Herbal medicines are widely used in the complex treatment of various diseases. Therefore, theoretical and practical interest is the in-depth study of drumstick primrose (Primula denticulata Smith). The study aimed to determine the optimal extraction mode of flavonoids and reducing compounds of drumstick primrose leaves. The concentration of ethanol, the ratio of raw materials and extractant, and extraction method were studied by dispersion analysis. This allowed reducing the number of experiments from 64 to 16. To obtain the alcohol extract of drumstick primrose leaves with the highest content of reducing compounds and flavonoids, found that maceration is the optimal method of extraction, the ratio of raw materials to extractant should be 1 to 5 and 40% ethanol is the most appropriate extractant.

З давніх-давен у народній медицині використовують види роду Первоцвіт (Primula L.). Відо-мо близько 500 видів роду, поширених по всій земній кулі. У країнах СНД росте 67 видів. Найбільш поширений первоцвіт весняний (Primula veris L.; Primula officinalis Hill.). Відвар кореневищ з коренями первоцвіту весняного використовують у народній і науковій медицині як відхаркувальний засіб; настій листя -як знеболювальний і сечогінний. На фармацевтичному ринку України відсутні вітчизняні препарати з сировини первоцвіту весняного.Метою роботи було визначення оптимальних умов одержання витяжок з листя та з кореневищ з коренями первоцвіту весняного для розробки нових вітчизняних лікарських засобів з протизапальною та відхаркуваль-ною активністю.Матеріали та методи. Об'єктом для досліджень було листя первоцвіту весняного, яке заготовляли на те-риторії Тернопільської області у 2015 році у період масового цвітіння рослин, та кореневища з коренями, які за-готовляли після відмирання надземної частини рослини. Сировину подрібнювали до розміру часток 3-5 мм. Для екстрагування біологічно активних речовин з листків первоцвіту весняного були досліджені такі розчинники -20 %, 40 %, 70 % етанол та гаряча вода очищена (80 °С). Співвідношення сировина:екстрагент становило 1 : 10. Настоювали впродовж доби при кімнатній температурі, перемішуючи; фільтрували та згущували на роторному вакуумному випарнику за температури 60-70 °С до густого стану. З метою вибору екстрагенту для одержання субстанції з кореневищ з коренями первоцвіту весняного брали 20 %, 30 %, 40 % етанол. Додатково отримали витяжку при екстрагуванні 20 % етанолом, попередньо змочивши сировину 95 % етанолом. Співвідношення сировина:екстрагент становило 1 : 8. Екстракцію проводили настоюванням впродовж семи днів з примусовою подачею екстрагенту та з періодичним перемішуванням. Отримані витяжки фільтрували та згущували на ро-торному вакуумному випарнику. Кількісне визначення вмісту суми флавоноїдів, суми гідроксикоричних кислот та суми фенольних сполук, сапонінів проводили спектрофотометричним методом. Сухий залишок гравіметрич-ним методом визначали згідно з вимогами ДФУ, окиснювані феноли -перманганатометричним.Результати та їх обговорення. При вивченні впливу екстрагенту на ступінь вилучення біологічно актив-них речовин з листя первоцвіту весняного встановлено, що максимальна кількість суми гідроксикоричних кис-лот, окиснюваних фенолів і сапонінів екстрагується 40 % етанолом, що становить 25,5 %, 12,13 %, 3,25 % відпо-відно; суми фенольних сполук і суми флавоноїдів -70 % етанолом (5,40 % і 12,70 % відповідно); екстрактивних речовин -гарячою водою очищеною (48,51 %). Introduction. Herbal medicines are increasingly being used in the treatment of various diseases. Significant number of resources, high-level availability and the possibility of cultivation have provided high prospects of plant raw materials in the development of new herbal medicines. Primula denticulate Smith. is one of the most interesting medicinal plant raw materials source in modern medicine and pharmacy with the wide spectrum of pharmacotherapeutic action, that is mainly used only in traditional medicine. Thus, nowadays the development of new and effective medicines based on Primula denticulate Smith. in tablet dosage form is the topical task of pharmacy. The aim of the work. To study the influence of excipients on the pharmacotechnological parameters of tablets with Primula denticulate Smith, are obtained by wet granulation method and to substantiate the choosing of the best excipients by the method of mathematical planning of the experiment. Materials and Methods. It was used a self-prepared dense extract of Primula denticulate Smith. and excipients, which are complied with the requirements of State Pharmacopoeia of Ukraine in terms of pharmaco-technological quality parameters, for developing tablets. The hyper Greek-Latin square 4x4 was used to study the four qualitative factors. Results and Discussion. In the course of the experiment, it was determined the dependence of pharmacotechnological parameters (friability, resistance to crushing and disintegration of tablets) on type of excipients. It was selected excipients with the optimal pharmaco-technological parameters for further research the tablets quality. Conclusions. According to the method of mathematical planning of the experiment, it’s chosen the optimal excipients for obtaining tablets based on dense extract of Primula denticulate Smith. Considering the investigated pharmaco-technological properties (friability, resistance to crushing and disintegration of tablets) the qualitative composition of excipients for obtaining tablets by the method of wet granulation was determined: Avicel PH - 105 and MCC 101, croscarmellose sodium, colloidal anhydrous silicon dioxide, magnesium stearate and Prosolv 90.
Вступ. Створення нових фітопрепаратів та вдосконалення технологій їх виробництва – важливий напрямок фармацевтичної науки і практики. Екстрагування сировини є складним фізико-хімічним процесом. На нього впливає багато чинників. Вагомим є вплив ступеня подрібнення рослинної сировини на інтенсифікування процесу екстрагування. Мета дослідження – визначити вплив ступеня подрібнення рослинної сировини на вилучення флавоноїдів та поліфенолів із примули дрібнозубчастої листків. Методи дослідження. Кількісний вміст флавоноїдів та поліфенолів у витяжках, отриманих із сировини зі ступенем подрібнення 2 мм, 3 мм, 4 мм і 5 мм, визначали спектрофотометричним методом на спектрoфoтoметрі Lambda 25 Perkin Elmer (США). Результати й обговорення. Найбільшу кількість флавоноїдів екстраговано з витяжок із ступенем подрібнення примули дрібнозубчастої листків до 4 мм, їх вміст становив 344,82 мкг/мл. Найоптимальнішим був ступінь подрібнення сировини для вилучення поліфенолів 2 та 4 мм. Враховуючи результати попередніх досліджень, у ході яких встановлено, що фракція 3–4 мм найменше поглинає екстрагенту, вирішено для оптимізації технології використовувати сировину зі ступенем подрібнення 4 мм. Висновки. Спектрофотометричним методом встановлено кількісний вміст флавоноїдів та поліфенолів у витяжках із примули дрібнозубчастої листків. Вміст флавоноїдів у витяжках, одержаних із сировини зі ступенем подрібнення 2, 3, 4 та 5 мм, становить 328,48, 305,00, 344,82 і 293,39 мкг/мл відповідно. Кількісний вміст поліфенолів у витяжках, отриманих із сировини зі ступенем подрібнення 2, 3, 4 та 5 мм, складає 411,88, 396,25, 386,34 і 357,95 мкг/мл відповідно. Найоптимальнішим є ступінь подрібнення примули дрібнозубчастої листків 4 мл, при якому вилучається найбільше флавоноїдів та поліфенолів і витрачається найменша кількість екстрагенту.
