The data obtained in the present study on spermiogenesis and spermatozoal ultrastructure of Pseudopimelodidae and Heptapteridae show that they share some characteristics, but greatly differ from each other. The main differences are the occurrence of type I spermiogenesis in Pseudopimelodidae and type III in Heptapteridae, the presence of nuclear fossa in Pseudopimelodidae and its absence in Heptapteridae, the presence of long midpiece in Pseudopimelodidae and short midpiece in Heptapteridae, the presence of cytoplasmic canal in Pseudopimelodidae and its absence in Heptapteridae, the presence of many large vesicles in the midpiece of Pseudopimelodidae and the presence of very long vesicles placed in the peripheral distal region in Heptapteridae, and mitochondria distributed all over the midpiece in Pseudopimelodidae, and very close to the nucleus in Heptapteridae. Heptapteridae and Pimelodidae share several characteristics, such as type III spermiogenesis, a similar chromatin condensation pattern, and the absence of nuclear fossa and flagellar lateral fins. The spermatozoa of Pseudopimelodidae is more similar to those of Siluridae. However, the absence of additional data on spermiogenesis and spermatozoa in siluriforms still limits a broader discussion in the order.
In Corydoradinae, the presence of spermatids in the lumen of the testicular tubules together with spermatozoa suggests that spermatogenesis is of the semicystic type, whereas in Callichthyinae, sperm production occurs entirely within spermatocysts in the germinal epithelium, characterizing cystic spermatogenesis. Spermiogenesis in Callichthyinae is characterized by an initial lateral development of the flagellum, the presence of nuclear rotation to different degrees, an eccentric or medial formation of a nuclear fossa, formation of a cytoplasmic channel, and presence of centriolar migration, being more similar to type I spermiogenesis. In Corydoradinae, spermiogenesis is characterized by eccentric development of the flagellum, the absence of nuclear rotation, an eccentric nuclear fossa formation, formation of a cytoplasmic channel, and absence of centriolar migration, differing from the types previously described. The process of spermatogenesis and spermiogenesis in Corydoradinae and Callichthyinae revealed unique characters for each of these subfamilies, corroborating the hypotheses that they constitute monophyletic groups. In relation to sperm ultrastructure, the comparative analysis of the callichthyid species shows that the general characteristics found in the spermatozoa were similar, thus, reinforcing the hypothesis that the family is monophyletic.
In the present study spermiogenesis was investigated in Cetopsis coecutiens (Cetopsidae), and Bunocephalus amazonicus (Aspredinidae), while spermatozoa ultrastructure was investigated in C. coecutiens, B. amazonicus, and Nematogenys inermis (Nematogenyidae). Aspredinidae and Cetopsidae share a spermatogenesis of the semicystic type, and a particular type of spermiogenesis process not reported in any fish group. In the three species analyzed, spermatozoa are biflagellate with flagella having the classical axoneme formulae (9 + 2). The analysis of thirteen characters showed the presence of eight characters shared by Cetopsidae and Aspredinidae, and six characters shared by Cetopsidae and Nematogenyidae, which may suggest that these three families may be more related than actually hypothesized, comprising a very primitive siluriform lineage originated after Diplomystidae.
SummaryRheumatoid arthritis (RA) may promote endothelial dysfunction. This phenomenon requires further investigation, especially in collagen-induced arthritis (CIA), as it is considered the experimental model most similar to RA. The objectives of this study were to identify CIA-induced changes in noradrenaline (NE) and acetylcholine (ACh) responses in mice aortas that may suggest endothelial dysfunction in these animals. Moreover, we characterize CIA-induced modifications in inducible nitric oxide synthase (iNOS) expression in the aortas and cardiac and renal tissues taken from these mice that may be related to possible endothelial dysfunction. Male DBA/1J mice were immunized with 100 lg of emulsified bovine collagen type II (CII) plus complete Freund's adjuvant. Twenty-one days later, these animals received a boost of an additional 100 lg plus incomplete Freund's adjuvant. Fifteen days after the onset of the disease, aortic rings from CIA and control mice were challenged with NE and ACh in an organ bath. In these animals, iNOS was detected through immunohistochemical analysis of aorta, heart and kidneys. Plasma nitrite concentration was determined using the Griess reaction. CIA did not change NE or ACh responses in mice aorta but apparently increased the iNOS expression not only in aorta, but also in cardiac and renal microcirculation. In parallel, CIA reduced nitrite plasma concentration. In mice, CIA appears to increase the presence of iNOS in aorta, as well as in heart and in kidney microcirculation. This iNOS increase occurs apparently in parallel to a reduction of the bioavailability of NO. This phenomenon does not appear to change NE or ACh responses in aorta.
Under these experimental conditions, the testicular function parameters showed that radiation produced marked damage that was not reversed by treatments. However, gonadal restructuring and recovery of spermatogenesis in treated animals may to reflect attenuation of radiation-induced damages and potential start of recovery.
Abstract Spadella, M. A., Oliveira, C. and Quagio-Grassiotto, I. 2006. Spermiogenesis and introsperm ultrastructure of Scoloplax distolothrix (Ostariophysi: Siluriformes: Scoloplacidae). -Acta Zoologica (Stockholm) 87 : 341-348 Spermiogenesis in scoloplacids is characterized by initial lateral development of the flagellum, nuclear rotation, medial nuclear fossa formation, complex centriolar migration, and cytoplasmic channel formation. The scoloplacid spermiogenesis is similar to those found in Diplomystidae, the most primitive siluriform family. The scoloplacid spermatozoa have all the main characteristics of introsperm. They exhibit a conic head, a symmetric midpiece, a medial flagellum, and no acrosome. The conic forward-elongated nuclei contain homogeneous chromatin. The thin extremity of the nuclei is strongly curved and along its internal face there is a well-developed membranous compartment. The centrioles are completely inside the medial nuclear fossa, perpendicular to each other and with an electron-dense material between them. In a cross view of the midpiece, the mitochondria form a ring surrounding internally the cytoplasmic channel, and in a longitudinal view they are organized in a row along it. Several elongated vesicles are distributed peripherally, mainly concentrated in the mid-piece basal region. The flagellum contains the classical axoneme (9 + 2) and has two lateral projections or fins. The spermatozoa of scoloplacids share several characteristics with those of Auchenipteridae. Since these two families are not phylogenetically related this similarity seems to be due to convergence once both families are, until now, the only known siluriform families with introsperm.
Purpose: To evaluate whether pre-treatment with rivastigmine is able to attenuate the I/R induced lesions in rat liver. Methods: SHAM animals or those submitted to I/R, non-treated or pre-treated with rivastigminine (2mg/kg) either 50 or 15 minutes before ischemia, were used. After I/R protocol, these animals were killed and their livers were harvested to measurement of the mitochondrial swelling as well as the malondialdehyde (MDA), nitrite and nitrate tissue concentration. Blood was also harvested for serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) determinations. Results: I/R promoted a significant increase of mitochondrial swelling in the studied animals. This increase of mitochondrial swelling was partially prevented by rivastigmine, but only if administered 50 minutes before ischemia. No significant modification of MDA, nitrite or nitrate tissue concentrations was observed in consequence of I/R, followed or not by rivastigmine treatments. In addition, I/R elevated both AST and ALT. These elevations of serum enzymes were not reversed by the different rivastigmine treatments. Conclusions: Rivastigmine administered 50 minutes before ischemia attenuates I/R-induced mitochondrial swelling, that indicates liver injury. This protective effect may be related to a greater stimulation of α7nAChR present in the Kupffer cells by the non-methabolized ACh, leading to an attenuation of I/R-induced inflammation.
