The aim of the present study was to explore the protective effect of small interfering RNA (siRNA) against nuclear factor κB (NF-κB) p65 on sepsis-induced acute lung injury (ALI) in mice. In total, 70 male Kunming mice were randomly divided into a healthy control group, a sepsis group, a specific interfering group and a scrambled control group (Sc), and the latter three groups were divided into post-operational 6 and 12 h subgroups, each of which consisted of 10 mice. The mice were administered with NF-κB siRNA, scrambled siRNA and normal saline via tail vein injection. Following 1 h, a mouse model of septic ALI was produced by cecal ligation and puncture (CLP) in the two siRNA groups and the sepsis control group. At 6 and 12 h post-operation, the experimental mice were sacrificed and the lung tissue samples were collected. Histopathological changes, wet/dry ratio of lung weight, NF-κB protein and NF-κB p65 mRNA levels, matrix metalloproteinase-9 (MMP-9) mRNA and protein activity were detected. Compared with the sepsis group and the Sc at the corresponding time, the expression levels of NF-κB p65 mRNA, the lung injury of experimental mice, the wet/dry ratio and the levels of MMP-9 mRNA and protein activity decreased, and significant differences were observed at 6 h post-operation (P<0.05). RNA interference against NF-κB p65 was able to decrease the expression of NF-κB and further inhibit the early phasic excessive inflammatory reaction in sepsis, which may alleviate ALI.
Abstract. Previous studies have indicated that microRNAs (miRNAs/miRs) may participate in the pathogenesis of hypertension. miR-145 has been demonstrated to serve important roles in the development of numerous cardiovascular diseases. However, the specific role of miR-145 in hypertension remains unclear. The present study aimed to investigate the role of miR-145 in spontaneously hypertensive rats (SHR) and rat vascular endothelial cells (RVECs). The results of the present study demonstrated that in the SHR group miR-145 expression was significantly upregulated in the thoracic aorta compared with the control group. Furthermore, a significant decrease in nitric oxide (NO) content was observed in the SHR group compared with the control rats. In RVECs, silencing miR-145 induced a significant increase in the expression of solute carrier family 7 member 1 (SLC7A1) and phosphorylated endothelial nitric oxide synthase, and a dual-luciferase reporter assay confirmed that SLC7A1 is a direct target of miR-145. The results of the present study indicate that miR-145 functions as a key mediator in the pathogenesis of hypertension via targeting SLC7A1, which suggests that miR-145 is a potential target for the treatment of hypertension.
a Polycaprolactone (PCL) filled with microcrystalline cellulose (MCC), wood flour (WF), or both were characterized before and after exposure to various environmental conditions for 60 days. PCL/WF composites had the greatest tensile strength and modulus compared to neat PCL or PCL composites containing MCC. Electron microscopy indicated better adhesion between WF particles and PCL than between MCC particles and PCL. Neither wood flour nor MCC cellulose appeared to significantly affect the crystallinity of PCL. Environmental conditioning resulted in only minor deterioration of mechanical properties, although samples soaked in water had greater deterioration of mechanical properties than those in high humidity or freezing environments. After a modified 12-week soil block test, specimens made with wood flour lost weight and showed signs of decay after exposure to the brown-rot fungus Gloeophyllum trabeum.
The purpose of this study was to investigate the relationship among potential athletic donors' beliefs, attitudes, subjective norms, and perceived behavioural control on donor intentions toward green stadium initiatives (GSI). Research participants were university students (N = 186) who responded to a questionnaire designed to measure these related concepts. Hierarchical regression analyses revealed that attitude and subjective norm mediated the impacts of behavioural beliefs and normative beliefs on donor intentions toward GSI. These findings support the applicability of the theory of reasoned action (TRA) in nurturing donation behaviours in the collegiate athletic context. The discussion is focused on developing educational and promotional programmes to enhance beliefs, generate positive attitudes, and develop supportive norms. Implications for practitioners are further presented.
