In favorable conditions Bacillus thuringiensis spores germinate and vegetative cells multiply, whereas in unfavorable conditions Bacillus thuringiensis sporulates and produces insecticidal crystal proteins. The development of B. thuringiensis strains was investigated in the larvae of insects belonging to the orders Lepidoptera and Diptera. Bacillus thuringiensis strains able to kill the insects did not always multiply in cadavers. Strains with no specificity to kill the insect sometimes multiplied when the insects were killed mechanically. These results indicate that some insect larvae represent an environment that favors the germination of B. thuringiensis spores and the multiplication of vegetative cells; however, there was no correlation between the toxin specificity and the specificity of the host.
Bacillus thuringiensis is a Gram positive, sporangial bacterium, known for its insecticidal habilities. Survival and conjugation ability of B. thuringiensis strains were investigated; vegetative cells were evaluated in non-sterile soil. Vegetative cells decreased rapidly in number, and after 48 hours the population was predominantly spores. No plasmid transfer was observed in non-sterile soil, probably because the cells died and the remaining cells sporulated quickly. Soil is not a favorable environment for B. thuringiensis multiplication and conjugation. The fate of purified B. thuringiensis toxin was analyzed by extractable toxin quantification using ELISA. The extractable toxin probably declined due to binding on surface-active particles in the soil.
Bacillus thuringiensis and Bacillus cereus belong to the B. cereus species group. The two species share substantial chromosomal similarity and differ mostly in their plasmid content. The phylogenetic relationship between these species remains a matter of debate. There is genetic exchange both within and between these species, and current evidence indicates that insects are a particularly suitable environment for the growth of and genetic exchange between these species. We investigated the conjugation efficiency of B. thuringiensis var. kurstaki KT0 (pHT73-Em) as a donor and a B. thuringiensis and several B. cereus strains as recipients; we used one-recipient and two-recipient conjugal transfer systems in vitro (broth and filter) and in Bombyx mori larvae, and assessed multiplication following conjugation between Bacillus strains. The B. thuringiensis KT0 strain did not show preference for genetic exchange with the B. thuringiensis recipient strain over that with the B. cereus recipient strains. However, B. thuringiensis strains germinated and multiplied more efficiently than B. cereus strains in insect larvae and only B. thuringiensis maintained complete spore germination for at least 24 h in B. mori larvae. These findings show that there is no positive association between bacterial multiplication efficiency and conjugation ability in infected insects for the used strains.
ABSTRACT:Bacillus has been widely studied and used for the control of pests and diseases. The adapted protocol proposed by POLANCZYK (2004) proved to be more efficient than the one by the World Health Organization (WHO, 1985) to isolate edaphic strains of Bacillus. However, it has not been assessed for isolation of endophytic strains, which are much less abundant in the nature and more difficult to be isolated. This study aimed to compare two methodological procedures for isolation of Bacillus, established by the WHO (1985) and by POLANCZYK (2004), regarding their efficiency for isolation of endophytics and edaphics Bacillus strains from inside the root tissue of sugarcane, as well as from the associated soil sample, collected from 11 locations; and to compare the density of bacteria in both environments. Endophytic and edaphic strains of Bacillus were isolated by both procedures. However, the isolation protocol performed by POLANCZYK (2004) made more efficient by having a greater number of colony forming units (CFU) per gram of soil and root indicating that this procedure is more useful, especially for isolation of endophytic strains of Bacillus, which are much less abundant in the nature than edaphic strains, being therefore more difficult to be isolated. Using the Polanczyk protocol (2004), Bacillus strains were recovered from all roots (endophytic) and soil (edaphic) samples of all the 11 fields, suggesting that the plant root may be another important source for isolation of Bacillus besides the soil. Higher densities of Bacillus were isolated from the edaphic environment compared with the endophytic environment, with significant differences when isolated by Polanczyk method (2004 (2004), quanto a sua eficiência para o isolamento de estirpes endofitas e edáficas de Bacillus originárias do interior do tecido radicular de cana-de-açúcar, bem como de amostras de solos associados, coletada de 11 locais; e comparar a densidade de bactérias em ambos os ambientes. As cepas endofíticas e edáficas de Bacillus foram isoladas por ambos os procedimentos. No entanto, o protocolo de isolamento realizado por POLANCZYK (2004) demonstrou-se mais eficiente por gerar maior número de unidades de formação de colônias (CFU) por grama de solo e raiz, indicando que esse procedimento é mais útil, especialmente para isolamento de estirpes endofíticas de Bacillus, que são muito menos abundantes na natureza do que as cepas edáficas, sendo, portanto, mais difíceis de serem isoladas. Usando o protocolo de POLANCZYK (2004), as cepas de Bacillus foram isoladas de todas as amostras de raízes (endofíticas) e de solo (edáficas) de todos os 11 campos, sugerindo que a raiz da planta pode ser outra fonte importante de isolamento de Bacillus além do solo. As densidades mais altas de Bacillus foram isoladas do ambiente edáfico em comparação com o ambiente endofítico, com diferenças significativas quando isoladas pelo método de POLANCZYK (2004). As plantas medicinais vêm sendo muito utilizadas para o tratamento, prevenção e cura de doenças, pois desde antigamente elas são usadas para fins medicinais pela humanidade. Por suas propriedades terapêuticas ou tóxicas conseguiram uma importância na medicina popular e convencional. Diante disso a presente pesquisa teve como objetivo avaliar através de predição in sílico os parâmetros toxicológicos de Melissa officinalis L. visando possível obtenção de novos candidatos a fármacos desta espécie. Foi um estudo básico, experimental, transversal, de natureza quantitativa realizado através de análise da toxicidade teórica com um software gratuito admetSAR. Para o composto acetil citrato de tributil, que para a maioria dos parâmetros não foi considerado tóxico, tem-se a possibilidade de ser utilizado de forma isolada como antifúngico, antibacteriano, anti-leshimania e inseticida, pois pertence ao componente do óleo essencial citral o qual faz parte majoritária presente na M. officinallis. Mas, é necessário que haja a continuidade de realização de trabalhos utilizando esta espécie cultivada na região sul do Tocantins, uma vez que foi verificado composto promissor para futuras investigações em testes in vitro e in vivo, podendo ser um forte candidato a novo fármaco.
Objetivo: Descrever a importância da atuação do enfermeiro no cuidado paliativo frente ao paciente oncológico. Métodos: Trata-se de uma revisão bibliográfica integrativa a coleta de dados foi realizada por meio de artigos atuais sobre o assunto estudado nos seguintes sites: SCIELO, Biblioteca Virtual da Saúde (BVS), Acervo+ Index Base e no mecanismo de busca do Google Acadêmico obedecendo marco temporal entre os anos de 2018 e 2022. Resultados: Os resultados apontam que o profissional enfermeiro possui um papel fundamental para prestar uma assistência humanizada estabelecendo uma relação com o paciente que minimize o sofrimento causado pela doença. Essa conexão começa a partir do diagnóstico até os cuidados paliativos, proporcionando uma melhor aceitação e ao mesmo tempo ofertando medidas de alívio ao sofrimento. Considerações finais: A profissional-paciente-família é um elo indispensável que se desenvolve através da comunicação interpessoal, entender o paciente nos mínimos detalhes, vendo-o de forma holística promovendo uma melhor qualidade de vida associado ao cuidado paliativo.
