Lack of a suitable assay has thwarted attempts to measure cytochrome c-552 in dark-grown wild type cells of Euglena gracilis var. bacillanis in mutants and in other situations where the concentrations are low. Purification methods are described based on electrofocusing which provide a cytochrome c-552 preparation homogeneous enough to elicit a single reactive antibody in rabbits; this antibody is then used as a specific and sensitive assay for cytochrome c-552. Microspores of Lilium longiflorum were isolated at various stages of development surrounding the mitotic interval and were analyzed for changes in the activities of n-glyceraldebyde-3-phosphate dehydrogenase and aldolase. Fructose 1,6 diphosphate was used as substrate. Activities were measured by the increase in optical density due to the reduction of diphosphopyridine nucleotide.It was found that mitosis occurs during the minimal activity of both aldolase and n-glyceraldehyde-3-phosphate, thus indicating that heightened glycolytic capacity is not necessarily related to mitosis. It was also found that soluble-SH levels were highest when the enzymes were least active. It appeared, therefore, that the "--SH enzymes" are not necessarily activated intracellularly by high concentrations of soluble thiol. These results are discussed in connection with the theory that soluble-SH compounds stimulate glycolysis and in this way initiate mitosis.
The extract of soluble proteins from Lilium anthers is characterized by four electrophoretic components. Three of the components show quantitative differences markedly correlated with the synchronized division of the microspores contained within the anthers. One of the components increases in amount before mitosis and disappears at mitosis; two of the components have peak values at the time of mitosis and decrease thereafter; a fourth component remains relatively constant in amount throughout.
Sequential measurements were made of the free amino-acid pool in the premitotic region of microspores of Lilium longiflorum. The basic amino acid arginine appears in the pool and then disappears.
