PurposeTo evaluate ocular surface alterations in two populations at different exposure levels to particulate matter (PM) in their living and work environments.MethodsA cross-sectional study was conducted, including 78 volunteers from Argentina who lived and worked under different pollution levels in an urban (U; n = 44) or industrial zone (I; n = 34). Mean exposure level to PM was evaluated. Responses to the Ocular Symptom Disease Index and McMonnies questionnaire were obtained from all subjects. Subsequently, an assessment through the Schirmer I test (ST), slit lamp microscopy, vital staining, and tear breakup time was conducted. Statistical analyses with Chi-square and Bartlett's tests, as well as Student's t-tests and principal component analysis (PCA), were performed.ResultsParticles of size < 2.5 μm (PM2.5) level was significantly higher in the I group than the U group (P = 0.04). Ocular surface parameters including bulbar redness, eyelid redness, and the degree of vital staining with fluorescein (SF) and lissamine green (SLG) exhibited difference between the groups. With regards to the tear film, statistically significant differences in the ST value and meibomian gland dysfunction between the groups were detected (P = 0.003 and P = 0.02, respectively). Conjunctival SF and SLG, and ST values were identified as factors which could distinguish groups exposed to different PM levels.ConclusionSubjects exposed to higher levels of PM in the outdoor air presented greater ocular surface alterations. Thus, ST, SF, and SLG values could be used as convenient indicators of adverse health effects due to exposure to air pollution.
Eye color prediction based on an individual's genetic information is of interest in the field of forensic genetics. In recent years, researchers have studied different genes and markers associated with this externally visible characteristic and have developed methods for its prediction. The IrisPlex represents a validated tool for homogeneous populations, though its applicability in populations of mixed ancestry is limited, mainly regarding the prediction of intermediate eye colors. With the aim of validating the applicability of this system in an admixed population from Argentina (n = 302), we analyzed the six single nucleotide variants used in that multiplex for eye color and four additional SNPs, and evaluated its prediction ability. We also performed a genotype-phenotype association analysis. This system proved to be useful when dealing with the extreme ends of the eye color spectrum (blue and brown) but presented difficulties in determining the intermediate phenotypes (green), which were found in a large proportion of our population. We concluded that these genetic tools should be used with caution in admixed populations and that more studies are required in order to improve the prediction of intermediate phenotypes.
To identify the changes in the lipid profile of the tear film in two human populations exposed to different levels of particulate material, and its relationship with dry eye, by gas chromatography with mass spectrometry (GC-MS) detection. A panel study involving 78 volunteers, who live and work in two locations in Argentina with different pollution levels: urban zone (n = 44) and industrial zone (n = 34). We measured the mean levels of particulate matter (PM) exposure. The tear samples were analyze by gas GC-MS detection and the dry eye was diagnose using Schirmer test, fluorescein breakup time, vital staining with fluorescein and lissamine green, and lid parallel conjunctival folds (LIPCOF). Statistical analysis was performed using Chi-Square, Bartlett's, Mann-Whitney tests, and Multiple Correspondence Analysis. PM 10 level was significantly higher in industrial zone than in urban area (p < 0.05). Subjects exposed to higher levels of PM 10 in outdoor air presented more presence of fatty acids (FA) of long chain, a higher proportion of saturated fatty acids (SFA), and lower unsaturated fatty acids (UFA), showing a differentiated profile, which may be associated with a PM level. The incidence of dry eye was greater in the industrial zone (p < 0.001), showing in both populations for this pathology higher FA ω-6 levels, which are responsible for the inflammation process. The lipid profile in populations exposed to higher levels of PM 10 , like the industrial zone, shows a differentiated profile of FA and more incidence of dry eye with higher FA ω-6 levels, which are responsible for the inflammation process.
In order to estimate air quality at work environments from small and medium-sized enterprises (SMEs), we determined both the concentration of particulate matter (PM and PM) and the presence of polycyclic aromatic hydrocarbons (PAHs), as the heavy metals in the composition of the particulate matter. Three SMEs located in the city of La Plata, Argentina, were selected: an electromechanical repair and car painting center (ERCP), a sewing work room (SWR), and a chemical analysis laboratory (CAL). The results evidenced high levels of PM exceeding the limits allowed by the USEPA and the presence of benzo(k)fluoranthene in all the analyzed sites and benzo(a)pyrene in the most contaminated site (ERCP). Regarding metals, the presence of Cd, Ni, Cu, Pb, and Mn, mainly in the fraction of PM, in the same workplace was found. As far as risk assessment at all the workplaces surveyed is concerned, risk values for contracting cancer throughout life for exposed workers (LCR) did not comply with the parameters either of USEPA or of WHO (World Health Organization).
Three areas are highlighted in Gran La Plata, Argentina: industrial, urban, and residential. In this work, the levels of volatile organic compounds (VOCs) in indoor air of homes and schools in those areas were analyzed, through the use of passive monitors. The study period is between 2007 and 2010. Higher levels of VOCs were found in homes and schools in the industrial zone, higher than the levels corresponding to urban and residential. Taking into account the relationship between indoor and outdoor levels of VOCs, they have ratios (I/O) between 1.5 and 10 are evidenced contributions of emission sources of VOCs both indoor and outdoor. Complementarily, we estimated the life time cancer risk (LCR) for benzene, styrene, trichloroethylene, and tetrachloroethylene in children who spend their time mostly in such indoor environments. The results show high LCR values for benzene, which exceed acceptable values for the US EPA.

The quality of life in large megacities is directly affected by its air quality. In urban environments, suspended particles from anthropogenic origin is one of the main air contaminants identified as highly genotoxic, mutagenic, or carcinogenic. Atmospheric monitoring is therefore imperative, and bioassays to detect the effects of genotoxic agents give usually excellent results. Analysis of micronucleus (MN) in exfoliated oral mucosa cells is a sensitive non-invasive method for monitoring genetic damage in human populations. The first aim of this study was to analyze and characterize levels of volatile organic compounds (VOCs), particulate matter (PM), and polycyclic aromatic hydrocarbons (PAHs) in two areas from Buenos Aires: La Plata city, an urban (U) area and Ensenada, an industrial (I) area. Secondly, we evaluated the possible health risk of its inhabitants through a simple genotoxic assay on exfoliated oral mucosa cells. Whole blood cell count and nuclear abnormalities frequencies were evaluated in the exfoliated oral mucosa cells from urban and industrial inhabitants. Smoking habit represented a significant factor increasing MN percentage while, age did not increase the production of any of the nuclear aberrations assayed (micronuclei, binucleated, karyorrhexis) when the inhabitants from the urban and the industrial areas were compared. In addition, changes in MN and binucleated cell percentages in males and females were found to be area-dependent. We suggest that regardless PM concentration, PM-specific characteristics (size, shape, chemical elements, etc.) and VOCs levels could be responsible for the different harmful genotoxic effects seen in the two areas. Although this is a preliminary study, our results allowed to recognize that individuals living in both the urban and the industrial areas could be considered susceptible groups and should periodically undergo biological monitoring and appropriate care.
El objetivo general del presente trabajo de tesis doctoral fue determinar las alteraciones oculares que pudieran asociar a los niveles de contaminación aérea de la ciudad de La Plata y Ensenada, a través de la concentración del material particulado. Frente a este objetivo es que se intentaron investigar dos hipótesis generales. Una que postula que dos poblaciones humanas expuestas a diferentes calidades de aire, presentaran cambios significativos en distintos parámetros oculares, debido a que la superficie anterior del ojo se encuentra expuesta directamente a los contaminantes aéreos y que los mismos pueden generar una pérdida de la estabilidad de la película lagrimal, lo que podría traducirse en diferentes afecciones de las estructuras del segmento externo. Y otra, que considera que las distintas calidades de aire, se asocian con distintos alteraciones oculares detectables en poblaciones humanas. Con estas premisas, se diseñó un estudio comparativo entre dos zonas, una urbana y otra inmersa en un área industrial, en donde se encuentra uno de los polos petroquímicos más importantes del país. Estas zonas fueron definidas en base a las características socio-culturales, económicas, de desarrollo urbano, etc. y según sus valores de material particulado (MP), hidrocarburos aromáticos policíclicos (HAPs), y los compuestos orgánicos volátiles (COVs), a los que se les suma los efectos adversos en la función respiratoria y carcinogénicos en los habitantes, los cuales han sido valorados al menos hace 10 años por el grupo de trabajo. En este marco, se cuantificaron los niveles de material particulado durante el periodo en el que se determinaron diferentes parámetros del segmento anterior del ojo y de la película lagrimal, así como también la composición proteínica y lipídica de la misma. Durante el diseño también se consideró encontrar posibles asociaciones con parámetros sistémicos, previamente asociados al material particulado. Con este objetivo se realizaron encuestas sobre características de la vivienda de los voluntarios y de antecedentes de salud, así como también se realizaron estudios espirométricos y sanguíneos, los cuales fueron tenidos en cuenta como datos complementarios del estudio.
