The pooled nectar method of selfing F1 rust cultures introduces sampling errors because of limited numbers (a) of pycnia selected, (b) of aecia sampled, (c) of cultures derived per aecium. The reciprocal method of selfing is more reliable. An improved method of calculating genetic ratios from the pooled nectar method of selfing is comparable to backcrossing and offers marked advantages over present methods.
