SUMMARYSera of 1255 individuals from Novi Sad, varying in age from < 1 month to 69 years, have been tested for neutralization antibodies to Herpes simplex virus type 1.The eight newborns tested and 97 % of the 507 adults were positive, with titres ranging from 1/4 to 1/256. The titres in newborns were significantly lower than the titres in adults.After birth the maternal antibodies declined rapidly and 94 % of infants at the age of > 6 months and < 2 years were negative.After the first year infants in Novi Sad start to acquire herpes-neutralizing antibodies actively, reaching a 50 % incidence of positives between the 2nd and 3rd year of age. Age-specific incidence rates of herpes positives found in Novi Sad have been compared with those reported from Edinburgh, Freiburg i. Br. and Louisiana. Possible influences of several circumstances upon the incidence rate of positives detected by the neutralization test are discussed.
Zusammenfassung Durch vergleichende Untersuchungen prüften wir das Auftreten und die Intensität des zytopathogenen Effektes in Zellkulturen, die mit Tonsillen‐, Gehirn‐ und Lungenmaterial natürlich mit Aujeszky‐Viren infizierter Schweine beimpft worden waren. In 21 von 24 Fällen trat der zytopathogene Effekt in denjenigen Zellkulturen, die mit Tonsillensuspensionen beimpft worden waren, 24 Stunden p.i. auf. Die Zellzerstörung war hier eher und deutlicher sichtbar als in Kulturen, die mit Gehirn‐ bzw. Lungensuspensionen inokuliert worden waren. Nur in einem Fall zeigte sich der zytopathogene Effekt nach Verimpfung von Gehirnsuspension eher als nach Inokulation von Tonsillenmaterial. Bei 13 von 15 untersuchten Tieren lagen die Virustiter in den Tonsillen höher als in den Gehirnen bzw. Lungen der gleichen Schweine. In den Tonsillen schwankte der Virustiter zwischen 10−40 und 10−70 KID50, in den Gehirnen bzw. Lungen zwischen 0 und 10−5,50 KID50. Nur in einem Fall war der Titer in Tonsillen und Lungen gleich hoch. Unsere Untersuchungsergebnisse weisen darauf hin, daß sich das Virus bei natürlich infizierten Schweinen primär in den Tonsillen vermehrt und von dort aus in die Blutbahn eindringt. Im Verlauf einer Virämie gelangt es in das Zentralnervensystem, in die Lungen, den Uterus gravidis etc. In diesen Organen kann es pathologische Veränderungen hervorrufen. Wir halten es allerdings auch für möglich, daß das Virus nach intranasaler Infektion direkt auf dem Nervenweg das Zentralnervensystem erreicht. Für eine Virusvermehrung in den Tonsillen sprechen pathologische Veränderungen in Form verschieden großer nekrotischer Herde in den lymphatischen Follikeln. Unsere Befunde zeigen, daß zur Isolierung des Aujeszky‐Virus bei Schweinen neben Gehirn und Lungen besonders die Tonsillen herangezogen werden müssen. Dadurch gelangt man schneller zu einer sicheren Diagnose. Summary The significance of the tonsils for the pathogenesis and diagnosis of Aujeszky's disease in pigs In comparative studies the authors studied the development and intensity of the cytopathogenic effects (CPE) in cell cultures inoculated with tonsil, brain and lung material from pigs infected with Aujeszky virus. In 21 of 24 samples the CPE appeared in cultures inoculated with tonsil suspensions 24 hours previously. Cell damage was earlier and clearer than in cultures inoculated with brain or lung suspensions. Only on one occasion did the CPE occur earlier with brain suspension than with tonsil material. In 13 of 15 animals the viral titre in the tonsils was higher than in the brain or lung, the tonsil titres ranging between 10−4.0 and 10−7.0 KID50 and the levels in brain and lungs between 0 and 10−5,50 KID50. Only in one animal was the titre in the lungs as high as in the tonsils. These studies show that in naturally‐infected swine the virus multiplies primarily in the tonsils and from there reaches the bloodstream from which it invades the central nervous system, the lungs, and the gravid uterus.
