The ability of surface acoustic waves to trap and manipulate micrometer-scale particles and biological cells has led to many applications involving "acoustic tweezers" in biology, chemistry, engineering, and medicine. Here, we present 3D acoustic tweezers, which use surface acoustic waves to create 3D trapping nodes for the capture and manipulation of microparticles and cells along three mutually orthogonal axes. In this method, we use standing-wave phase shifts to move particles or cells in-plane, whereas the amplitude of acoustic vibrations is used to control particle motion along an orthogonal plane. We demonstrate, through controlled experiments guided by simulations, how acoustic vibrations result in micromanipulations in a microfluidic chamber by invoking physical principles that underlie the formation and regulation of complex, volumetric trapping nodes of particles and biological cells. We further show how 3D acoustic tweezers can be used to pick up, translate, and print single cells and cell assemblies to create 2D and 3D structures in a precise, noninvasive, label-free, and contact-free manner.3D acoustic tweezers | cell printing | 3D cell manipulation | cell assembly | 3D particle manipulationT he ability to precisely manipulate living cells in three dimensions, one cell at a time, offers many possible applications in regenerative medicine, tissue engineering, neuroscience, and biophysics (1-3). However, current bioprinting methods are generally hampered by the need to reconstruct and mimic 3D cellto-cell communications and cell-environment interactions. Because of this constraint, bioprinting requires accurate reproduction of multicellular architecture (4, 5). Several approaches have been developed to produce complex cell patterns, clusters, assembled arrays, and even tissue structures. These approaches use many disparate technologies which include optics, magnetic and electrical fields, injection printing, physical or geometric constraints, or surface engineering (6-11). The ability to precisely maneuver micro/nano objects in fluids in a contactless, biocompatible manner can enable innovative technologies and may have far-reaching impact in fields such as biology, chemical engineering, and nanotechnology. Here, we report a design for acoustically powered bubble-based microswimmers that are capable of autonomous motion in three dimensions and selectively transporting individual synthetic colloids and mammalian cells in a crowded group without labeling, surface modification, or effect on nearby objects. In contrast to previously reported microswimmers, their motion does not require operation at acoustic pressure nodes, enabling propulsion at low power and far from an ultrasonic transducer. In a megahertz acoustic field, the microswimmers are subject to two predominant forces: the secondary Bjerknes force and a locally generated acoustic streaming propulsive force. The combination of these two forces enables the microswimmers to independently swim on three dimensional boundaries or in free space under magnetical steering.
Rheotaxis is a common phenomenon in nature that refers to the directed movement of micro-organisms as a result of shear flow. The ability to mimic natural rheotaxis using synthetic micro/nanomotors adds functionality to enable their applications in biomedicine and chemistry. Here, we present a hybrid strategy that can achieve both positive and negative rheotaxis of synthetic bimetallic micromotors by employing a combination of chemical fuel and acoustic force. An acoustofluidic device is developed for the integration of the two propulsion mechanisms. Using acoustic force alone, bimetallic microrods are propelled along the bottom surface in the center of a fluid channel. The leading end of the microrod is always the less dense end, as established in earlier experiments. With chemical fuel (HO) alone, the microrods orient themselves with their anode end against the flow when shear flow is present. Numerical simulations confirm that this orientation results from tilting of the microrods relative to the bottom surface of the channel, which is caused by catalytically driven electro-osmotic flow. By combining this catalytic orientation effect with more powerful, density-dependent acoustic propulsion, both positive and negative rheotaxis can be achieved. The ability to respond to flow stimuli and collectively propel synthetic microswimmers in a directed manner indicates an important step toward practical applications.
The multicellular spheroid is an important 3D cell culture model for drug screening, tissue engineering, and fundamental biological research. Although several spheroid formation methods have been reported, the field still lacks high-throughput and simple fabrication methods to accelerate its adoption in drug development industry. Surface acoustic wave (SAW) based cell manipulation methods, which are known to be non-invasive, flexible, and high-throughput, have not been successfully developed for fabricating 3D cell assemblies or spheroids, due to the limited understanding on SAW-based vertical levitation. In this work, we demonstrated the capability of fabricating multicellular spheroids in the 3D acoustic tweezers platform. Our method used drag force from microstreaming to levitate cells in the vertical direction, and used radiation force from Gor’kov potential to aggregate cells in the horizontal plane. After optimizing the device geometry and input power, we demonstrated the rapid and high-throughput nature of our method by continuously fabricating more than 150 size-controllable spheroids and transferring them to Petri dishes every 30 minutes. The spheroids fabricated by our 3D acoustic tweezers can be cultured for a week with good cell viability. We further demonstrated that spheroids fabricated by this method could be used for drug testing. Unlike the 2D monolayer model, HepG2 spheroids fabricated by the 3D acoustic tweezers manifested distinct drug resistance, which matched existing reports. The 3D acoustic tweezers based method can serve as a novel bio-manufacturing tool to fabricate complex 3D cell assembles for biological research, tissue engineering, and drug development.
For decades, scientists have pursued the goal of performing automated reactions in a compact fluid processor with minimal human intervention. Most advanced fluidic handling technologies (e.g., microfluidic chips and micro-well plates) lack fluid rewritability, and the associated benefits of multi-path routing and re-programmability, due to surface-adsorption-induced contamination on contacting structures. This limits their processing speed and the complexity of reaction test matrices. We present a contactless droplet transport and processing technique called digital acoustofluidics which dynamically manipulates droplets with volumes from 1 nL to 100 µL along any planar axis via acoustic-streaming-induced hydrodynamic traps, all in a contamination-free (lower than 10−10% diffusion into the fluorinated carrier oil layer) and biocompatible (99.2% cell viability) manner. Hence, digital acoustofluidics can execute reactions on overlapping, non-contaminated, fluidic paths and can scale to perform massive interaction matrices within a single device.
Platelets contain growth factors which are important in biomedical and clinical applications. In this work, we present an acoustic separation device for high-throughput, non-invasive platelet isolation. In particular, we separated platelets from whole blood at a 10 mL/min throughput, which is three orders of magnitude greater than that of existing acoustic-based platelet separation techniques. Without sample dilution, we observed more than 80% RBC/WBC removal & platelet recovery. High throughput, high separation efficiency, and biocompatibility make this device useful for many clinical applications.
Engines and motors are everywhere in the modern world, but it is a challenge to make them work if they are very small. On the micron length scale, inertial forces are weak and conventional motor designs involving, e.g., pistons, jets, or flywheels cease to function. Biological motors work by a different principle, using catalysis to convert chemical to mechanical energy on the nanometer length scale. To do this, they must apply force continuously against their viscous surroundings, and because of their small size, their movement is "jittery" because of the random shoves and turns they experience from molecules in their surroundings. The first synthetic catalytic motors, discovered about 15 years ago, were bimetallic Pt-Au microrods that swim in fluids through self-electrophoresis, a mechanism that is apparently not used by biological catalytic nanomotors. Despite the difference in propulsion mechanisms, catalytic microswimmers are subject to the same external forces as natural swimmers such as bacteria. Therefore, they follow similar scaling laws, are subject to Brownian forces, and exhibit a rich array of biomimetic emergent behavior (e.g., chemotaxis, rheotaxis, schooling, and predator-prey behavior). It was later discovered, quite by accident, that the same metallic microrods undergo rapid autonomous movement in acoustic fields, converting excitation energy in the frequency (MHz) and power range (up to several W/cm) that is commonly used for ultrasonic imaging into axial movement. Because the acoustic propulsion mechanism is fuel-free, it can operate in media that have been inaccessible to chemically powered motors, such as the interior of living cells. The power levels used are intermediate between those of ultrasonic diagnostic imaging and therapy, so the translation of basic research on microswimmers into biomedical applications, including in vivo diagnostics and drug delivery, is possible. Acoustic and chemical propulsion are applied independently to microswimmers, so by modulating the acoustic power one can achieve microswimmer functionalities that are not accessible with the individual propulsion mechanisms. These include motion of particles forward and backward with switching between chemical and acoustic propulsion, the assembly/disassembly equilibrium of particle swarms and colloidal molecules, and controllable upstream or downstream propulsion in a flowing fluid. This Account relates our current understanding of the chemical and acoustic propulsion mechanisms, and describes how their combination can be particularly powerful for imparting enhanced functionality to micromotors.
Acoustic-based fluorescence activated cell sorters (FACS) have drawn increased attention in recent years due to their versatility, high biocompatibility, high controllability, and simple design. However, the sorting throughput for existing acoustic cell sorters is far from optimum for practical applications. Here we report a high-throughput cell sorting method based on standing surface acoustic waves (SSAWs). We utilized a pair of focused interdigital transducers (FIDTs) to generate SSAW with high resolution and high energy efficiency. As a result, the sorting throughput is improved significantly from conventional acoustic-based cell sorting methods. We demonstrated the successful sorting of 10 μm polystyrene particles with a minimum actuation time of 72 μs, which translates to a potential sorting rate of more than 13,800 events/s. Without using a cell-detection unit, we were able to demonstrate an actual sorting throughput of 3,300 events/s. Our sorting method can be conveniently integrated with upstream detection units, and it represents an important development towards a functional acoustic-based FACS system.
