Infections with Trichuris trichiura and other trichurid nematodes have been reported to display protective effects against atopy, allergic and autoimmune diseases. The aims of the present study were to investigate the immunomodulatory properties of T. trichiura adult worm extract (TtE) and its fractions (TtEFs) on the production of cytokines by peripheral blood mononuclear cells and to identify their proteinaceous components. Fourteen TtEFs were obtained by ion exchange chromatography and tested for effects on cytokine production by peripheral blood mononuclear cells. The molecular constituents of the six most active fractions were evaluated using nano-LC/mass spectrometry. The homology between T. trichiura and the related nematode Trichinella spiralis was used to identify 12 proteins in TtEFs. Among those identified, fructose biphosphate aldolase, a homologue of macrophage migration inhibitory factor and heat-shock protein 70 may contribute to the immunomodulatory effects of TtEFs. The identification of such proteins could lead to the development of novel drugs for the therapy of allergic and other inflammatory diseases.
Smallanthus sonchifolius is a traditional Andean plant which has been cultured mainly in Brazil, Japan and New Zealand due to its medicinal properties. A study of the endophytic fungi associated to the plant was carried out in order to characterize new cytotoxic agents. Thirty two fungal strains were isolated and submitted to cultivation and extraction producing 186 extracts. Of these, 12% displayed moderate to high cytotoxic activities and were considered promising anticancer compound sources. The ethyl acetate fractions of Nigrospora sphaerica and Phoma betae liquid fermentations contained the synergistic compounds 8-hydroxy-6-methoxy-3-methylisocoumarin and (22E,24R)-ergosta-4,6,8(14),22-tetraen-3-one which are potential compounds for drug discovery. Another isolated compound, pimara-7,15-dien-3-beta-ol diterpene is being characterized for the first time through a detailed spectroscopic analysis including GC/MS, homo- and hetero-nuclear correlated NMR experiments (HMQC, HMBC, COSY and NOEdiff) along with its optical rotation.
CHEMICAL BIOLOGY: A MODERN STRATEGY FOR THE NATURAL PRODUCTS RESEARCH. INTRODUÇÃOA pesquisa em produtos naturais é uma das áreas mais tradicionais da Química no Brasil, devido a fatores históricos amplamente discutidos 1 e à grande biodiversidade do país. A formação científica dos pesquisadores e a infraestrutura dos laboratórios são bastante adequadas para o desenvolvimento de pesquisa de qualidade na área. A maioria dos trabalhos está voltada para o isolamento e a identificação de substâncias, muitas vezes associada a ensaios biológicos ou estudos quimiotaxonômicos. Estes trabalhos têm gerado um número significativo de publicações em revistas indexadas internacionais e nacionais, além de contribuir expressivamente para a formação de recursos humanos competentes na área de produtos naturais. Porém, algumas diferenças são observadas quando as pesquisas brasileiras em produtos naturais são comparadas com as desenvolvidas no exterior. Uma das principais diferenças está relacionada com a forte inserção dos produtos naturais na área biológica, fato que nas pesquisas brasileiras é observado quase que exclusivamente na busca de substâncias bioativas. Mesmo assim, a baixa produtividade em relação à identificação de produtos naturais efetivamente úteis como ferramentas de pesquisas biológicas ou como candidatos a protótipos de fármacos e outras substâncias de interesse sugere que a integração entre a Biologia e a Química não tem sido feita de forma eficiente na pesquisa em produtos naturais no Brasil.Neste sentido, o objetivo principal deste artigo é mostrar como alguns dos avanços na Biologia, especialmente na Biologia Molecular, podem ser associados à pesquisa em produtos naturais e vice-versa, ou seja, como os produtos naturais podem ser inseridos nas pesquisas biológicas de forma eficiente, contribuindo para a área de Biologia Química. Além disso, aspectos da política científi-ca no país e seu impacto na pesquisa também são discutidos. Siphoneugena Berg (Myrtaceae) é um pequeno gênero eugenióide segregado de Eugenia por Berg. A investigação fitoquímica de Siphoneugena densiflora foi realizada com o objetivo de se descobrir metabólitos secundários que possam ser considerados marcadores quimiotaxonômicos e auxiliar na distinção entre os dois gêneros. Cinco taninos hidrolisáveis inéditos foram isolados do extrato metanólico da casca da raiz e caracterizados como 4-O-α-L-2"-O-e seu isômero 4-O-α-L-3"-O-acetilramnopiranosídeo do ácido elágico, siphoneugenina, constituído por uma nova aglicona de estrutura dibenzo-1,4-dioxínica, 4-O-β-D-6"-e 4-O-β-D-3",6"-di-O-acetilglucopiranosídeo do ácido 3,4'-di-O-metilelágico, juntamente com o 4-O-α-L-4"-O-acetilramnopiranosídeo do ácido elágico, eschweilenol C, sitosterol, daucosterol, ramnose, casuarinina, castalagina e ácidos elágico, gálico e siríngico. Do extrato metanólico das folhas, além dos compostos conhecidos como a quercetina, quercitrina, guiajaverina, reinoutrina, chebulosídeo II, ácidos terminólico, madecássico e asiático, lupeol, α-e β-amirina, foi isolado um novo triterpeno pentacíclico nomeado como 2α,3β,6β-triidroxiolean-12-en-28-ato de β-D-glucopiranosila. The phytochemical survey of Siphoneugena densiflora was carried out in order to find secondary metabolites which may be considered as chemotaxonomic characters and help to distinguish between the two genera. Five novel hydrolysable tannins were isolated from the methanolic extract of root bark and were characterized as ellagic acid 4-O-α-L-2"-O-and its isomer 4-O-α-L-3"-O-acetylrhamnopyranoside, siphoneugenin, that supports a new aglycone with a dibenzo-1,4-dioxin structure, 3,4'-di-O-methylellagic acid 4-O-β-D-6"-O-and 4-O-β-D-3",6"-diacetylglucopyranoside, accompanied by ellagic acid 4-O-α-L-4"-O-acetylrhamnopyranoside, eschweilenol C, sitosterol, daucosterol, rhamnose, casuarinin, castalagin, ellagic, gallic and syringic acids. From methanolic extract of leaves, in addition to the well known compounds quercetin, quercitrin, guiajaverin, reynoutrin, chebuloside II, terminolic, madecassic and asiatic acids, lupeol, α-and β-amyrin, a new pentacyclic triterpene was isolated and named as β-D-glucopyranosyl-2α,3β,6β-trihydroxyolean-12-en-28-ate. Structures were established on the basis of spectroscopic and chemical evidence, along with the comparison of the data reported in the literature. The effects of crude extracts, fractions and isolated compounds from Vitex polygama Cham. and Siphoneugena densiflora Berg were evaluated on the development of Spodoptera frugiperda JE Smith, a destructive insect pest of corn and several other crops. The extracts and fractions were incorporated into an artificial diet at 1 mg g(-1) and offered to the insect during its larval stage. Length and viability of larval and pupal stages as well as pupal weight were assessed. Isolated compounds were tested through superficial contamination of the diet at 0.1 mg g(-1). Weight and viability of ten-day-old larvae were determined. Methanolic and hydroalcoholic S. densiflora extracts caused 100% larval mortality, while leaf and fruit hydroalcoholic extracts from V. polygama were the most active. Among the isolated compounds, flavonoids presented the best insecticidal results, and tannins the best larval growth inhibition.
