Lallemantia iberica (Bieb.) Fisch. and C. A. Mey is high valued annual ornamental and medicinal plant from Lamiaceae family that prefers dry sunny hillsides, roadsides, slopes, and fallow fields over an altitude of 500–2150 m. It bears beautiful white flowers and bloom from April to June each year. This study reports L. iberica micropropagation using cotyledon node explants isolated from 15-day-old in vitro regenerated plantlets. The cotyledon node explants were cultured on MS medium containing 0.50, 1.00 plus 2.00 mg/L BAP, 0.00, 0.01, and 0.02 mg/L NAA. Maximum shoot regeneration was noted on MS medium containing 0.50 mg/L BAP. Well-developed micropropagated shoots were rooted on MS medium containing 1.00 mg/L IBA. The rooted plants were easily hardened in the growth chamber and acclimatised in greenhouse.
Mentha spicata subsp. spicata tıbbi ve aromatik bir bitkidir. Bu çalışmada Mentha spicata subsp. spicata'nın in vitro' da çoğaltımı amacıyla hipokotil eksplantları BAP ve NAA 'nın farklı kombinasyonlarını içeren, %3 sukroz ilave edilmiş ve % 0.65 agar ile katılaştırılmış MS besin ortamında kültüre alınmıştır. Araştırma sonuçları kallus oluşum yüzdesi, rejenere sürgün yüzdesi, eksplant başına düşen ortalama sürgün sayısı, ortalama sürgün uzunluğu, eksplant başına düşen ortalama kök sayısı ve ortalama kök uzunluğu için BAP-NAA arasındaki kombinasyoların etkili olduğunu göstermiştir. Maksimum kallus oluşum yüzdesi 0.25 mg/L BAP + 0.50 mg/L NAA içeren MS besin ortamında, maksimum rejenere sürgün yüzdesi 2 mg/L BAP + 0.25 mg/L NAA içeren MS besin ortamında, maksimum eksplant başına düşen ortalama sürgün sayısı 2 mg/L BAP içeren MS besin ortamında, maksimum ortalama sürgün uzunluğu 2 mg/L BAP + 0.50 mg/L NAA içeren MS besin ortamında, kaydedilmiştir. Background Seed weevils are one of the causes of post harvest rice yield losses in Turkey. There are many cultural and chemical methods to control these insects.PurposeThis study aimed to control these using Agrobacterium tumefaciens mediated genetic transformation in two rice cultivars namely Hamzadere and Osmancı-97.Methods and results Two days old germinated seeds of the two cultivars with 2-3 mm long shoots were used as explants. They were treated with Agrobacterium tumefciens containing a pTF101AoPR1AcBar plant expression vector containing the cry1Ac gene under the control of the wound-induced (AoPR1) promoter. The transformation event was genotype-dependent. Out of 3000 and 2600 explants of cv Hamzadere and cv Osmancık-97, only 92 (3.07%) and 32 (1.230%) plants were selected on MS medium containing 3.0 and 2.5 mg/l bialaphos. These were validated using PCR, bioassay, and ELISA tests. All plants were held in the greenhouse until T0 and T1 seed settings. three (0.10%) plants of cv Hamzadere and 5 (0.19%) plants of cv. Osmancık 97 could be validated through all tests in T₀ and T₁ generations in both cultivars of rice for the first time using a novel and a very simple transformation procedure.ConclusionThese results are very encouraging and will influence future rice transformation techniques and improve rice breeding technologies.
