This study was focused on analyzing the effects of dietary inclusion of raw chickpea seed as a replacement of soybean meal as a primary protein source on bone structure in broiler chickens. Broiler chickens (n = 160) received in their diet either soybean meal (SBM) or raw chickpea seeds (CPS) as a primary protein source throughout the whole rearing period (n = 80 in each group). On the 42th day randomly selected chickens from each group (n = 8) were slaughtered. Collected tibiotarsus were subjected to examination of the biomechanical characteristics of bone mid-diaphysis, microstructure of the growth plate and articular cartilages; the analysis of mineral content and crystallinity of mineral phase, and the measurements of thermal stability of collagen in hyaline cartilage were also carried out. The inclusion of chickpea seeds resulted in increase of bone osteometric parameters (weight, length and mid-diaphysis cross-sectional area) and mechanical endurance (yield load, ultimate load, stiffness, Young modulus). However, when loads were adjusted to bone shape (yield and ultimate stress) both groups did not differ. Mineral density determined by means of densitometric measurements did not differ between groups, however the detailed analysis revealed the differences in the macro- and microelements composition. The results of FT-IR and XRD analyses showed no effect of diet type on mineral phase crystallinity and hydroxyapatite nanocrystallites size. In trabecular bone, the increase of real bone volume (BV/TV) and number of trabeculae was observed in the CPS group. Total thickness of articular cartilage was the same in both groups, save the transitional zone, which was thicker in the SBM group. The total thickness of the growth plate cartilage was significantly increased in the CPS group. The area of the most intense presence of proteoglycans was wider in the SBM group. The structural analysis of fibrous components of bone revealed the increase of fraction of thin, immature collagen content in articular cartilage, trabeculae and compact bone in the CPS group. The dietary inclusion of CPS affected the thermal stability of collagen, as decrease of net denaturation enthalpy was observed. This study showed a beneficial effect of CPS on the skeletal development, improving the overall bone development and the microarchitecture of cancellous bone. It suggests that CPS can be a promising replacement for SBM in broilers feeding in the aspect of animal welfare related to the development of the skeletal system.
Simple Summary: A soybean meal, commonly used as a primary source of protein in animal diets, is often obtained from processing genetically modified soybean varieties. The tendency to reduce the use of feeds containing transgenic constituents increases the need for alternative sources of dietary protein. In this study, we fed broiler chickens diets containing either soybean meal or raw chickpea seeds. We examined the effect of such a substitution on broilers' subcutaneous fat tissue, which has numerous important physiological functions in chickens.Abstract: In this study, the effect of soybean meal substitution by raw chickpea seeds on the thermal properties and fatty acid profile of subcutaneous fat tissue of broiler chickens was examined. The experiment, performed on Ross 308 chickens, lasted for 42 days. Tight subcutaneous fat tissue was analyzed using differential scanning calorimetry (DSC) measurements while the fatty acid composition of subcutaneous adipose tissue was determined chromatographically. There was no effect of soybean meal substitution on fat crystallization temperature or crystallization enthalpy. However, the total calorimetric enthalpy of the melting of low-melting monounsaturated and saturated triacylglycerols differed between groups. Fatty acid proportions in the subcutaneous fat tissue of broiler chickens were also altered. Among others, chickpea seed inclusion decreased the content of main saturated acid (palmitic acid) and increased the content of main monounsaturated (oleic) and tri-unsaturated (linolenic) acids. The results show that the soybean meal substitution by raw chickpea seeds in the feed can affect the structural properties of adipose tissue in broiler chickens, including the thermal transformation of unsaturated fatty acids. Due to the numerous physiological functions of subcutaneous fat tissue, understanding these mechanisms can promote the use of alternative protein both in poultry and human nutrition.
Currently, it is important in aquaculture to study and use products and additives, which have positive effects on the digestibility of nutrients by helping to normalize the intestinal microflora, thereby modulating fish immune response. The aim of the study was to assess the effect of a prebiotic based on mannan oligosaccharides produced from the outer walls of Saccharomyces cerevisiae cells on the histological structure, morphometric state of the intestine and activity of cellular and humoral components of nonspecific resistance of age -2+ carp. The experiment was conducted in four analogous ponds, which were stocked with age-1 scaly carp with an average weight of 55–58 g with a stocking density of 1000 fish/ha. Carp had been fed balanced compound feed for 60 days. Experimental groups of fish were fed with a prebiotic as a feed additive in the amount of: 0.025 % – Experiment 1, 0.05 % – Experiment 2 and 0.075 % – Experiment 3. Use of the studied prebiotic did not damage the intestinal histological structure of carp of the experimental groups. The villi height and intestinal crypt depth of carp in Experiment 1 (P < 0.05) and Experiment 2 (P < 0.01) were larger compared to the control group. Experiment 3 showed destructive changes in intestinal mucosa. The results of histological examination indicated the effectiveness of the use of the studied prebiotic in the amount of 0.025 and 0.05 % in the diet of age -1+ carp due to the absence of pathological changes and active morphofunctional state of the intestine, which, in turn, had a positive effect on feed absorption and metabolic processes in fish body. Analysis of the indicators of cellular and humoral links of natural resistance showed that in carp of the second and third experimental groups lysozyme activity of serum was 5 and 11 % (P < 0.001) higher than in the control group. Similar changes, though found in a greater extent, were recorded in the study of bactericidal activity of blood serum, especially in individuals of the second and third experimental groups (by 6.8 (P < 0.01) and 15.2 %). While with carps of the third experimental group, which used a prebiotic drug in the amount of 0.075 %, the content of CEC was 20.5 % higher than in the control group, which indicates an additional antigenic load on the body and is an unfavorable diagnostic factor. At the same time, with the fish of the second experimental group, which received, respectively, 0.05 % of the supplement of the drug “Actigen” to the main diet, probably higher phagocytic activity of blood neutrophils was recorded. A direct dependence of the growth of the phagocytic index and the phagocytic number of neutrophils on the dose of the studied prebiotic was detected. Immunological studies indicated a dose-dependent effect of the prebiotic on the activity of cellular and humoral components of non-specific resistance of carp. Given this, for the effective functioning of innate protective mechanisms, for the increase of the body's immune potential and productivity, it is most appropriate to apply to the diet of carp prebiotic supplement “Actigen” in the amount of 0.025 and 0.05 %.
The article presents the results of a study of the acute and subacute toxicity of the drug TILOTAR (a water-soluble powder for oral administration) made on the basis of tylosin tartrate. As a result of the research it was found that the drug "TILOTAR" refers to the 4th class of toxicity, that is, to low-toxic substances. After intragastric administration of the drug to white mice and rats at doses of 2500, 5000, 7500 mg/kg, all animals remained alive. Short-term inhibition was recorded in laboratory animals, which were given the drug at the maximum dose. Further changes in the clinical state of the animals of the experimental groups were not observed. The study of subacute toxicity of the drug "TILOTAR" showed that 14-day administration of a therapeutic dose of the drug leads to a decrease in erythropoiesis, the appearance of old forms of erythrocytes, damage to hepatocytes, the appearance of eosinophils, γglobulins, a decrease in protein biosynthesis, increased kidney function. The introduction of a 10-fold dose within 14 days reduced the body weight of the rats, leading to abnormalities in the liver and kidneys, and a decrease in the mass of the spleen. An increase in urea blood concentration and a decrease in creatinine may indicate impaired renal function in animals that were given a tenfold dose of the drug, and an increase in the level of α-and γ-globulins against the background of a decrease in body weight of rats indicates an overload of the body's immune system. In rats of the therapeutic group, homeostasis was maintained by enhancing kidney function, accompanied by a decrease in the number of erythrocytes due to young forms and the appearance of mature erythrocytes with a high level of hemoglobin (MCH), some destruction of hepatocytes (high level of alkaline phosphatase activity) while reducing the amount of albumin, an increase in relative levels of α-and γ-globulins. In the conducted experiments it was established that the macroscopic and microscopic structure of the internal organs of rats, while studying the subacute toxicity of the drug "TILOTAR", was preserved, no macroscopic changes were revealed. In rats receiving a 10-fold therapeutic dose of the drug for 14 days, focal protein dystrophy of the liver and kidneys was histologically established. 1 Державний науково-дослідний контрольний інститут ветеринарних препаратів та кормових добавок, м. An important feature of living organisms, acquired in the process of evolution, is the ability to adapt to various external influences and changes in the environment, to maintain homeostasis and regulatory processes in organs, systems and the body as a whole. However, with modern breeding technologies, the physiological capabilities of the animal's body are often unable to change as quickly as required by the conditions of industrial maintenance, which leads to stress. Vitamin preparations and biologically active feed additives are widely used by veterinary medicine specialists to increase resistance to stressful situations, disease prevention, increase resistance, safety and productivity of farm animals. Preparation L-tsyn, solution for injection (manufactured by BIOTESTLAB LLC), is a drug, the action of which is due to the prescription combination of active substances, namely: butaphosphane, B vitamins (vitamin B12, vitamin B3) and L-carnitine. The aim of the study was to investigate the effectiveness of the drug L-tsyn on horses under the influence of stressors of various natures. Two experiments on working horses with constant, excessive physical activity with chronic exercises and with complex therapy of intoxication with signs of acute renal failure in horse were conducted. Clinical trials of the drug were performed on working horses aged 5–15 years, of different sexes. The drug was administered in a dose of 20–25 ml per animal, 1 time per day for 5 consecutive days. The clinical condition of the experimental animals was observed for 21 days from the start of the drug. The morpho-functional state of the animal's body before and after the drug was established according to the indicators of the clinical condition, morphological and biochemical parameters of the blood, which were determined according to generally accepted methods. In working horses under excessive load the drug L-tsyn activated the processes of hematopoiesis, improved the physiological state of the body and contributed to the increase the general tone of the body. The drug as part of complex therapy in the treatment of intoxication, showed hepato- and nephroprotective properties, reduced the manifestations of intoxication, helped to restoration of the animal’s normal physiological state. According to the results of clinical, hematological and biochemical studies, it can be concluded that the drug L-tsyn has a stimulating effect on metabolic processes in the animals’ body, shows tonic properties, increases the body's resistance to toxins and adverse environmental factors and can be used as monotherapy and for the complex treatment of diseases of various etiologies in horses.
