Penicillium janthinellum, isolado como um fungo endofítico dos frutos de Melia azedarach, foi cultivado por 20 dias em milho branco triturado e autoclavado, onde os policetídeos conhecidos citrinina, emodina, 1,6,8-triidróxi-3-hidroximetilantraquinona, e uma nova antraquinona modificada, denominada janthinona, foram produzidos e isolados por procedimentos cromatográficos clássicos e identificados por extensivos estudos espectroscópicos, principalmente RMN 1D e 2D e EM. Essas substâncias foram ensaiadas contra diversas bactérias. Citrinina foi ensaiada pela primeira vez contra Leishmania e inibiu 100% o crescimento de cepas depois de 48h a uma concentração de 40 µg mL -1 . Penicillium janthinellum, isolated as an endophytic fungus from fruits of Melia azedarach, was cultivated over 20 days on ground and autoclaved white corn, where the known polyketides citrinin, emodin, 1,6,8-trihydroxy-3-hydroxymethylanthraquinone, and a new modified anthraquinone, named janthinone, were produced and isolated by classical chromatographic procedures and identified by MS and 1D and 2D NMR spectroscopic data. The antibacterial properties of these polyketides were investigated. Citrinin inhibited 100% of Leishmania growth after 48h at a concentration of 40 µg mL -1 .
SUMMARYThe objective was to estimate the prevalence of Blastocystis hominis, to evaluate the effectiveness of different techniques for its diagnosis as well as to estimate the prevalence of other intestinal parasites in the community of Campo Verde, a district of Pitanga. The work was carried out from August to October 2004. Samples of feces from children and adults were collected and submitted to the techniques of direct wet mount, flotation in zinc sulphate solution, tube sedimentation, sedimentation in formalinether and staining by Kinyoun and iron hematoxylin methods. From 181 studied individuals, 128 (70.7%) showed protozoa and/or helminths in stool samples. The most prevalent species were Endolimax nana (33.7%); B. hominis (26.5%); Giardia lamblia (18.2%); Entamoeba coli (17.1%); Ascaris lumbricoides (16.6%); Iodamoeba bütschlii (9.4%); and ancylostomatidae (7.7%). B. hominis was only identified by the techniques of direct wet mount, sedimentation in formalin-ether and staining by iron hematoxylin, though the latter was less sensitive than the other methods. The high frequency of B. hominis demonstrated by this study indicates the need to include laboratory techniques that enable identification of the parasite on a routine basis.
Resumo Visando a obtenção de informações sobre os casos de malária registrados no Estado do Paraná, analisaram-se os relatórios de atendimento de casos suspeitos de malária elaborados pela Fundação Nacional de Saúde, regional do Paraná, no período de janeiro de 1994 a dezembro de 1999. Das 31.975 amostras de sangue examinadas, 7,4% mostraram-se positivas, sendo 86,4% para Plasmodium vivax; 12,7% para P. falciparum; 0,04% para P. malariae e 0,9% para P. vivax e P. falciparum. Com relação à classificação epidemiológica, 84,5% representaram casos importados e 15,5% casos autóctones. Os municípios com maior número de casos autóctones foram Foz do Iguaçu, Santa Terezinha do Itaipu e Santa Helena, região de influência do lago de Itaipu, onde devem-se, portanto, concentrar os esforços de vigilância. Malária no Paraná.Abstract To collect data regarding registered cases of malaria in the state of Paraná, attendance reports of suspected cases of malaria performed by Fundação Nacional de Saúde, Paraná regional center, were analyzed from January, 1994 through December, 1999. Of 31,975 blood samples examined, 7.4% were positive: 86.4% for Plasmodium vivax, 12.7% for P. falciparum, 0.04% for P. malariae and 0.9% for P. vivax and P. falciparum. As to the epidemiological classification, 84.5% represented heterochthonous cases and 15.5% represented autochthonous cases. The municipalities showing higher rates of autochthonous cases were Foz do Iguaçu, Santa Terezinha do Itaipu and Santa Helena, a region influenced by the Itaipu reservoir, where prevention and control actions must be concentrated.
Leishmania (Leishmania) amazonensis LV79 (MPRO/BR/72/M1841) has been adapted to grow at 33 C as amastigote-like (AL) organisms in modified UM-54 medium initially adjusted to a pH of 4.8-5.0. Axenic cultures could be routinely restarted from parasites recovered from footpad lesions obtained by inoculation of BALB/c mice with preadapted culture stages. Morphological features, proteinase activities, and infectivity of AL organisms were examined during the in vitro growth cycle, and differences were found between log- and stationary-phase parasites. Stationary-phase AL organisms were morphologically similar to lesion amastigotes, did not react with a paraflagellar rod-specific monoclonal antibody in western blots, and contained proteinase activities resolving identically to the enzymes of lesion amastigotes in gelatin gels. Whereas typical megasomes could be identified in about a third of the stationary-phase AL population, the organelles were rarely seen in log-phase organisms. Azocaseinolytic activity progressively increased during the exponential growth phase and reached its highest values (approximately 65-70% of those determined in lesion amastigotes) at the stationary phase; the association of total proteinase activity with increased expression of cysteine proteinases was indicated by the strong inhibition of azocasein hydrolysis by E-64, the intensified banding of the 28-, 31-, and 35-kDa proteinases in gelatin gels, and the higher susceptibility of stationary-phase AL organisms to L-leucine methyl ester. Although overall axenic amastigotes were less infective to BALB/c mice than were lesion-derived parasites, stationary-phase AL organisms were more infective than were log-phase parasites. Medium pH increased during the exponential growth phase, but dropped in the stationary phase, when the observed morphological, biochemical, and biological changes became apparent.
Cistos de Giardia duodenalis e Entamoeba coli foram observados quanto à flutuação em soluções de sacarose de massas específicas diversas (1.040, 1.050, 1.060, 1.070,1.080,1.090,1.100, 1.150,1.200 e 1.250 kg/m³), contidas em câmaras de contagem de 0,17 mm de altura. Contaram-se os cistos que flutuaram e os que sedimentaram tendo sido calculadas as porcentagens respectivas. As diferenças de flutuabilidade dos cistos de cada uma das espécies não foram consideráveis. Soluções de massa específica igual a 1.200 kg/m³ fizeram flutuar 88,49% dos cistos de G. duodenalis e 95,71% dos de E. coli. Os maiores valores de flutuabilidade estiveram associados à massa específica 1.250 kg/m³ e foram, respectivamente, 89,15% e 98,59% para cistos de G. duodenalis e de E. coli.
Brine flotation and gravity sedimentation coproscopical examinations were performed in stool samples from 69 of the 147 Iaualapiti Indians of the Xingu Park, Mato Grosso State, Brazil. Intestinal parasites were present in 89.9% of the population examined. High rates of prevalence were found for some parasite species: Ancylostomidae, 82.6%; Enterobius vermicularis, 26.1%; Ascaris lumbricoides, 20.3%; and Entamoeba coli, 68.1%. Infection by Trichuris trichiura, Schistosoma mansoni, Taenia spp. and Hymenolepis nana was not detected. Helminth's prevalence in children aged one year or less was comparatively low (33.3%). Quantitative coproscopy was done in positive samples for Ascaris and Ancylostomidae and the results expressed in eggs per gram of feces (EPG). Quantitative results revealed that worm burdens are very low and overdispersed in this Indian tribe, a previously unreported fact.
O rendimento de sedimentação em tubos, aplicada à pesquisa de ovos de Ancylostomidae, Trichuris trichura e Ascaris lumbricoides foi estudado em suspensões fecais diluídas a 2,0%; 1,5% e 1,0% em colunas de 80 mm de altura. De cada um dos helmintos citados, as amostras continham quantidades correspondentes a 12,5; 25,0; 50,0; 100,0; 150,0 e 200,0 ovos por grama de fezes (opg). Avaliou-se o rendimento do processo em termos de freqüências de resultados positivos e de contagens de ovos por lâmina de sedimento (0,078 ml). Ambos aumentaram com os valores de opg. As várias diluições não influíram nos graus de positividade ou nas contagens de ovos por lâmina de sedimento. Os sedimentos de suspensões fecais diluídas a 1,0% (quando o valor de opg era de 150,0). Em trabalho rotineiro é desnecessária a determinação instrumental das quantidades de fezes usadas nas suspensões. Diluições em torno de 1,0% representam um compromisso entre bom rendimento e limpidez dos esfregaços, especialmente quando os valores de opg forem baixos.
