Introdução: o cultivo in vitro de células animais é uma tecnologia moderna, baseada em fundamentos e princípios científicos com inúmeras aplicações, levando inclusive, a união de organizações internacionais no sentido de validar modelos de ensaios in vitro para avaliar toxicidade de produtos químicos, tendo em vista que estes são de baixo custo, maior produtividade e fornece resultados mais rápidos. Objetivo: Abordar por meio de revisão bibliográfica os métodos de avaliação de citotoxicidade mais empregados, além de outros ensaios biológicos necessários para testes de biocompatibilidade. Metodologia: foram selecionados os principais trabalhos e normas por meio de busca nas bases de pesquisa de trabalhos científicos da Scielo, Google Acadêmico e livros sobre o tema, a fim de possibilitar um embasamento para um adequado delineamento de ensaios in vitro de citotocicidade. Resultados: A atual disponibilidade de uma variabilidade de metodologias e linhagens celulares em bancos nacionais e internacionais tornou possível a larga utilização da cultura de células como modelos in vitro para avaliações toxicológicas. Conclusão: testes iniciais de citotoxicidade apresentam limitações quando correlacionados com as situações clínicas, porém são validos para determinar o comportamento biológico dos materiais e/ou seus componentes, estabelecendo informações essenciais na fase inicial de investigação de um biomaterial e direciona os testes in vivo necessários. Além dos ensaios de citotoxicidade, que não devem ser equiparados a biocompatibilidade, a avaliação de um dispositivo médico deve ser baseada em testes in vivo que analisem a toxidade aguda e crônica, inflamação, irritação, sensibilidade, carcinogenicidade e mutagenicidade, reações alérgicas, hemocompatibilidade e toxidade sistêmica.
