Contamination of fresh milk with Staphylococcus aureus was assessed comparatively through routine phenotypic (coagulase tube test and coagulase slide test) and genotypic (PCR) screening of 128 S. aureus strains isolated from 555 milk samples. These samples were collected from 362 cows with subclinical mastitis, hosted in different dairy herds at various locations of the Northern and Northeastern rural areas of the State of Rio de Janeiro, 39.7% of which were CMT-positive. All S. aureus isolates tested positive for the presence of the coagulase gene by PCR and the isolates could be grouped into four distinct classes according to the size of the PCR product. The strains also yielded variable results when assayed with coagulase test. Taken together, these data indicate the existence of extensive polymorphism at the coagulase gene locus in the genus Staphylococcus and exemplifies the extent of molecular and phenotypic heterogeneity associated with the strains circulating in rural herds.
O leite contaminado por resíduos de antimicrobianos é considerado adulterado e impróprio para o consumo, representando riscos à saúde do consumidor, riscos tecnológicos para a indústria de laticínios e rejeição da imagem da empresa pelo consumidor. Com os objetivos de determinar a freqüência da contaminação por resíduos de antimicrobianos no leite pasteurizado dos tipos "C" e "Integral Fazenda" comercializado na região Norte do Estado do Rio de Janeiro foram analisadas 300 amostras de leite pasteurizado tipo "C" (130 amostras) e "Integral Fazenda" (170 amostras) oriundas dessa região, de abril de 1996 a abril de 1997. Foi encontrada uma freqüência de 4.33% de contaminação por resíduos de inibidores microbianos no leite, utilizando-se, paralelamente, o método de inibição microbiana "Delvotest P" e o de proteínas ligadoras de penicilinas "bL Snap Test". A utilização isolada do "Delvotest P" revelou uma ocorrência de 3,66 % e do "bL Snap Test" uma freqüência de 1,66%. Para as amostras "Delvotest P" positiva e "bL Snap Test" negativa, foram utilizados testes para detectar tetraciclinas ("Tetraciclina Snap Test") e gentamicina ("Cite Probe Gentamicin"). Conclui-se que houve contaminação no leite de consumo, por vários tipos de antimicrobianos, na Região Norte do Estado do Rio de Janeiro.
The presence of Leptospira spp. and Brucella spp. antibodies was investigated in serum samples from 28 collared anteaters (Tamandua tetradactyla) kept in seven Brazilian zoos. Sera were tested against 19 Leptospira serovars using microscopic agglutination. Samples reacted to the following serovars: two (7.14%) to Patoc, three (10.71%) to Tarrasovi, three (10.71%) to both Patoc and Tarrasovi, two (7.14%) to Wolffi, and one (3.57%) to Australis. Two (7.14%) samples reacted to the buffered Brucella antigen test, but no confirmatory reaction occurred using the 2-mercaptoethanol slow slide agglutination test. No sample was reactive in the agar gel immunodiffusion test for rugose species of Brucella. The presence of anti-leptospira agglutinins in captive T. tetradactyla serum indicates that this species may be susceptible to infection by these bacteria.
Foram identificados Staphylococcus aureus e estafilococos coagulase-negativos produtores de enterotoxinas, a partir de amostras de leite de cabras. Coletaram-se 184 isolados estafilocócicos de 8 capris do estado do Rio de Janeiro. As amostras foram analisadas de acordo com técnicas microbiológicas de rotina e inoculadas em ágar estoque. A determinação das cepas enterotoxigênicas utilizou o método de Optimum Sensibility Plate (OSP). As cepas produtoras de enterotoxinas foram identificadas por método automatizado MiniApi pelo uso de galerias ID32Staph. Do total de amostras, 36 isolados (19,6%) foram produtoras de enterotoxinas (A, B, C, D). As espécies identificadas foram:
