Durante essa alternância, Photorhabdus inverte um segmento de DNA que corresponde ao promotor do operon mad, que codifica proteínas integrantes de uma fímbria necessária para a adesão ao intestino da fêmea do nematoide (SOMVANSHI et al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
This inseticide property is due to a rather peculiar kind of symbiosis established between the worm and bacteria, the latter being the main responsible for the death of the insect. Besides offering interesting data to understand the biological mechanisms involved in symbiosis, the entomopathogenic nematodes represent a potential tool to control insect pests in agriculture and even arachnids like ticks. Additionally, the study of entomopathogenic bacteria is leading to the identification of different bioactive compounds of microbial origin.
