Salamander axolotl has been emerging as an important model for stem cell research due to its powerful regenerative capacity. Several advantages, such as the high capability of advanced tissue, organ, and appendages regeneration, promote axolotl as an ideal model system to extend our current understanding on the mechanisms of regeneration. Acknowledging the common molecular pathways between amphibians and mammals, there is a great potential to translate the messages from axolotl research to mammalian studies. However, the utilization of axolotl is hindered due to the lack of reference databases of genomic, transcriptomic, and proteomic data. Here, we introduce the proteome analysis of the axolotl tail section searched against an mRNA-seq database. We translated axolotl mRNA sequences to protein sequences and annotated these to process the LC-MS/MS data and identified 1001 nonredundant proteins. Functional classification of identified proteins was performed by gene ontology searches. The presence of some of the identified proteins was validated by in situ antibody labeling. Furthermore, we have analyzed the proteome expressional changes postamputation at three time points to evaluate the underlying mechanisms of the regeneration process. Taken together, this work expands the proteomics data of axolotl to contribute to its establishment as a fully utilized model.
Giriş ve Amaç:Rejenerasyon, fonksiyonel bir kayıp yaşamadan hasarlı dokunun, organın ya da uzvun restore edilmesi işlemidir. Su semenderlerinden olan Aksolotl kendisini öldürmeyecek iç organ, merkezi sinir sistemi ve uzuv hasarlarını başarı ile tamir edebilmektedir. miRNA'lar hedef mRNA'ların translasyon düzeyini düşürerek yazılım sonrası gen ifadesinin kontrolünde önemli roller üstlenirler. Biz de bu çalışmada Aksolotl'un kuyruk rejenerasyonu sırasında değişen miRNA profilini ve bu miRNA'ların moleküler yolaklara olası etkilerini araştırdık. Yöntem ve Gereçler: 6-8 aylık 60 adet Aksolotl'un kuyruk bölgesinden ampütasyon yapılmış ve hayvanlar 4 gruba ayrılmıştır. Ampütasyondan hemen sonra (0. gün) ve rejenerasyonun 1., 4. ve 7. günü örnek toplanmıştır. Toplanan örneklerden yeni nesil dizileme yöntemi ile miRNA tespiti yapılmıştır. Bulunan miRNA'lar içinde rejenerasyonun farklı evrelerinde miktarı anlamlı olarak değişenlerin hangi yolakları regüle ettiği analiz edilmiştir. Bulgular: Bulunan miRNA'lar içinden 52 tanesinin miktarı 1.5 kat ve üzeri değiştiği (arttığı veya azaldığı) için bu miRNA'lar yolak analizlerinde kullanılmıştır. Rejenerasyon ile birlikte artış gösteren miRNA'ların hemostazla ilişkili yolakları regüle ettiği görülmüştür. Rejenerasyon sürecinde azalış gösteren miRNA'ların ise hücre bölünmesi ve hücre bölünmesinin kontrolü ile ilgili yolakları regüle ettiği bulunmuştur. Tartışma ve Sonuç: miRNA'lar canlılıkta birçok görev (hemostaz, gelişim, büyüme ve hastalık durumunun regüla-syonu vb.) üstlenirler. miRNA profilinin değişmesi rejenerasyonun başarı ile sürmesi ve tamamlanması için gerekli bir mekanizmadır. Biz de bu çalışmada Aksolotl'un kuyruk rejenerasyonu sürecinde miktarı artan ve azalan miRNA'larının dinamik değişimini tanımlayıp, bu değişimin moleküler yolaklar üzerine etkisini araştırdık.
