The association of tamsulosin/tadalafil reduces detrusor pressure at maximum flow without changing the maximum flow rate during micturition and significantly improves lower urinary tract symptoms compared with the isolated use of tamsulosin.
O carcinossarcoma 256 de Walker tem despertado o interesse de muitos pesquisadores como modelo experimental para estudo da biologia tumoral. OBJETIVO: estabelecer um modelo de tumor renal que possa ser usado para estudar in vivo e in vitro, as alterações impostas pelas neoplasias. MÉTODOS: utilizados vinte ratos Wistar, machos, adultos, pesando entre 250-300 g, oriundos do Laboratório de Cirurgia Experimental da Universidade Federal do Ceará. Sob anestesia inalatória procedia-se uma pequena incisão supraumbilical, e com manobra delicada fazia-se a exposição do rim direito. Neste órgão eram inoculadas 3x10(5) células tumorais viáveis. Os animais então eram mantidos em gaiolas individuais com as mesmas condições ambientais e com água e dieta ad libitum. RESULTADOS: o Carcinossarcoma 256 de Walker, implantado no parênquima do rim direito de ratos Wistar apresentou índice de pega de 100%, e crescimento rápido, invadiu por contiguidade as estruturas vizinhas, porém sem apresentar metástases, no entanto, levando os animais a óbito no curso médio de 14 dias. CONCLUSÃO: o modelo de implante de tumor de Walker no parênquima do rim direito de ratos Wistar é eficiente, tem reprodutibilidade, apresentando um índice de pega de 100%, e permitindo seu uso em linhas de pesquisa.
Treatment of pelvic neoplasms with radiotherapy may develop sequelae, especially RHC. An 85-year-old male patient was admitted to a hospital emergency with gross hematuria leading to urinary retention and was diagnosed with RHC. The urinary bladder was probed, unobstructed, and maintained in continuous three-way saline irrigation. During 45 days of hospitalization, the patient underwent two cystoscopic procedures for urinary bladder flocculation, whole blood transfusions, and one platelet apheresis. None of these interventions led to clinical resolution. As the patient hematological condition was deteriorating, dexamethasone (4 mg i.v., bolus of 6/6, 12/12, and 24 h during five days) and epoetin alpha (1000 IU, 1 ml, s.c., for four weeks) were administered which led to the remission of the urinary bleeding. Dexamethasone therapy may be considered for RHC, when conventional treatments are not effective or are not possible, avoiding more aggressive interventions such as cystectomy.
Uroguanylin (UGN) is a peptide shown to stimulate membrane bound guanylate cyclase receptors (GC) which has been identified in human corpora cavernosa (HCC). This study tests whether UGN induces HCC relaxation and evaluate its interaction with soluble GC. Strips of HCC were preconstricted with phenylephrine (PHE) and concentration‐response curves (10−10 to 10−5 M) to human UGN were performed. In other experimental set tissues were treated with 10−7 M vardenafil 10 min before PHE challenge. UGN effects were evaluated after 10−4M L‐NAME or ODQ and also compared with atrial natriuretic peptide (ANP). UGN induced maximal relaxation of 55 ± 7.5 % (PD2 7.4) when used alone and 82.6 ± 4.5% (PD2 14.1) (p<0.05) after vardenafil pre‐treatment. L‐NAME or 7‐NI were unable to change the maximal response or PD2 values for UGN. The maximal relaxation after L‐NAME was 52.6 ± 5.8% and with 7‐NI was 46.1 ± 3.2%. ANP induced 52.6 ± 5.8 % maximal relaxation with PD2 of 4.0. The effects of eletrical field stimulation (40–50 V; 0.5 ms; 2 and 20 Hz) or ACh (10−7M) were not altered when tissues were incubated with UGN (10−7M). UGN induces HCC relaxation downstream to NO release from nerves or endothelium and with similar profile to ANP. This study shows for the first time that UGN, a GC‐C agonist, relaxes human corpus cavernosum and that vardenafil can increase this effect, and may point out to new therapeutic targets to treat erectile dysfunction.
