RESUMOO presente trabalho teve como objetivo analisar os efeitos de diferentes concentrações de sacarose no desenvolvimento in vitro de protocormios Cyrtopodium cachimboense L. C. Foram utilizados protocormios de Cyrtopodium cachimboense L. C. obtidos da germinação de sementes em meio de cultura alternativo, sem a presença de carvão, o delineamento experimental foi inteiramente casualizado. Após 90 dias a partir da germinação da semente, 5 protocormios foram transferidos para cada frasco com capacidade para 300 mL de meio de cultura. Foram realizadas 10 repetições de cada tratamento. Os tratamentos constituíram de: 0, 10, 20, 30, 40, e 50 gL -1 de sacarose, na ausência de carvão ativado. Os meios de cultura tiveram o pH ajustado para 5,5. Após três meses do desenvolvimento da plântulas, foram avaliados tamanho total de plantas, números de folhas, comprimento das folhas, números de raízes, comprimento médio das raízes, comprimento da maior raiz, e peso da massa fresca. Foi possível inferir que para o desenvolvimento das características fitotécnicas: número de raízes, número de folhas, peso da massa fresca e comprimento da maior raiz, a dosagem de 40g L -1 de sacarose, foi a mais efetiva. Para as variáveis comprimento da maior folha a dosagem de 10g L -1 e 30g L -1 apresentaram os melhores resultados e para comprimento total das plantas a dose recomendada é de 30 g L -1 de sacarose.
