Bacterial populations in goldfish feces were characterized by the fluorescent in situ hybridization (FISH) method. A total of nine different group‐specific rRNA‐targeted oligonucleotide probes were used. Approximately half of the microbial cells (57.8 ± 16.7%) were detected with a probe EUB338 and found to be bacteria. The microbial cells in 33–35 of the 35 samples from five specimens strongly hybridized with probes ALF1b, BET42a and GAM42a, suggesting that goldfish intestinal bacteria are mainly composed of α, β and γ‐subclasses of Proteobacteria. The fact that a probe AER66 reacted with 25.6 ± 14.2% of the total microbial cells in all 35 samples, demonstrated that genus Aeromonas was the dominant species in the goldfish intestines. Genus Bacteroides including Bacteroides type A detected with a probe BAC303 was observed in 15 of 35 samples while other taxonomic groups determined with HGC69a, CF39a and P72 were detected in 6–11 of 35 samples. These results strongly suggest that Bacteroides shows the greatest daily fluctuation and interindividual variation in the intestines of goldfish. Moreover, the FISH method was proven to be useful for rapid enumeration of taxonomic groups of fish intestinal bacteria.
Uji coba budidaya dalam keramba telah dilakukan untuk mengetahui kelayakan kolam pengendap tambang batubara untuk areal budidaya ikan bagi masyarakat sekitar apabila kegiatan tambang telah ditutup, serta untuk mengetahui apakah produk ikan yang dihasilkan cukup aman untuk dikonsumsi manusia. Percobaan dilakukan pada bekas kolam pengendap DS2 milik PT KPC, dengan mengunakan 5 buah keramba apung berukuran 3 m x 1 m x 1 m yang dalam setiap keramba ditebar benih dari alam untuk ikan repang (Barbodes schwanenfeldii) ukuran rata-rata 20,2 g; puyau (Osteichilus kappenii) ukuran 66,1 g; dan mas (Cyprinus carpio) ukuran 28,96 g dari pemijahan di laboratorium sebanyak masing-masing 200 ekor, serta pepuyu (Anabas testudineus) ukuran rata-rata 41,4 g dari alam sebanyak 50 ekor, udang galah (Macrobrachium rosenbergii) ukuran 113,8 g juga dari alam sebanyak 50 ekor. Pakan diberikan secara sampai kenyang (ad libitum). DO, pH, suhu, DHL, dan kekeruhan diukur harian, sedangkan ikan diukur pertumbuhan bobotnya. Setelah dipelihara selama 4 bulan dan memenuhi ukuran konsumsi, ikan dan udang diperiksa kandungan Sb, Se, As, Hg, Mn, Cd, Fe, Cu, Pb, dan Zn. Hasil penelitian menunjukkan bahwa bekas kolam pengendap tambang batubara cukup layak untuk dikembangkan sebagai areal budidaya ikan dengan memberikan pertumbuhan bobot populasi sebesar 570,79% (repang, SR: 95%), 202,57% (puyau, SR: 97%), 573% (mas, SR: 2,5%), 238,92% (udang galah, SR: 10%), 447,10% (pepuyu, SR: 14%) dan produknya cukup aman untuk dikonsumsi, dengan kandungan Sb (0,24-2,45 mg/L, rasio terdeteksi dari sample: 100%), Se (0,00-0,06, 57,14%), As (tidak terdeteksi/ttd), Hg (0,00-0,06 mg/L, 50%), Mn (ttd-1,68 mg/L, 14,29%), Cd (ttd), Fe (ttd-5,45 mg/L, 7,14%), Cu (ttd), Pb (ttd), dan Zn (7,82-61,50 mg/L, 100%).The experiment was conducted to study the feasibility of settling or sedimentation pond of coal mining to be used for culturing fish in net cage for local people post mining activities. Experimental 3 m x 1 m x 1 m of 5 floating net cages were located in DS 2, an abandoned settling pond owned by Kalimantan Timur Prima Coal (KPC). Each cage was stocked with natural barb seed (Barbodes schwanenfeldii) with mean weight of 20.2 g, fresh water carp or puyau (Osteichilus kappenii) with mean weight of 66.1 g, and common carp (Cyprinus carpio) with mean weight of 28.96 g from fish hatchery with density of 200 seeds per species per cage, wild seed of climbing pearch (Anabas testudineus) with mean weight of 41.4 g and density of 50 seeds per cage, and giant prawn (Macrobrachium rosenbergii) with mean weight 113.8 g and density of 50 seeds per cage. The fishes were feed ad libitum. Water quality parameters were measured daily consisting of DO, pH, temperature, conductivity, and turbidity, while fish growth was determined by measuring its weight. After 4 months of rearing and reached marketable size, the fish were then measured for the evidence of Sb, Se, As, Hg, Mn, Cd, Fe, Cu, Pb, and Zn. The result shows that the settlement pond of coal mining is feasible to rear fish in cage. The recorded fish growths were 570.79% for barb fish with survival rate/SR: 95%, 202.57% for local carp with SR: 97%, 573% for common carp with SR:2.5%), 238.92% for giant prawn with SR:10%), 447.10% for climbing perch with SR:14%. All of the reared fish are safe to be consumed because they have normal content of Sb (0.24 to 2.45 mg/L, and detected ratio in all sample was: 100%), Se (0.00 to 0.06; 57.14%), As (not detected/nd), Hg (0.00 to 0.06 mg/L, 50%), Mn (nd to 1,68 mg/L, 14.29%), Cd (nd), Fe (nd-5.45 mg/L, 7.14%), Cu (nd), Pb (nd), and Zn (7.82-61.50 mg/L, 100%).
Penelitian pengembangan pembenihan ikan betok (Anabas testudineus) untuk skala rumah tangga telah dilakukan untuk mengatasi sulitnya ketersediaan benih ikan betok untuk keperluan budidaya di Kalimantan Timur. Sebanyak tiga orang warga Kota Samarinda telah dipilih untuk mengembangkan pembenihan ikan betok selama delapan bulan. Sebanyak 20 ekor indukan jantan dan 30 ekor indukan betina ukuran 30-110 g/ekor, dipelihara dalam kotak kayu ulin kedap air ukuran 2 m x 1 m x 0,75 m dengan menggunakan media air hujan dan diberi makan ad libitum pagi dan sore dengan menggunakan pelet mengandung protein minimal 30%. Indukan yang telah matang gonad disuntik dengan hormon Ovaprim sesuai dosis anjuran, dan selanjutnya 2 ekor jantan dan 1 ekor betina dipijahkan dalam akuarium kaca ukuran 60 cm x 40 cm x 40 cm dengan ketinggian media air hujan 30 cm. Larva ikan betok diberi pakan 2x sehari pagi dan sore berupa larutan infusoria, dan kutu air (Moina sp.) hidup sebagai persiapan pakan lanjutan. Selain pengamatan terhadap pemahaman pembenihan ikan betok oleh pelaksana, dilakukan juga pengamatan terhadap tingkat keberhasilan pemijahan, bobot telur (butir), derajat penetasan (%), sintasan (ekor, %), jenis plankton di media, panjang dan bobot anakan pada umur empat bulan, serta kualitas air. Data yang dikumpulkan dianalisis secara deskriptif. Hasil penelitian menunjukkan pengembangan pembenihan ikan betok dapat dilakukan di tingkat rumah tangga pada lahan sempit dengan metode induced breeding, dengan 100% terjadi pemijahan, jumlah telur: 8.978-39.868 butir, derajat penetasan: 69,40%-98,14%, derajat sintasan anakan hingga usia empat bulan: 0,17% dan 0,54%, panjang total anakan usia empat bulan adalah (nilai rata-rata ± simpangan baku): 49,51 ± 15,71 mm, dan bobot: 2,46 ± 2,50 g (n= 24), hubungan antara panjang total (X) terhadap bobot (Ŷ) pada umur yang sama adalah: Ŷ=4,59 + 0,15X dan keeratan korelasi (r) sebesar: 0,96. Ikan betok dapat dibudidayakan pada media air hujan dengan kejenuhan oksigen: 1,50%-47,4%, kelarutan oksigen: 0,12-3,80 mg/L, dan pH: 3,45-5,85. Rendahnya sintasan benih ikan betok utamanya disebabkan oleh tingginya pemangsaan antar sesama (kanibalisme) sejak benih berusia lebih dari satu minggu, dan sulit untuk dikendalikan.
Tiga spesies tanaman sayur, kangkung darat (Ipomoea reptana), sawi (Brassica juncea), dan kemangi (Ocimum basilicum) dibandingkan guna mengonversi ammonium dan nitrat nitrogen dari sistem akuakultur. Tanaman tersebut ditanam secara hidroponik menggunakan teknik rakit (rafting technique) dengan tata letak rancangan acak kelompok lengkap (RAKL). Hasil percobaan menunjukkan bahwa tingkat konversi nitrogen oleh ketiga jenis tanaman berbeda secara nyata dengan tingkat retensi nitrogen tertinggi pada tanaman kangkung sebesar 0,73±0,28 g; diikuti oleh kemangi (0,30±0,17 g); dan terakhir oleh sawi (0,03±0,07 g). Secara keseluruhan ketiga tanaman mampu menyisihkan limbah nitrogen sebesar 6,70% dari total produksi TAN dari sisa metabolisme ikan yang dibudidayakan.
