Solanum nigrum L. plants were exposed for 28 days to 100 and 200 μmol/L copper (Cu) in a hydroponic system to analyze the antioxidant defense response. A dose‐dependent reduction in growth (fresh mass of root and shoot, shoot height, and root elongation) with increasing concentration of Cu was observed, whereas Cu treatments did funt affect total chlorophyll and carotefunids content. An enhanced lipid peroxidation, in terms of malondialdehyde (MDA) content, was quantified in shoots when the plants were subjected to the highest Cu level, while in roots MDA levels showed a dose‐dependent increase along the increasing Cu concentrations applied. An increase of proline in roots of plants exposed to 200 μmol/L Cu was found. Antioxidant enzymes superoxide dismutase (SOD) and catalase (CAT) did funt show significant changes with respect to control, in both roots and shoots, despite mRNA‐specific accumulations varied between Cu levels and organs. Ascorbate peroxidase (APX) was negatively affected in shoots by the highest Cu level. Gene expression of the subtype 2d metallothioneins (MT) revealed to be Cu‐enhanced throughout the plant body and correlated with Cu tissue levels, with the other MT1 and MT2 gene members downregulated in roots and upregulated in shoots, contributing more as antioxidants in the latter organs than in Cu homeostasis. MT3s are not involved in Cu homeostasis and phytochelatin (PC) production was enhanced in roots of plants exposed to 200 μmol/L Cu, contributing to a higher Cu accumulation in these organs.

Solid-phase microextraction commercial fibers present a few drawbacks such as relatively low recommended operating temperature, instability and swelling in organic solvents, breakage of the fiber, stripping of coatings, and bending of the needle. Some of these problems have been obviated by covalent bonding of the polymer phase to the fused-silica substrate by sol-gel, but the easy breakage of the fiber remains a problem. In the present work, the known occurrence of titanol groups at the surface of titanium wire was exploited to produce sol-gel fibers supported on this unbreakable substrate. Scanning electron microscopy analysis revealed the film formation on titanium wire surface while temperature and solvent stability as well as durability tests showed that the sol-gel film was tightly attached to the substrate, thus suggesting covalent bonding. The use of this type of fiber is currently generalized in our laboratory without any breakage or stripping out incidents up to the moment.

Four monovarietal red wines, Tinta Roriz, Tinta Barroca, Touriga Nacional, and Touriga Francesa, were studied for their complexometric properties toward copper. For the purpose, ion selective electrode potentiometric titrations were carried out, and the data were treated by two methods suitable for heterogeneous ligands: the Scatchard plot, which provided the complexation capacity (CC(total) = maximum number of moles of sites per liter), and the differential equilibrium function (DEF), which provided the conditional stability constants of the sites effectively involved in the metal complexation (K(DEF)) at a given titration point. The different wines displayed similar properties: CC(total) ranged from 4.5x10(-3) (Tinta Barroca) to 4.9x10(-3) M (Touriga Francesa), and log K(DEF) values were between 4.2 and 6.5. The range of log(|Cu|(bound)/CC(total)) = log theta embraced in the titration was between -0.2 and -1.9. With regard to the natural levels of copper, from 4.4x10(-7) (Tinta Roriz) to 1.3x10(-6) M (Touriga Nacional), and ligands (CC(total)) in the wines, it can be expected that copper will be strongly complexed in the studied wines (log K(DEF) > 6). Studies carried out, in parallel, for solutions of isolated seed tannins and skin polyphenols (mainly anthocyans) showed that their contribution to the CC(total) of the wines was <40% for tannins and <2% for skin polyphenols. However, skin polyphenols were shown to be much stronger copper ligands (log K(DEF) = 6.7, log theta = -1.5) than tannins (log K(DEF) = 4.5, log theta = -1.5).

Overuse of pesticides has resulted in environmental problems, threating public health through accumulation in food chains. Phytoremediation is a powerful technique to clean up contaminated environments. However, it is necessary to unravel the metabolic mechanisms underlying phytoremediation in order to increase the efficiency of this process. Therefore, growth, physiological and biochemical responses in leaves and roots of Solanum nigrum L. exposed to the commonly used fungicide metalaxyl were investigated. This species shows characteristics that make it valuable as a potential tool for the remediation of organic pollutants. We found that once inside the plant, metalaxyl altered carbon metabolism, which resulted in a reduction of growth and lower biomass accumulation due to impairment of carbohydrate production (total soluble sugar, starch, rubisco) and increased photorespiration (glycolate oxidase, Gly/Ser ratio). A significant increase of antioxidant defenses (polyphenols, flavonoids, tocopherols, ascorbate, glutathione, superoxide dismutase, catalase, peroxidases, monodehydroascorbate- and dehydroascorbate reductase, gluthatione reductase) kept reactive oxygen species (ROS) levels under control (superoxide anion) leaving cell membranes undamaged. The results suggest that enhancing carbon assimilation and antioxidant capacity may be target parameters to improve this species’ phytoremediation capacities.Highlights• Metalaxyl inhibits growth by reducing photosynthesis and inducing photorespiration• Elevated antioxidant defenses protect metalaxyl-treated plants from oxidative damage• Ascorbate and glutathione are key antioxidants in metalaxyl tolerance.
