Urinary excretion of purine derivatives (PD) was used to estimate the microbial N (MN) supply to sheep in three experiments designed to examine the effects of DMI and BW on the efficiency of microbial N supply (EMNS) to the host animal. In Exp. 1, four sheep of about 45 kg BW were given 328, 656, 984, and 1313 g of DM/d of a hay/concentrate diet in a Latin square design. Excretion of PD per kilogram of digestible organic matter intake (DOMI) increased with intake, and EMNS increased from 12.0 to 28.3 g of MN/kg of OM digested in the rumen (DOMR). In Exp. 2, 19 sheep ranging from 22 to 73 kg BW were all offered 820 g of DM/d of the same diet as that fed in Exp. 1. Although DM digestibility was relatively constant, PD excretion varied from 4.5 to 13.5 mmol/d and EMNS from 8 to 36 g of MN/kg of DOMR, both inversely related to animal BW. In Exp. 3, five sheep of 48 to 57 kg BW were given a different diet at 702, 966, or 1,237 g of DM/d. Purine derivative excretion per kilogram of DOMI increased with the DMI:BW ratio. Calculated EMNS ranged from 23 to 35 g of MN/kg of DOMR. Pooled data from all experiments showed EMNS to be related to the DMI:BW ratio. It is suggested that the DMI:BW ratio defines the ruminal digesta passage rate and hence outflow of microbial protein. The results imply that the EMNS for a given diet is not constant, but changes with intake.

Young growing rats weighing approximately 83 g were fed on diets containing kidney bean (Phaseofus vulgaris) lectin (phytohaemagglutinin, PHA) in the range of 0 4 4 5 g / k g body weight for 10d to ascertain whether there was a minimum dose below which the lectin had no significant effect on body and skeletal muscle weights in comparison with pair-fed lectin-free controls. Averaged over all experiments, PHA doses of less than 10 mg/d (012 g/kg body weight) reduced body dry weight by 1.14 (SE 0.25) g when compared with controls. Between 10 and 27 mg/d ( 0 1 2 4 3 2 g/kg body weight) a further reduction of 0-64 (SE 0.21) g occurred, suggesting a slight but steady decline of body dry weight with increasing dose. However, above 27 mg/d the depression of growth and changes in body composition accelerated. The difference between the proportional losses of skeletal muscle and body weight was not significant at doses of PHA below 10 mg/d (0.12 g/kg body weight) but the ratio of these losses rose to 1.5-2.0 at doses above this. The proportional decrease in lipid weight exceeded that of both body and skeletal muscle weights at all lectin doses, suggesting that lipid catabolism was the first target of the PHA effect.Plasma insulin level was depressed at the PHA dose of 0.02 g/kg body weight at which growth depression and muscle atrophy were minimal but, contrary to expectations, plasma glucose levels remained stable over the whole PHA dose range. It appears that despite a PHA-induced lowering of blood insulin, glucose catabolism is elevated by an unknown, possibly hormonal, compensatory mechanism. Thus, because low insulin levels facilitate the mobilization and catabolism of lipids, it may be possible to use low doses of PHA to reduce hyperglycaemia and body fat.

Experiments were conducted to compare the rates of apparent absorption (disappearance) of individual essential amino acids (EAA) from the small intestine with their net fluxes across the mesenteric- (MDV) and portal- (PDV) drained viscera in sheep given a pelleted alfalfa diet at two levels of intake. Disappearances of individual EAA across the region of the small intestine drained by the mesenteric arcade (jejunum to ileum) were similar to those across the whole of the small intestine (duodenum to ileum). The net MDV flux of each EAA was similar to its rate of disappearance, but, with the exception of threonine on the low intake level, the net PDV flux was lower (P < .05). Increasing the intake of alfalfa from 800 to 1,200 g/d increased the rate of disappearance of individual EAA between the duodenum and ileum by .56 (range .43 to .65) and between the jejunum and ileum by .51 (range .45 to .60). The MDV and PDV blood flows increased by .35 and .39, respectively, and, with the exception of valine, net fluxes of individual EAA increased by .39 (range .20 to .50) across the MDV and by .44 (range .21 to .71) across the PDV. When net fluxes across the MDV and PDV were measured simultaneously, the ratio of PDV: MDV flux for each EAA was less than (P < .05) unity (mean .61, range .55 to .69), even though all MDV blood enters the PDV, contributing approximately .45 of the total portal flow. This observation suggests that, in regions of the tract not drained by the MDV, extraction of arterial blood EAA for tissue and secretory protein synthesis must exceed the release of protein degradation products into the venous drainage. The results are discussed in terms of endogenous protein secretions into the lumen of the tract anterior to the small intestine.

Although the importance of root production and mortality to nutrient fluxes in ecosystems is widely recognized, the difficulties associated with root measurements have limited the availability of reliable data. We have used minirhizotrons and image analysis to measure root longevity of Prunus avium L., Picea sitchensis (Bong.) Carrière, Acer pseudoplatanus L. and Populus x canadensis cv. Beaupre directly in cohorts of roots. Major differences in the longevity of roots among species were identified. For example, 40% of Prunus avium roots but only 6% of Picea sitchensis roots survived for more than 14 days. Survival analysis of cohorts of roots of Prunus avium and Populus x canadensis revealed differences in the distribution of longevity among cohorts. Genetic, biotic and abiotic factors that may influence longevity are discussed.
