Heat stress (HS), one of the major problems of tropical and subtropical countries, adversely affects the production performance of poultry. Keeping this in view, the present study was designed to investigate some of the biological markers of HS in broilers as modulated by dietary supplementation of mannan-oligosaccharide (MOS) and a Lactobacillus-based probiotic (LBP), either alone or in combination. Two hundred fifty 1-d-old-chicks were randomly divided into 5 groups. From d 22, the birds were either kept at the thermoneutral zone (TN) or exposed to HS to the conclusion of study, d 42. Birds were fed either a corn-based basal diet (TN and HS groups) or the same diet supplemented with 0.5% MOS (HS-MOS group), 0.1% LBP (HS-LBP group), or their combination. Birds were immunized against Newcastle disease virus on d 4 (intraocular; live attenuated) and d 20 (drinking water; live attenuated) and infectious bursal disease virus on d 8 (intraocular; live intermediate strain) and d 24 (drinking water; live attenuated). Birds were killed on d 42 to collect serum for determination of cortisol, thyroid hormones, cholesterol, C-reactive protein, and postvaccinal antibody titers. Results revealed that dietary supplementations decreased (P < 0.05) the serum cortisol and cholesterol concentrations and increased (P < 0.05) thyroxine concentration compared with the HS group without affecting triiodothyronine concentration. The percentage of the C-reactive protein-positive birds was higher (P < 0.05) in the HS group compared with the TN group. Dietary supplementations improved humoral immunity against Newcastle disease virus and infectious bursal disease virus during HS. In conclusion, dietary supplementation of either MOS or LBP alone or in combination can reduce some of the detrimental effects of HS in broilers.

The objective of the present study was to determine the relationship of age to body weight (BW), scrotal circumference (SC), number of pixels of testicular ultrasonograms (NP), and semen quality in Sahiwal bulls. The study was based on 128 Sahiwal bulls of different age groups (from 0 to >100 months of age). Bulls were evaluated for SC, BW, and NP. Semen was evaluated once a week for five consecutive weeks from regularly collected donor bulls (n = 86) ranging in age from 25-30 to >100 months. Ejaculate volume, sperm motility, sperm concentration, sperm morphology, percent live sperms, sperm plasma membrane integrity, and normal acrosome were compared among different age groups. Mean SC and BW increased (P < 0.05) in a curvilinear manner from birth to >100 months of age. Mean NP of testicular ultrasonograms increased (P < 0.05) from 0 to 24 months and then plateaued until >100 months of age. Body weight, SC, and NP were positively correlated with age from birth until >100 months (r = 0.91, 0.87, and 0.40, respectively). Ejaculate volume (5.7 ± 0.2 vs. 4.6 ± 0.09 ml) and sperm concentration (1,281.6 ± 17.7 vs. 1,115.8 ± 55.9 × 10(6)/ml) increased (P < 0.05) in mature bulls compared to younger ones. However, motility (68.6 ± 0.3%), plasma membrane integrity (50.8 ± 1.0%), and normal acrosome (74.8 ± 0.5%) remained insignificant due to age. In six of eight age groups studied, morphological abnormalities were well within the range (18.1 ± 0.3%). In conclusion, the BW, SC, and NP of testicular ultrasonograms, ejaculate volume, and concentration increased with age. Moreover, semen quality is fairly independent of age except volume and concentration in Sahiwal bulls.

1. Seven 35-week-old Hubbard broiler breeder males were subjected to three semen collection frequencies either once every 2 d (48 h), daily (24 h) or twice daily (12 h). 2. Semen characteristics including motility, volume, concentration and sperm numbers per ejaculate were determined for each ejaculate. 3. Sperm motility was unaffected by collection interval, but semen volume was lower at 12 than 24h intervals. Sperm concentration was lower at 12 than 48h intervals. 4. At 24 and 48 h number of sperm per collection (1.7+/-0.2, 1.8+/-0.2 x 10(9)) were higher than at 12 h (1.2+/-0.1 x 10(9)). 5. The number of semen doses over a 6-d period increased linearly as the frequency of collection increased from once every 2 d to twice daily. 6. It is concluded that output was theoretically maximal at twice daily collection, but in practice not all cockerels may be able to maintain full performance with such a demanding regime.

The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, which measures the reduction of MTT, is commonly used to validate the viability of metabolically active cells. This study was conducted to evaluate and validate the MTT assay to assess the spermatozoal viability of Nili-Ravi buffalo bulls and compare the efficiency of the test with the supravital staining technique (eosin and nigrosin) and the hypoosmotic swelling test. Fresh semen samples from breeding Nili-Ravi buffalo bulls (n = 25) were collected using an artificial vagina. After assessing the quality of the semen for routine variables, the MTT assay was carried out in PBS. Results revealed a correlation (r = 0.979; P < 0.001) between the viability of spermatozoa and the rate of reduction of MTT. The other proportions of same semen samples showed a poor relationship between the eosin and nigrosin test (r = -0.25), the hypoosmotic swelling test (r = -0.12), and motility (r = -0.15). However, the MTT assay was found to be superior compared with other tests because it was able to determine those spermatozoa that were more than 90% viable. In conclusion, the MTT assay is a simple, robust test that can be used to select Nili-Ravi buffalo bulls on the basis of spermatozoa quality.

Introduction Metformin is one of the safest, first-line oral hypoglycemic agents used in type-2 diabetes mellitus patients. This study aims to study the effect of metformin on thyroid-stimulating hormone (TSH) in hypothyroid and euthyroid individuals, as both these diseases have an increased prevalence and coexistence. Method This hospital-based study was conducted in Jinnah Allama Iqbal Institute of Diabetes and Endocrinology (JAIDE), Allama Iqbal Medical College/Jinnah Hospital Lahore, Pakistan, from October 2019 to April 2020. One hundred and sixty type-2 diabetic participants, aged 25-60 years and meeting the inclusion criteria were enrolled in the study after informed consent. They were divided into two groups, the hypothyroid group who were already on levothyroxine therapy and had a stable TSH in the normal range, and a euthyroid group who had no thyroid dysfunction. Both the groups were started on metformin therapy for the control of type-2 diabetes mellitus and followed for six months. Their blood samples for TSH and free thyroid hormone (fT4) were drawn both prior to and after the study period. Results Out of the 160 type-2 diabetic patients, TSH levels showed a significant reduction in the hypothyroid patients (2.33 ± 0.70, p < 0.001) with no significant changes in the euthyroid patients (3.87 ± 0.40, p = 0.206) following six months of metformin therapy. However, there was no significant difference in the fT4 levels in either of the groups. Conclusion Metformin has the effect of significantly lowering TSH levels in hypothyroid individuals. However, no such effect was observed in euthyroid patients.

ABSTRACT:The study was conducted to evaluate the potential cryoprotective effect of butylated hydroxytoluene (BHT) through post-thaw evaluation of canine semen and its optimal inclusion level. Ejaculated canine semen was extended in TRIS-glucose egg yolk extender containing various concentrations of BHT (0.5, 1.0, 1.5, 2.0, and 2.5mM). Semen was frozen at −196°C using 200 × 10 6 spermatozoa per 0.5 ml straws and post-thaw evaluation was carried out in terms of sperm motility, viability, plasma membrane integrity, and acrosomal integrity through phase-contrast microscope, supravital staining, hypo-osmotic swelling test, and normal acrosomal ridge, respectively. BHT was found to improve (P > 0.005) all post-thawed semen quality parameters at an inclusion level of 1.0mM in the extended semen. However, higher concentrations than this were found to have detrimental effects.
