Across the seed-seedling transition, several germination criteria are used in studies of palm-seed germination. In Oenocarpus bataua, these criteria have differential tolerance to thermal stress. In this study, we evaluated the tolerance of germination criteria to seed desiccation of the congeneric Oenocarpus bacaba. We dried seeds to different moisture contents (MC) before scoring first cataphyll, second cataphyll, enclosed eophyll and expanded eophyll. Seeds without drying had 41.7% MC. Germination success reached close to 70% after 75 and 105 days, depending on the germination criterion. Safe MC was close to initial MC and all seeds were dead with MC < 26.7%. As the primordial organs of the cataphylls and the eophyll are already detectable in the palm-seed embryo, all were affected by drying. Critical MC, defined here as 50% loss of germination capacity, increased from 35.4% (first cataphyll) to 37.1% (expanded eophyll) and confirmed that, across the seed-seedling transition, more advanced germination stages had a higher sensitivity to desiccation. During germination and development, the criteria appear in sequence over several weeks. Consequently, the desiccation damage was only detectable when the last criterion was evaluated. To avoid an underestimation of damages, we suggest that seed-stress studies in palms should take into account an adequate period for seedling development, which, for O. bacaba, was 105 days until the expansion of the eophyll.
A germinação pós fogo tem sido relatada a partir de uma enorme diversidade de espécies vegetais, uma vez que o calor e a fumaça podem quebrar a dormência de sementes enterradas no solo. Apesar dos muitos estudos em regiões temperadas, a contribuição do fogo na germinação de espécies tropicais ainda é pouco compreendida. Além da complexidade da composição química da fumaça, as condições de estresse durante a germinação podem levar a diversas alterações metabólicas, desencadeando a produção excessiva de espécies reativas de oxigênio (EROs). O objetivo dessa pesquisa foi avaliar a germinação e as alterações enzimáticas que ocorreram em sementes de Senna macranthera (Fabaceae), uma árvore nativa do Cerrado. Aplicou-se fogo em sementes enterradas a 2 e 5 cm no solo; defumação pela queima de feno; e fumaça líquida com uso de extratos pirolenhosos (EPs) produzidos do carvão de Eucalyptus sp. e Anadenanthera macrocarpa (angico) diluídos em diferentes concentrações. Utilizou-se sementes embebidas em água destilada, sem aplicação do fogo e fumaça, como testemunha. O teste de germinação durou uma semana e a protrusão da raiz foi o critério germinativo. Um corte foi realizado para verificar a viabilidade de sementes não germinadas. Foram calculadas a porcentagem de germinação (G%), tempo médio e índice de velocidade de germinação. A composição química dos EPs foi identificada por cromatografia gasosa. Foram analisadas as enzimas superóxido dismutase (SOD), catalase (CAT) e peroxidase (POX). A germinação da testemunha após sete dias foi de 0%, porém o teste de viabilidade atestou que estavam vivas (intumescimento do tegumento e cotilédones de coloração verde). Houve efeito positivo do fogo na germinação de sementes dormentes enterradas a 5cm embebidas em EP de eucalipto (14,0 G%), seguida das sementes incendiadas a 2cm (11,2 G%) e das que não foram embebidas em EP (10,0 G%). Sementes defumadas apresentaram intumescimento do tegumento, porém estavam mortas (0 G%). Registrou-se 124 compostos orgânicos nos EPs de eucalipto e angico. Piridonas e aldeídos podem ter sido responsáveis pela germinação das sementes embebidas com EP de eucalipto. A germinação das sementes embebidas com um dos EPs de angico, pode ter sido influenciada pelo (Tetrahydrofuran-2-yl) metanol, um derivado do éter. As atividades da SOD aumentaram após EP de eucalipto (F2-1:75) e exposição ao fogo. As atividades da CAT e POX reduziram após EP de angico (F2-1:50) e fumaça. O efeito do calor e da fumaça líquida na quebra de dormência de sementes sugere que S. macranthera é adaptada ao fogo. No entanto, a exposição das sementes à fumaça gasosa reduziu todas as atividades enzimáticas antioxidantes, indicando que este estresse é o mais prejudicial para a germinação, e em cenários de incêndios florestais frequentes seu desenvolvimento ficará prejudicado. Este trabalho indica como prática silvicultural o uso de EP de eucalipto e de angico no tratamento de sementes de S. macranthera, devido sua composição antioxidante. É possível que a POX atue na parede celular das sementes reduzindo peróxido, sinalizando dano oxidativo e removendo EROs. Os estudos nessas áreas são encorajados. Palavras-chave: Germinação pós fogo.
