The effect of farnesol, a sesquiterpene alcohol, on the production of laccases by Trametes versicolor and Pycnoporus sanguineus in pineapple waste solid-state fermentation was evaluated. Extracellular laccase production reached a maximum of 77.88 ± 5.62 U/g (236% above control) in farnesol-induced cultures of T. versicolor on the 17th day, whereas in a similar P. sanguineus culture, a maximal laccase activity of 130.95 ± 2.20 U/g (159% increase) was obtained on the 17th day. A single 45 KDa laccase was produced by both fungi under the influence of farnesol. These and other data allow us to conclude that farnesol acted as an inducer of the same form of laccase in both fungi. Farnesol disfavored fungal growth by increasing the lag phase, but it also clearly improved the oxidative state of the cultures. Contrary to the results obtained previously in submerged cultures, farnesol did not promote hyperbranching in the fungal mycelia. This is the first demonstration that farnesol is an excellent inducer of laccases in T. versicolor and P. sanguineus in solid-state cultivation. In quantitative terms, the results can be regarded as an excellent starting point for developing industrial or at least pre-industrial procedures to produce laccases using T. versicolor and P sanguineus under the stimulus of farnesol.
Diante do aumento da população mundial, consumo em excesso e consequente poluição ambiental, a busca por alternativas eficientes que contribuam para a descontaminação das águas é de grande relevância. Os efluentes industriais apresentam composição física, química e biológica complexa, sendo que o tipo de matéria prima usada pela indústria definirá sua aparência, coloração, densidade, acidez e basicidade, temperatura e presença de compostos orgânicos e/ou inorgânicos. O descarte incorreto desse passivo ambiental pode causar danos à saúde e prejuízos ao ecossistema. Nesse contexto, a utilização da tecnologia de Wetlands Construídas (WC) para o tratamento de águas residuais industriais tem se tornado uma perspectiva crescente, visto suas inúmeras vantagens face aos tratamentos convencionais, como baixo custo, simplicidade na implantação e alta eficiência na remoção de poluentes emergentes. WC é um mecanismo, composto por lagoas ou canais artificiais rasos, que abrigam plantas aquáticas que possuem um leito filtrante, onde essas espécies vegetais promovem a atenuação de diversos contaminantes presentes no efluente. Assim, o objetivo deste trabalho é realizar uma revisão bibliográfica, preferencialmente no período de (2011/2021), sobre o uso de WC na fitorremediação de efluentes industriais, que ainda é uma alternativa pouco difundida no contexto nacional. Nesta revisão, diversas modalidades de WC que apresentam alto grau de eficiência para remoção de compostos poluentes dos efluentes industriais foram encontradas. Para além do mais, pode-se concluir que a aplicação dessa tecnologia representa a possibilidade de expandir o tratamento descentralizado de efluentes, alcançando diversas localidades de países em desenvolvimento, e também reduzir as emissões de gases de efeito estufa, gerados pelos tratamentos anaeróbios.
RESUMOFungos endofíticos são considerados um nicho em potencial na produção de enzimas de interesse biotecnológico. Entre elas destaca-se a asparaginase, utilizada no tratamento de leucemia aguda. Atualmente a enzima produzida em escala industrial e disponível comercialmente é de origem bacteriana, entretanto, está associada à ocorrência de reações adversas, além de sua produção ser intracelular, tornando o processo de extração e purificação difícil e oneroso. Muitos fatores físico químicos relacionados ao processo fermentativo, podem ser utilizados para otimizar a produção enzimática por microrganismos, tais como diferentes fontes nutricionais, pH, temperatura, tempo de cultivo dentre outros. Neste contexto, o estudo teve como objetivo avaliar a influência de diferentes tempos de cultivo e fontes de carbono na produção da enzima Lasparaginase por fungos endofíticos de Moringa oleifera. A influência do tempo de incubação foi avaliada cultivando os endófitos a 30 o C no meio específico por 48, 72, 96 e 120 horas. Para avaliar a produção da enzima em diferentes fontes de carbono, os fungos foram cultivados a 30 °C por 120 horas em glicose, glicerol e etanol. A atividade enzimática extracelular foi avaliada pela relação entre o diâmetro médio do halo de degradação e o diâmetro médio da colônia e expressa como índice enzimático. A glicose foi a fonte de carbono preferencial para a maioria dos fungos, assim como o cultivo por 120 horas favoreceu maior produção da enzima. As variações no índice enzimático nos diferentes parâmetros analisados (fonte de carbono e tempo de cultivo) mostram as individualidades fisiológicas de cada microrganismo, influenciando na produção de Lasparaginase pelos fungos, evidenciando que a produção enzimática é variável, estando relacionada com as diferentes condições de cultivo.
