A poliamina espermidina é encontrada em todos os seres eucariotos e na grande parte dos procariontes. Conforme sua natureza policatiônica, as poliaminas podem juntar facilmente a biomoléculas com carga negativa, especialmente o DNA, RNA, proteínas e fosfolipídios, modificando em muitas ocasiões a função dessas macromoléculas. Dada a sua habilidade em se ligar em outras moléculas, eles estão associados em diversas funções, sobretudo vinculadas ao crescimento, autofagia, sobrevivência e proliferação celular. Objetivou-se com este estudo identificar os efeitos da poliamina espermidinano metabolismo e na longevidade. Trata-se de um estudo de revisão de literatura integrativa, foi utilizada as seguintes bases de dados SciELO, PUBMED, LILACS e Periódicos da Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (Capes). A espermidina exerce nas atividades de promoção da longevidade sendo ligada à sua capacidade de manter proteostase por meio da estimulação da autofagia citoprotetora, ocasionando uma retirada do acúmulo intracelular de detritos tóxicos produzidos pelo envelhecimento e doenças. Além da indução da autofagia, a atuação da espermidina tem sido referente à modulação de espécies reativas de oxigênio, replicação, transcrição e tradução de DNA, propriedades antiinflamatórias, atividade mitocondrial modificada, proteostase melhorada, crescimento da hipusinação. As poliaminas têm uma relação complexa e difusa com as doenças. Podendo ser prejudiciais, neutras ou benéficas, variando de acordo com a poliamina e a doença específica. Contudo, parece que a espermidina expressou efeitos mais positivos, estando em concordância com seus efeitos benéficos expressados acerca do tempo de vida e o estresse.
