RESUMO -A produção de enzimas está cada vez mais escassa devido aos seus altos custos. Diversos estudos estão sendo realizados de uma forma mais econômica, utilizando biomassa lignocelulósica (resíduos agroindustriais), como substrato em fermentação semissólida para a obtenção de celulases, ocasionando menores despesas, acarretando uma maior procura pelo processo. Este trabalho teve como objetivo realizar um acompanhamento cinético da produção de celulase, pelo fungo Trichoderma reesei LCB 48 em fermentação semissólida, usando como substrato sorgo sacarino, cultivar IPA P15, suplementado com farelo de trigo. O substrato foi caracterizado para a obtenção de suas propriedades físico-químicas e lignocelulósicas. O resultado da caracterização foi satisfatório, apresentando um índice de celulose de 40,55%, tornando-se um material com potencial para a produção de enzimas celulolíticas. A fermentação foi realizada em 216h e as amostras foram coletadas periodicamente para análise de pH, umidade, açúcares redutores e atividade enzimática expressa em unidade de enzima por grama de substrato (U/g). A melhor atividade foi de 1,82 U/g, obtida em 120h de fermentação, em um meio com 60% de umidade e pH 5,0.
