RESUMOIntrodução: O diabetes melito (DM) constitui problema de saúde pública no Brasil. Este estudo teve por objetivo investigar a prevalência de DM, sua relação com características sociodemográficas e outros fatores de risco cardiovascular em uma população adulta do sertão de Pernambuco/Brasil. Sujeitos e métodos: Foi conduzido em 2008/2009 estudo transversal na população adulta e urbana do Distrito de Canaã, município de Triunfo, sertão de Pernambuco/ Brasil. Amostra representativa constituída por 198 indivíduos com idade média de 57,7 anos foi aleatoriamente selecionada, em que 80,0% tinham apenas o ensino fundamental, e 81,3% tinham renda mensal inferior a um salário-mínimo. Resultados: Foi encontrada prevalência de 13,6% de DM e de 7,6% de disglicemia (6,6% de glicemia de jejum alterada e 1,0% de tolerân-cia diminuída à glicose). Dos diabéticos, 24,0% não tinham diagnóstico. Nos diabéticos, 87,5% apresentavam síndrome metabólica (SM) e 68,0% eram hipertensos. Houve associação positiva e significativa entre DM e IMC e entre DM e SM. Conclusão: Foi encontrada elevada prevalên-cia de DM nessa população adulta urbana do sertão nordestino. Esses resultados assim como a sua associação com obesidade e síndrome metabólica indicam a necessidade de ações mais efetivas na prevenção dessas enfermidades. The objective of this study was to investigate the prevalence of DM and its relation with socio-demographic features and other cardiovascular risk factors in a adult population from the wilderness of Pernambuco, Brazil. Subjects and methods: In 2008/2009 a cross-sectional study in the adult urban population of Canãa district, city of Triunfo, in the wilderness of Pernambuco/Brazil was conducted. A representative sample of 198 individuals, with average age of 57.7 years, 80% with primary education and 81.3% with a monthly income of less than 1 minimum wage was randomly selected. Results: A prevalence of diabetes of 13.6% and 7.6% of disglycemia (6.6% of impaired fasting glycemia, and 1.0% of impaired glucose tolerance) was found. Among those with diabetes, 24% had no prior diagnosis. Among diabetics, metabolic syndrome (MS) and arterial hypertension (AH) were diagnosed in 87.5% and 68%, respectively. Besides these, there was a positive and significant association between DM and BMI, and between DM and MS. Conclusion: These data demonstrate a high prevalence of DM in the adult urban population from the wilderness of Pernambuco/Brazil. These results as well as their association with obesity and MS, indicate that more effective interventions are necessary for the prevention of these diseases also outside principal urban centers. The allelic distribution of human leukocyte antigen (HLA) class I genes (HLA-A, HLA-B, and HLA-Cw) of the population from the State of Pernambuco in Northeastern Brazil was studied in a sample of 101 healthy unrelated individuals. Low to medium resolution HLA class I typing was performed using polymerase chain reaction-amplified DNA hybridized to sequence specific primers (PCR-SSPs). Twenty allele groups were detected for HLA-A, 28 for HLA-B, and 14 for HLA-Cw. The most frequent alleles were HLA-A*02(0.2871), HLA-B*15(0.1238), and HLA-Cw*04(0.2277), and the most frequent genotypes were A*02/A*02(0.0990), B*15/B*15(0.0594), and Cw*04/Cw*04 and Cw*07/Cw*07, both with a frequency of 0.0792. The observed heterozygosity for the studied loci was 79.21% for HLA-A, 87.13% for HLA-B, and 77.23% for HLA-Cw. The most frequent haplotype was A*02-Cw*04-B*35(0.0485), which is also present in Western European, Amerindian, and Brazilian Mulatto populations, but absent in African populations. Taken together, these data corroborate the historic anthropological evidences of the origin of the Northeastern Brazilian population from Pernambuco.

The sequences of the two hypervariable (HV) segments of the mitochondrial DNA (mtDNA) control region were determined in 167 randomly selected, unrelated individuals living in the state of Alagoas, north-eastern Brazil. One hundred and forty-five different haplotypes, associated with 139 variable positions, were determined. More than 95% of the mtDNA sequences could be allocated to specific mtDNA haplogroups according to the mutational motifs. Length heteroplasmy in the C-stretch HV1 and HV2 regions was observed in 22 and 11%, respectively, of the population sample. The genetic diversity was estimated to be 0.9975 and the probability of two random individuals presenting identical mtDNA haplotypes was 0.0084. The most frequent haplotype was shared by six individuals. All sequences showed high-quality values and phantom mutations were not detected. The diversity revealed in the mitochondrial control region indicates the importance of this locus for forensic casework and population studies within Alagoas, Brazil.

Samples of venous blood were collected using the anticoagulant ACD (citric acid 0.038 M; tribasic sodium citrate 0.075 M; dextrose 0.033 M), from 247 non-related, healthy individuals from the population of the State of Rio Grande do Norte, in Northeastern Brazil. The DNA was extracted by rapid mini salting-out and digestion with proteinase K (1). The PCR was performed in a Perkin-Elmer 2400 thermal cycler, using amplification protocols described in Promega Manual (Madison, WI). The amplified products underwent electrophoresis in a denaturing polyacrylamide gel in a manual sequencer from Amershan Pharmacia (SQ3 Sequencer). The statistical analysis was performed using the Genetic Data Analysis (2) and the PowerStat (3) softwares. The complete dataset is available in www.nlink.com.br/mauricio.
