This study evaluated the antidepressant-like effect of insulin compared to sertraline and a combination of insulin and sertraline in streptozotocin (STZ)-induced type 2 diabetes mellitus (T2DM) rats submitted to the forced swim test (FST). Male Wistar rats were daily treated for 21 days with insulin (1 or 2 IU/kg, i.p.), with the selective serotonin reuptake inhibitor (SSRI), sertraline (10 mg/kg, i.p.), or with a combination of insulin (1 or 2 IU/kg, i.p.) and sertraline (10 mg/kg, i.p.) and submitted to the FST. We also evaluated the water and food intake, urine volume and weight gain of the rats. Rats treated with STZ showed impaired glucose tolerance. Chronic treatment with sertraline showed an antidepressant-like effect in non-diabetic and diabetic rats. Furthermore, sertraline promoted lower weight gain in diabetic rats. Insulin reduced the immobility behaviour in T2DM rats with impaired glucose tolerance. In conclusion, our results showed that insulin has an antidepressant-like effect comparable to that of sertraline. Sertraline is effective as an antidepressant and reduces weight gain, which reinforces its superiority over other SSRIs in the treatment of major depression disorder in patients with T2DM.Global estimates indicate that approximately 382 million people worldwide have diabetes, 316 million have impaired glucose tolerance and 90% of patients are diagnosed with type 2 diabetes mellitus (T2DM) [1]. Major depressive disorder (MDD) is the most common psychiatric comorbidity diagnose in patients with T2DM [1], with a global prevalence ranging from 11% to 30% [2].MDD amplifies the perception of diabetes symptoms [3], worsens glycaemic control and increases the complications of diabetes such as retinopathy, neuropathy, sexual dysfunction and macrovascular complications [4]. The treatment of MDD is necessary to improve the quality of life and to increase the adherence to glycaemic control therapies in patients with diabetes. Currently, selective serotonin reuptake inhibitors (5-HT, SSRIs) are the main therapeutic option in the treatment of MDD in patients with or without diabetes due to low incidence of adverse effects compared to tricyclic antidepressants, such as anticholinergic effects, on the cardiovascular system and glycaemic control [5,6].Although the mechanisms involved in the relationship between T2DM and MDD are not yet fully understood, studies show the involvement of insulin on different cognitive brain functions such as learning and memory [7]. A wealth of evidence indicates that the activation of 5-HT1A and 5-HT2A receptors in the dorsal periaqueductal grey matter (dPAG) inhibits escape, a panic-related defensive behaviour. Results that were previously obtained with the elevated T-maze test of anxiety/panic suggest that 5-HT1A and μ-opioid receptors in this midbrain area work together to regulate this response. To investigate the generality of this finding, we assessed whether the same cooperative mechanism is engaged when escape is evoked by a different aversive stimulus electrical stimulation of the dPAG. Administration of the μ-receptor blocker CTOP into the dPAG did not change the escape threshold, but microinjection of the μ-receptor agonist DAMGO (0.3 and 0.5 nmol) or the 5-HT1A receptor agonist 8-OHDPAT (1.6 nmol) increased this index, indicating a panicolytic-like effect. Pretreatment with CTOP antagonised the anti-escape effect of 8-OHDPAT. Additionally, combined administration of subeffective doses of DAMGO and 8-OHDPAT increased the escape threshold, indicating drug synergism. Therefore, regardless of the aversive nature of the stimulus, μ-opioid and 5-HT1A receptors cooperatively act to regulate escape behaviour. A better comprehension of this mechanism might allow for new therapeutic strategies for panic disorder.

Introdução: A qualidade de vida de pacientes oncológicos tem sido objeto de estudo em muitos trabalhos brasileiros. Contudo, apesar da alta prevalência de indivíduos submetidos a radioterapia, poucos estudos com ênfase nesse grupo de pacientes foram identificados. Objetivo: Avaliar a qualidade de vida e a prevalência de sintomas depressivos em pacientes com neoplasias malignas durante o tratamento radioterápico. Método: Estudo transversal quantitativo realizado com 153 pacientes oncológicos em vigência de tratamento radioterápico em um centro especializado em oncologia e radioterapia, localizado no Noroeste do Estado do Parana. Os dados foram coletados entre marco e setembro de 2018. O European Organization for Research and Treatment of Cancer Quality of Life Questionnaire (EORTC QLQ-C30) e o Inventario de Depressão de Beck foram utilizados para avaliar a qualidade de vida e os sintomas depressivos, respectivamente. Resultados: Os domínios “qualidade de vida”, “função cognitiva” e “função social” foram os que menos se mostraram prejudicados na amostra estudada, enquanto “insônia”, “perda de apetite” e “dificuldades financeiras” destacaram-se entre os maiores preditores de baixa qualidade de vida. Ademais, contatou-se que 22% dos indivíduos avaliados apresentaram algum grau de transtorno de humor, sendo 11% diagnosticados com depressão. Conclusão: O declínio na qualidade de vida e a prevalência de sintomas depressivos em pacientes oncológicos, mesmo os em vigência de radioterapia, enaltecem a importância de intervenções precoces que visem a restabelecer a funcionalidade e o bem-estar.

Introdução: A dor pode ser incapacitante dependendo da sua intensidade e duração, fazendo com que o indivíduo tenha uma perda na qualidade de vida. Atualmente os tratamentos preconizados apresentam efeitos benéficos que se escassa a curto prazo e seus efeitos adversos faz com que seja imprescindível novas terapias farmacológicas, neste sentido observa-se atualmente o uso do Canabidiol. Objetivo: Analisar o efeito do canabidiol em indivíduos com dor crônica e aguda. Método: Trata-se de uma revisão integrativa da literatura bibliográfica sobre os efeitos do Canabidiol como tratamento de dor crônica e aguda. A busca será realizada por meio das palavras-chave baseadas nos Descritores em Ciências da Saúde (DeCS) nos idiomas português: Canabidiol, Dor, Dor Crônica, Dor aguda, Neuropatias.  Ao final do projeto, utilizará o Protocolo PRISMA para que os artigos sejam selecionados. Resultados: O trabalho evidenciou resultados, predominantemente, positivos em relação ao uso do canabidiol (CBD) para manejo da dor. De maneira geral, nos estudos analisados, o composto derivado da Cannabis mostrou boa eficácia nos efeitos antinociceptivos dos indivíduos observados. Conclusão: a utilização do canabidiol para manejo da dor é uma terapêutica promissora, porém ainda necessita de maior compreensão sobre seu uso prolongado e mecanismos de ação.
