The experiment was conducted at the Tissue Culture Laboratory as well as in the net house of Oilseed Research Centre, Bangladesh Agricultural Research Institute (BARI), Joydebpur, Gazipur on microspore culture and the performance of microspore derived doubled haploid (DH) in Brassica juncea (L.). The variety BARI Sarisha II of B. junccea was used for microspore culture and 8 DH genotypes viz., BJDH 01, BJDH 05, BJDH ii, BJDH 12, BJDH 14, BJDH 17, BJDH 18, and BJDH 20 along with their parent Rajat collected from the Plant Research International, the Netherlands were used for performance evaluation. Four-day cold treatment at 6 °C along with bud size 3.0-3.1 mm of BARI Sarisha II showed better response in the embryonic development. Significant variation was observed for the different yield contributing characters among the DH genotypes along with their parent Rajat. All the DH genotypes were late both for days to flower and days to maturity compared to the parent. Most of the DH genotypes produced higher seed yield than the parent. The genotypes BJDII 14 (7 g) and BJDH 01(7 g) were bold seeded and almost double or more in 1000-seed weight compared to the parent. Total oil content of the DH genotypes (40.64-42.28) were significantly different from the parent (3 g). Oil content of BJDH 01 was almost 3% higher than the parent could be selected to incorporate in the future breeding program.
Cellulose is the richest plant biomass on earth and is an unbranched polymer composed of D-glucose residues joined by β-1, 4-D-glycosidic bonds. The utmost abundant carbohydrates in nature are considered to be cellulases and hemicellulases. Cellulases are inducible enzymes that catalyze the hydrolysis of a β-1, 4-glycosidic bond to release the glucose units in a cellulose molecule. Thermophilic cellulases are relatively expensive and a very significant industrial enzyme. In this study, the recombinant plasmid pET22b (+) containing the cellulase encoding gene was transformed in E.coli BL21 codon plus. A Shake flask fermentation study was performed using modified M9NG media. Lactose and IPTG were used as an inducer. After SDS-PAGE analysis, the predicted molecular weight of a protein was 62kDA Batch culture fermentation was performed using LB and modified M9NG media. Lactose was used as the cheapest inducer. Under optimized fermentation conditions, the enzyme displayed maximum activity at 37oC and pH 7. The specific activity of the enzyme was 70U/ml. The production of the recombinant enzyme was enhanced approximately 6 times in E.coli BL21 as compared to wild type strain. The expression level of the recombinant cellulase was round about 30%-40%. Molecular cloning of the cellulase encoding genes resulted in the maximum production of the cheapest enzymes that can be used for industrial purposes.
Trehalose is a stress-response disaccharide commonly found in yeasts, bacteria, plants, and invertebrates. In plants, its purpose is uncertain, but some studies suggest it protects against abiotic stressors like chilling, heat, drought, and salinity. It is an osmoprotectant and a stabilizing agent for higher animals and plants. Some trehalose metabolism mutants show growth abnormalities, which could compromise plant physiology. Biotic stress causes contradictory results as well. It is also crucial for pathogen infectivity and plant defensive response. In our opinion, trehalose should not be considered only as a defensive sugar but rather as a molecule with two identities. It is an exceptional molecule that maintains the plant's vigor despite severe environmental conditions. Arabidopsis plants have been used to treat Alzheimer's, Parkinson's, and Huntington's diseases. There's no doubt that what we have just learned about this disaccharide will take us down many roads we've never been down before.
Barley (Hordeum vulgare L.). Sprouting has been used to improve the nutritional value of grains. The main objective of this study was to analyze the Physical analysis (pH, Acidity, TSS, Sensory Attributes, Color, Flavor, Taste) analysis of Lemon Barley RTD. Fine powder of barley Haider-93 utilized with combination of Sprouted barley powder to develop Lemon (RTD).Sensory evaluation of product was done on nine point hedonic scales. The product was packed in PET bottles and was evaluated for 90 days storage. Data was evaluated statistically and were interpreted.
