Natural Occurrence of Beauveria bassiana (Bals.) Vuilleman (Moniliales: Moniliaceae) on the Lesser Mealworm, Alphitobius diaperinus (Panzer) (Coleoptera: Tenebrionidae), in a Poultry House in Cascavel, PR ABSTRACT -One of the main problems in poultry houses in Brazil is the lesser mealworm, Alphitobius diaperinus Panzer. This insect inhabits the litter, manure and soil of poultry houses, feeding on bird manure, cadavers and spilt feed. The lesser mealworm serves as a reservoir of many animal pathogens. Chickens also feed readily on the beetles in infested litter and this sometimes causes nutritional problems and affects weight gain. Current control measures are generally unsatisfactory and microbial control may be a promising strategy to control A. diaperinus. Natural occurrence of entomopathogens is very important for the natural control of insect pests and this paper records the occurrence of Beauveria bassiana (Bals.) Vuill. on the lesser mealworm in commercial poultry houses, in Brazil. The evaluation was carried out weekly for six months (September/2002 to February/2003. The soil around the poultry house was collected and transferred to Zoology Laboratory from Unioeste, at Cascavel, PR. Infected larvae and adults were always present, but infected pupae were registered twice. Larvae were more susceptible than pupae and adults (incidence ranging from 49% to 100%, mean 74%, 33% to 50%, mean 6.9%; 0.9% to 100%, mean 40.7%, respectively). The pathogenicity of the fungus was confirmed under laboratory conditions following procedures outlined in Koch's postulates. O inseto infesta os aviários, transmite doenças às aves e afeta seu desenvolvimento quando ingerido no lugar da ração. Seu controle é dificultado pelos hábitos crípticos e pela presença constante das aves nos aviários e o controle microbiano pode ser uma alternativa viável para o problema. Sabe-se que a presença de entomopatógenos no ambiente contribui para o controle natural dos insetos-praga. Este trabalho relata a ocorrência natural de Beauveria bassiana (Bals.) Vuill. sobre larvas e adultos do cascudinho em aviários comerciais no Brasil. O experimento foi realizado de setembro/2002 a fevereiro/2003, com amostragem semanal de insetos no solo periférico de um aviário comercial em Cascavel, PR. O fungo esteve presente em todas as avaliações, infectando principalmente larvas (49% a 100%, média de 74,4%) e adultos (0,9% a 100%, média de 40,7%) encontrados no solo, sendo que pupas infectadas ocorreram apenas em duas avaliações (33% a 50,0%, média de 6,9%). O isolado de fungo obtido nos cadáveres foi identificado como sendo B. bassiana e teve sua patogenicidade ao cascudinho comprovada pelo postulado de Koch, em condições de laboratório. The purpose of this paper was to evaluate the liquid culture media for the production of Metarhizium anisopliae (IBCB 425) and Beauveria bassiana (IBCB 66), as well as the tolerance of these seedlings to the ultraviolet action and to the temperature. Twelve treatments composed of combinations between carbon and nitrogen concentrations were assessed. In order to determine the effect of ultraviolet radiation, plates with blastos-pores were exposed to it for 25 and 50 seconds. To determine the temperature efect, blastospores from culture media were exposed to 20, 25, 30 and 35°. For the virulence experiments, caterpillars of Diatraea saccharalis were sprayed with 2 mL of fungal suspension with the aid of a Potter tower. Te best media for M. anisopliae are 16.00 g (carbon) + 7.00 g (nitrogen) and 14.40 g (carbon) + 7.00 g (nitrogen), whereas for B. bassiana: 20.00 g (carbon) + 6.30 g (nitrogen) and 20.00 g (carbon) + 7.00 g (nitrogen). Te longer the exposure to ultraviolet radiation, the smaller the number of colonies. At 35°, there is a significant decrease in the formation of colonies. Te produced seedlings of fungi are pathogenic to D. saccharalis.
RESUMO O trabalho teve os seguintes objetivos: 1) avaliar diferentes substratos para a produção semissólida dos fungos Metarhizium anisopliae e Beauveria bassiana; 2) verificar a tolerância dos conídios produzidos ao efeito da radiação ultravioleta e da temperatura; e 3) avaliar a patogenicidade dos conídios a Diatraea saccharalis. Para ambos os fungos, foram utilizadas 6 repetições para cada tratamento: amido de milho, arroz integral, arroz parboilizado tipo1, arroz tipo 1 e 2, aveia em flocos, canjiquinha, farelo de trigo, farinhas de mandioca crua, de milho amarela e de trigo especial, fubá, milho em grãos, polvilho azedo, soja em grãos, trigo moído e turfa. Os conídios foram quantificados em câmara de Neubauer e a determinação da viabilidade foi realizada através da observação em microscópio, dos conídios germinados e não germinados, após espalhamento da suspensão fúngica em placas de Petri contendo BDA. No ensaio com radiação os fungos foram expostos à radiação por 25 e 50 segundos e foi considerado um tratamento sem exposição; para temperatura os fungos foram expostos a 20, 25, 30 e 35o C. Utilizando a torre de Potter, 2 mL de cada suspensão fúngica dos tratamentos foram pulverizados em lagartas de D. saccharalis. As maiores concentrações de conídios de M. anisopliae e B. bassiana foram encontradas nos tratamentos com arroz parboilizado tipo 1, arroz tipo 1 e 2, farinha de milho amarela, fubá e trigo moído e a viabilidade dos conídios produzidos superou os 94%. Quanto maior o tempo de exposição à radiação menor foi o número de conídios viáveis; também ocorreu perda significativa da viabilidade dos conídios quando expostos à temperatura de 35o C. Os fungos foram patogênicos para D. saccharalis.
RESUMO O efeito de oito inseticidas e doze fungicidas, utilizados na cultura da goiaba, foi avaliado sobre o fungo Beauveria bassiana sob condições in vitro e de semi-campo. Em laboratório, o efeito foi verificado misturando-se os agrotóxicos ao meio de cultura BDA, nas concentrações recomendadas, e os parâmetros avaliados foram o diâmetro médio das colônias, o número de conídios produzidos e a virulência sobre Galleria mellonella. Em condições de semi-campo, preparações dos produtos químicos e do entomopatógeno foram pulverizadas em mudas de goiaba de três diferentes formas, a saber: primeiramente os produtos químicos e depois o fungo; primeiro o fungo e posteriormente o produto químico e os dois simultaneamente. Após a aplicação, folhas foram coletadas e lavadas obtendo-se uma suspensão que foi plaqueada em BDA para avaliação da esporulação, viabilidade e virulência dos conídios dos fungos. Os resultados mostraram que, in vitro, somente o inseticida Actara 250 WG foi compatível com o fungo e os demais produtos foram classificados como moderadamente compatível, tóxicos e muito tóxicos. Para os testes de semicampo, a forma de aplicação interferiu no desenvolvimento do fungo e, além disso, Cuprozeb PM, Manzate 800 PM e Cerconil PM não afetaram o entomopatógeno em nenhum dos parâmetros avaliados, portanto são indicados para serem utilizados num programa de manejo integrado de pragas. Nos testes de virulência, os conídios não tiveram sua viabilidade afetada, mostrando-se patogênicos a traça-dos-favos, G. mellonella.
RESUMO Este trabalho teve como objetivo avaliar, em condições de laboratório e estufa, a compatibilidade de Lecanicillium lecanii, JAB 02, com agrotóxicos. A compatibilidade em laboratório foi verificada misturando-se os agrotóxicos ao meio de cultura BDA e os parâmetros utilizados para a avaliação foram: crescimento vegetativo, esporulação e viabilidade do entomopatógeno. Em condições de estufa, as suspensões dos agrotóxicos e, posteriormente, a do fungo foram pulverizadas em plantas de crisântemo. Após a aplicação, folhas foram coletadas e lavadas obtendo-se uma suspensão que foi plaqueada em BDA, sendo então avaliado o crescimento das colônias do fungo. Verificou-se, em laboratório, que a maioria dos inseticidas e os acaricidas foram compatíveis ao fungo, com exceção do inseticida Thiodan® classificado como muito tóxico. Todos os fungicidas testados foram classificados como tóxico e muito tóxico. Em condições de estufa, no tratamento com o fungicida Rovral® foi observado um número de colônias fúngicas formadas compatível com a testemunha, não afetando o desenvolvimento do fungo. Essa compatibilidade repetiu-se para os produtos Alto 100® e Thiodan® em alguns dos tempos avaliados. Os produtos Rovral® e Alto 100® podem ser utilizados, conjuntamente, quando aplicados previamente ao entomopatógeno.
