S U M M A R Y Human leukocyte antigen-G (HLA-G) is a non-classical major histocompatibility complex class Ib molecule that acts as a specific immunosuppressor. Some studies have demonstrated that human papillomavirus (HPV) seems to be involved in lower or absent HLA-G expression, particularly in cervical cancer. In this study, we performed a cross-sectional study, systematically comparing the qualitative expression of the HLA-G5 isoform in invasive cervical carcinoma ( Two biopsies were collected from each patient (one from the tumor lesion and one from a lymph node). Immunohistochemistry analyses were performed for the HLA-G5 isoform, for HPV detection, and virus typing. HLA-G5 isoform molecules were detected in 25 cases (31.6%), 17 (32.7%) without metastasis and 8 (29.6%) with metastasis. HPV was detected in the cervical lesions of 74 patients (93.7%), but low expression of the HLA-G5 isoform was observed in all HPV-related cases. These findings are important; however, additional studies are necessary to identify the influence of HPV with HLA-G5 isoform expression on invasive cervical malignancies. (TGAS,CPS) S U M M A R Y The larynx is the most common site of malignancy in the upper aerodigestive tract. In Brazil, malignant laryngeal lesions represent 2% of all cancers, with ?3000 annual deaths. The association between human papillomavirus (HPV) and laryngeal cancer is still controversial. The aim of the present retrospective study was to determine the expression of galectin-3 immunoperoxidase in laryngeal carcinoma by examining paraffin-embedded larynx biopsies from 65 patients, 10 in situ laryngeal carcinomas, 27 laryngeal carcinomas without metastases, and 28 with metastases. Twenty-eight cervical lymph nodes from patients with metastatic lesions were also evaluated. Nested PCR was performed to detect and type HPV DNA. Galectin-3 expression was assessed by immunohistochemistry using a computerassisted system. Among 65 patients, 55 (84.6%) were positive to beta-globin (internal control); 10 (15.4%) patients were beta-globin negative and were excluded from the HPV evaluation. Thus, 7 (12.7%) out of 55 patients were HPV positive and 48 (87.3%) out of 55 patients were HPV negative. High expression of galectin-3 was observed in invasive laryngeal tumors, suggesting that galectin-3 could be associated with the invasiveness and aggressiveness of laryngeal carcinomas. O mecanismo de oncogênese na laringe pode ser controlado por vários fatores, entre eles fatores envolvidos na regulação do ciclo celular e outros de risco, tais como exposição prolongada ao fumo e álcool. O desenvolvimento do câncer de laringe também pode estar associado à infecção pelo HPV. Este estudo, análise imunoistoquímica quantitativa de p53, p27 e Mdm2, foi realizado em 54 pacientes com carcinoma invasivo de laringe subdivididos em: carcinoma sem metástase (laryngeal squamous cell carcinoma without metastasis -LSCC WT ), com metástase (laryngeal squamous cell carcinoma with metastasis -LSCC W ) e linfonodos cervicais (limph nodes biopsies -LB). A detecção e tipificação do HPV foram realizadas pela reação em cadeia da polimerase (PCR) e os tipos de HPV avaliados foram HPV 6, 11, 16, 18, 31 e 33. Na análise quantitativa, alta expressão de p53, p27 e Mdm2 foi observada nos grupos LSCC W e LSCC WT assim como nas biópsias dos linfonodos cervicais, indicando que a avaliação dessas proteínas poderia não discriminar carcinomas de laringe metastáticos e nãometastáticos. Detecção do HPV foi verificada em apenas 7.4% dos casos. Dentre os pacientes HPV positivos, verificou-se expressão negativa de p53. Por outro lado, alta expressão de p27 e Mdm2 foi observada. Em conclusão, a avaliação quantitativa de p53, p27 e Mdm2 não permite traçar um perfil complementar em lesões metastáticas de laringe. Palavras-chave: p53, p27, Mdm2, metástase, carcinoma de laringe.
