RESUMO Objetivou-se comparar a digestibilidade in vitro da matéria seca (DIVMS) e da fibra em detergente neutro (DIVFDN) de forragens e concentrados obtidos por intermédio das incubadoras DaisyII e TE-150, combinadas com filter bags tipo F57 (Ankon®) ou tecido não-tecido (TNT, 100g/m²), em relação aos valores obtidos utilizando-se o método de Tilley e Terry (1963). Foram utilizadas 25 amostras de alimentos concentrados e 25 de forragens. Não houve diferenças entre filter bags (P>0,39) para a DIVMS de forragens. Maiores valores de DIVMS (P<0,01) foram verificados para a incubadora TE-150 em relação à DaisyII. As estimativas de DIVMS obtidas com incubadoras e filter bags foram superiores (P<0,01) àquelas obtidas com o método Tilley e Terry (1963). Observaram-se maiores valores de DIVFDN de forragens (P<0,01) com filter bags F57 em relação ao TNT e com a incubadora TE-150 em relação à DaisyII. Todos os valores de DIVFDN obtidos com incubadoras e filter bags foram superiores (P<0,01) aos obtidos com o método Tilley e Terry (1963). Todos os métodos apresentaram-se positiva e fortemente correlacionados (P<0,01), tanto para DIVMS como para DIVFDN. As repetibilidades e variâncias entre amostras para DIVMS e DIVFDN foram similares entre os métodos. Portanto, conclui-se que, em se tratando de avaliações comparativas entre alimentos, todos os métodos avaliados possuem capacidade similar de discriminação.

Rigorously standardized laboratory protocols are essential for meaningful comparison of data from multiple sites. Considering that interactions of minerals with organic matrices may vary depending on the material nature, there could be peculiar demands for each material with respect to digestion procedure. Acid digestion procedures were evaluated using different nitric to perchloric acid ratios and one- or two-step digestion to estimate the concentration of calcium, phosphorus, magnesium, and zinc in samples of carcass, bone, excreta, concentrate, forage, and feces. Six procedures were evaluated: ratio of nitric to perchloric acid at 2:1, 3:1, and 4:1 v/v in a one- or two-step digestion. There were no direct or interaction effects (p>0.01) of nitric to perchloric acid ratio or number of digestion steps on magnesium and zinc contents. Calcium and phosphorus contents presented a significant (p<0.01) interaction between sample type and nitric to perchloric acid ratio. Digestion solution of 2:1 v/v provided greater (p<0.01) recovery of calcium and phosphorus from bone samples than 3:1 and 4:1 v/v ratio. Different acid ratios did not affect (p>0.01) calcium or phosphorus contents in carcass, excreta, concentrate, forage, and feces. Number of digestion steps did not affect mineral content (p>0.01). Estimated concentration of calcium, phosphorus, magnesium, and zinc in carcass, excreta, concentrated, forage, and feces samples can be performed using digestion solution of nitric to perchloric acid 4:1 v/v in a one-step digestion. However, samples of bones demand a stronger digestion solution to analyze the mineral contents, which is represented by an increased proportion of perchloric acid, being recommended a digestion solution of nitric to perchloric acid 2:1 v/v in a one-step digestion.

ABSTRACT. The effects of the salt-to-metal catalyst ratio and amount of digestion mixture on total nitrogen content in different materials using the Kjeldahl method were evaluated. Four samples with low nitrogen contents and four samples with high nitrogen contents were analyzed. The study evaluated two ratios of salt (sodium sulfate) to metal catalyst (copper sulfate) in the digestion mixture (10:1 and 20:1) and three amounts of digestion mixture per sample (1.0, 1.5, and 2.0 g 200 mg -1). There was an interaction between low-nitrogen material and amount of digestion mixture on nitrogen contents. Samples of cattle feces and corn presented higher nitrogen content when 1.5 and 2.0 g of digestion mixture were used. The high-nitrogen materials presented higher nitrogen contents when 2.0 g of the digestion mixture was used. However, there was an interaction between high-nitrogen material and the ratio of the digestion mixture components. The cattle carcass sample showed higher nitrogen content when the 20:1 ratio was used. The digestion mixture amount in the Kjeldahl method must be 2.0 g with a salt-to-metal catalyst ratio of 20:1 when samples of approximately 200 mg are analyzed.

-The objective of this work was to evaluate the accuracy of digestion techniques using nitric and perchloric acid at the ratios of 2:1, 3:1, and 4:1 v v -1 , in one-or two-step digestion, to estimate chromium contents in cattle feces, using sodium molybdate as a catalyst. Fecal standards containing known chromium contents (0, 2, 4, 6, 8, and 10 g kg -1 ) were produced from feces of five animals. The chromium content in cattle feces is accurately estimated using digestion techniques based on nitric and perchloric acids, at a 3:1 v v -1 ratio, in one-step digestion, with sodium molybdate as a catalyst.

Background Crude ash is categorized as an empirical method playing an important role in the nutritional interpretation of animal feeds, allowing indirect estimation of total organic matter. Objective Our objective was to evaluate variations in laboratory procedures for crude ash quantification regarding physical parameters (i.e., time, temperature) and ashing aids and their influences on crude ash, repeatability, and discrimination power among feeds. Method The “control” method was based on a simple ignition time of 3 h at 550 °C. The variations are briefly described: increasing ashing time to 6 h; increasing temperature to 600 °C; and using two 3-h ignition cycles at 550 °C with ashing aids inclusion between them: fresh air supply, fresh air supply plus distillate water, and fresh air supply plus hydrogen peroxide. A color evaluation was also performed using a colorimetric technique. Twenty four study materials from eight different feed types were evaluated. Results The crude ash results differed among the method variations, but a consistent decrease in the estimates was observed when liquid aids were applied, which also improved repeatability. Ash residues did not present a consistent color pattern among methods, but the residues were darker when the control method was applied. Conclusions The method of obtaining ash residues in animal feeds based on the binomial 550 °C × 3 h does not have enough robustness and may overestimate crude ash in some feeds. Adjustments in either ignition time or temperature might improve crude ash test results, but the best test results are obtained using liquid ashing aids between two ignition cycles. Highlights The recommended method is based on the use of 550 °C and two 3-h ignition cycles with water added to the ash residue between cycles.
