Using gas chromatography mass spectrometry and the PacBio single molecule with real-time sequencing technology (SMRT), we analyzed the detailed metabolomic profiles and microbial community dynamics involved in ensiled Medicago sativa (alfalfa) inoculated without or with the homofermenter Lactobacillus plantarum or heterofermenter Lactobacillus buchneri. Our results revealed that 280 substances and 102 different metabolites were present in ensiled alfalfa. Inoculation of L. buchneri led to remarkable up-accumulation in concentrations of 4-aminobutyric acid, some free amino acids, and polyols in ensiled alfalfa, whereas considerable down-accumulation in cadaverine and succinic acid were observed in L. plantarum-inoculated silages. Completely different microbial flora and their successions during ensiling were observed in the control and two types of inoculant-treated silages. Inoculation of the L. plantarum or L. buchneri alters the microbial composition dynamics of the ensiled forage in very different manners. Our study demonstrates that metabolomic profiling analysis provides a deep insight in metabolites in silage. Moreover, the PacBio SMRT method revealed the microbial composition and its succession during the ensiling process at the species level. This provides information regarding the microbial processes underlying silage formation and may contribute to target-based regulation methods to achieve high-quality silage production.
Dietary selection and intake affect the survival and health of mammals under extreme environmental conditions. It has been suggested that dietary composition is a key driver of gut microbiota variation; however, how gut microbiota respond to seasonal dietary changes under extreme natural conditions remains poorly understood. Sequencing plant trnL (UAA) region and 16S rRNA gene analysis were employed to determine dietary composition and gut microbiota in freely grazing yaks on the Tibetan plateau. Dietary composition was more diverse in winter than in summer, while Gramineae and Rosaceae were consumed frequently all year. Turnover of seasonal diet and gut microbiota composition occurred consistently. Yaks shifted enterotypes in response to dietary change between warm and cold seasons to best utilize nitrogen and energy, in particular in the harsh cold season. Our findings provide insights into understanding seasonal changes of diet–microbiota linkages in the adaptation of mammals to high altitudes.
Yaks are a multifunctional and dominant livestock species on the Qinghai-Tibetan plateau. The yak grazing system is a traditional transhumance system. Research is focussed on both understanding and improving the system. Development of local economies and the use of new knowledge from yak research and modern technology are improving the system. However, the Qinghai-Tibetan plateau ecosystem is becoming dysfunctional through adverse climate sequences, over-population and over-grazing. The Chinese Government has implemented regional projects to address these degradation problems. Several policies have recently been introduced to improve yak grazing management. This paper reviews current knowledge on the system and the options for improving and optimising the system.
Rangeland management can affect plant diversity and plant functional groups of native grassland communities. To improve pasture for livestock grazing from the existing poisonous and ruderal vegetation in the 'black soil land' grassland in the headwaters of the Yangtze and the Yellow Rivers (HAYYR) of the Qinghai-Tibetan Plateau, three treatments (fencing-FT, artificial seeding with local grasses-ST and abandonment-AT) were applied during a 5-year period (2000)(2001)(2002)(2003)(2004)(2005). Plant diversity and groups (sedges, grasses and dwarf shrubs) in the treated plots were compared with non-treated control plots. Results showed that (1) FT promoted an increase in forbs species, but there was no change in the other plant functional groups; (2) with ST, species richness of forbs decreased but grass species increased indicating that sowing local grasses could change plant composition of black soil land in the short-term, and increase the proportion of grasses and, thus, forage for grazing and (3) AT, after plowing, was similar to CK plots, with forbs being the dominant plant functional group for the 5 years. In conclusion, the goal to alter black soil land cover from poisonous and ruderal plants to more useful plants for livestock grazing by either fencing, artificial seeding or abandonment was not successful in the long term.
Using light and scanning electron microscopy, the morphological adaptations of the yak (Bos grunniens) tongue to its foraging environment in the Qinghai-Tibetan Plateau were studied. The tongue of the yak was compared with that of cattle (Bos taurus). Compared with cattle, yak tongues are on average 4 cm shorter (P < 0.001), and yak consume forages using the labia oris, rather than by extending the tongue into the harsh environment. The lingual prominence of yak is greater (P < 0.001) and more developed than in cattle. The conical papillae on the prominence surface of yak are slightly larger (diameter: P = 0.068 and height: P = 0.761) and more numerous (P < 0.001) than in cattle. The lenticular papillae on the prominence surface of yak are larger (diameter: P = 0.002 and height: P = 0.115) and more numerous (P = 0.007) than in cattle. Such characteristics may improve the digestibility of forage by the grinding of food between the tongue and the upper palate. Filiform, conical, lenticular, fungiform, and vallate papillae were observed on the dorsal surface of the tongues studied; no foliate papillae were observed. The papillae were covered by keratinized epithelium, which was thicker (P < 0.001) in the yak than in cattle. It is suggested that the development of characteristic filiform papillae, and more numerous lingual gland ducts and mucus-secreting pores in the lenticular, fungiform and vallate papillae, fungiform papillae, probably having mechanical functions, are all morphological adaptations by yak to diets with greater fiber and DM content as provided by the plants within the Qinghai-Tibetan Plateau environment. On average, yak has 26 vallate papillae and cattle have 28. In the vallate papillae of the yak, the taste buds are arranged in a monolayer within the epithelium, whereas they are multilayered (2 to 4) in those papillae in cattle. The number of taste buds in each vallate papillae was less (P < 0.001) in the yak than in cattle. Therefore, the gustatory function of the yak was weaker than in cattle. Yaks graze throughout the year on diverse natural grasslands and have evolved morphological characteristics enabling them to consume a wide variety of plant species, thereby better adapting them to the typically harsh characteristics of their pastures.
Tibetan nomads have lived since ancient times in the unique and harsh environment of the Qinghai-Tibetan Plateau with average altitudes over 4000 m. These people have been able to live and multiply healthily over numerous generations under the extreme stress of high-altitude environment, including cold, hypoxia, and strong ultraviolet radiation, and with a simple diet devoid of vegetables and fruits for most of the year. Their survival depends heavily on yak milk, and its products comprise the main portion of their daily diet. In this review, yak milk and its derived products are examined in detail and compared with milk from other ruminant species. Yak milk products seem to be particularly rich in functional and bioactive components, which may play a role in maintaining the health status of Tibetan nomads. This includes particular profiles of amino acids and fatty acids, and high levels of antioxidant vitamins, specific enzymes, and bacteria with probiotic activity (yoghurt is the main food). Based on that, it is proposed that the Tibetan nomads have developed a nutritional mechanism adapted to cope with the specific challenges posed by the environment of the world's highest plateau. Systematic studies are required to demonstrate this in a more mechanistic way.
We studied the effects on alfalfa preservation and chemical composition of the addition of different levels of malic acid and citric acid at ensiling as well as the utilization efficiency of these 2 organic acids after fermentation. Alfalfa was harvested at early bloom stage. After wilting to a dry matter content of approximately 40%, the alfalfa was chopped into 1- to 2-cm pieces for ensiling. Four levels (0, 0.1, 0.5, and 1% of fresh weight) of malic acid or citric acid were applied to chopped alfalfa at ensiling with 4 replicates for each treatment, and the treated alfalfa forages were ensiled for 60 d in vacuum-sealed polyethylene bags (dimensions: 200 mm × 300 mm) packed with 200 to 230 g of fresh alfalfa per mini silo and an initial density of 0.534 g/cm. The application of malic or citric acids at ensiling for 60 d led to lower silage pH than was observed in the control silage (0% of malic or citric acids). Application of the 2 organic acids led to higher lactic acid concentration in alfalfa silage than in the control silage except with the application rate of 1% of fresh weight. Silages treated with both organic acids had lower nonprotein nitrogen concentrations than the control silages, and the nonprotein nitrogen concentrations in ensiled forages decreased with the increase in malic or citric acid application rates. The application of the 2 organic acid additives led to lower saturated fatty acid proportions and higher polyunsaturated fatty acid proportions in ensiled alfalfa than in the control silage. The amount of malic and citric acids degraded during ensiling of alfalfa was 1.45 and 0.63 g, respectively. At the application rate of 0.5% of fresh weight, residues of malic acid and citric acid in alfalfa silage were 11.1 and 13.6 g/kg of dry matter. These results indicate that including malic or citric acids at the ensiling of alfalfa effectively improved silage fermentation quality, limited proteolysis, improved fatty acid composition of the ensiled forage, and could provide animals with additional feed additives proven to promote animal performance. However, when the application rate of both organic acids reached 1%, the concentration of lactic acid in silages decreased notably. Additionally, 0.5 and 1% application rates also increased the yeast count in ensiled alfalfa.
Abstract. The Tibetan plateau is the source of most of the major rivers of Asia and has a huge impact on the livelihoods of the population, who have for centuries engaged in traditional herding practices. Sustainable management of the plateau is of critical importance not only for maintaining livelihoods but also because of its vital ecological function. The major problem of sustainable development in these grassland-livestock systems is the conflict between forage and livestock production. Despite considerable investment of manpower, material resources and capital over many years, attempts to resolve the problem have not been successful. The magnitude of conflict between forage and livestock is addressed by presenting 19 resolution strategies based on numerous research data. Each of these strategies is evaluated in terms of how it can be implemented, its potential benefits for livestock production, current progress and the requirement for further research. The 19 strategies have been divided into four topic categories, namely; grassland-forage, livestock, economy and market, societyculture, which cover the basic elements of sustainable development in this pastoral region. It is argued that improved planning and implementation of the proposed strategies must be based on the background investigation of natural and social status of the pastoral region. Particular attention needs to be given to genetic resources and technology in order to ensure the successful implementation of these proposals. In addition to expanding the use of 18 currently practiced strategies, the authors propose a further novel strategy of replacing the current intensive form of ecological migration with a model incorporating two semipermanent settlements. The objective of this novel strategy is to retain the nomadic element of pastoral husbandry while increasing the temporal and spatial scale of rotational stocking to reduce pasture degradation on the Tibetan plateau. It is argued that these proposals should be urgently incorporated into two national plans for the pastoral livestock industry, and the construction of an ecologically safe shelter zone on the Tibetan plateau, while ensuring a sustainable livelihood of its pastoral residents. Ensuring a successful implementation of these strategies in resolving the conflict between grassland and livestock, and promoting sustainable development on the Tibetan plateau, requires willing support at the level of national investment and policy commitment and from the herdsmen.
