δ( 13 C-GIAO-6-31G*//6-31G*) ) 0.893δ( 13 C-exp) -2.554 (r 2) 0.991) are derived. Combined together with recently reported 11 B NMR correlations between experiment and theory, a significant means is added to the arsenal of carborane NMR structure proof methods available to the experimentalist having access to only modest computational resources. And this procedure, of course, also has the additional feature of yielding reasonably good structural information (bond distances, angles, etc.).

Reactions of both closo-9,12-I2-1,2-C2B10H10 and closo-9,10-I2-1,7-C2B10H10 with an excess of aryl magnesium bromide in the presence of [PdCl2(PPh3)2] afford the corresponding closo-9,12-(4-R-C6H4)2-1,2-C2B10H10 [R=H (1), Me (2), OMe (3), SMe (4), N(CH3)2 (5), Cl (6)] and closo-9,10-(4-R-C6H4)2-1,7-C2B10H10 [R'=Me (7), OMe (8), N(CH3)2 (9), Cl (10), and -C[(OCH2)2]CH3 (11)] compounds in high yields. The anisole derivatives 3 and 8 were deprotected to yield the corresponding bis-phenols 12 and 13, respectively. Structural analyses of compounds 1, 3, 6, and 12 are reported. Re-etherification of compound 12 by using gamma-bromotriethyleneglycol methyl ether provided 14 (R=(CH2CH2O)3CH3). Oxidation of 4 with ceric(IV) ammonium nitrate (CAN) generated the bis-sulfoxide 15 (R=S(O)Me). Deprotection of compound 11 led to the corresponding acetyl derivative 18 (R'=C(O)Me). Bis-anisole 3 was tethered with 1,3-dibromopropane, 1,6-dibromohexane, 1,8-dibromooctane, 4,4'-bis(iodomethyl)-1,1'-biphenyl, and alpha,alpha'-dibromo-2,6-lutidine to afford the dimers 20b, 21b, 22b, 23b, and 24b, respectively. The tetrameric carboracycles 27a and 30a, as well as the dimeric 29c were obtained through repetitive coupling of the dimeric compounds 20b, 24b, and 22b with 1,3-dibromopropane, alpha,alpha'-dibromo-2,6-lutidine, and 1,8-dibromooctane, respectively. The tetrameric carboracycle 28a was obtained upon consecutive reactions of 1 with 1,4-dibromobutane. Hexameric carboracycle 28b was identified as a byproduct. Exhaustive ether cleavage of 27a generated octaphenol 31a. Re-etherification of 31a with trimethylenesultone provided the octasulfonate 32a, the first example of a water-soluble carboracycle. Linkage of dimer 23b with alpha,alpha'-dibromolutidine yielded the cyclic tetrameric tetrapyridyl derivative 30a in low yield. The structures of the carboracycles 27a, 28a, 28b, and 30a have been confirmed by Xray crystallography. In addition, the compounds 28a,b are the first reported carboracycles that interact with solvent molecules in a host-guest fashion.
