BACKGROUND: Uncaria gambir (local name: gambir) is a plant native to Sumatera, Malaya and Borneo. This plant is potential as local wisdom for therapeutics. In Sumatera, gambir was used as a traditional treatment for fever, diarrhoea, diabetics and wound healing. AIM: To explore the efficacy of gambir extract on TNF alpha level, prostaglandin E2 level, lesson area, body weight, lipid profile and leptin level in Wistar rat-model gastritis. METHODS: This study was an experimental study, with a pre-post-test control group design. The subjects in this study were 30 male rats, 8 weeks old, weight 150-200 gram. Rats were administered with gambir extract at the dose of 20, 40 and 80 mg/kg BW/day for 3 days. Gambir was extracted by maceration methods. Statistical analysis was performed by SPSS 18. RESULTS: Gambir extract at the dose of 80 mg/kg BW exhibited the highest efficacy in reducing TNF alpha level, lesion area and increasing prostaglandin E2 level compared to gambir extract at doses of 20 mg/kg BW, 400 mg/kg BW, negative control, and positive control. CONCLUSION: Gambir extract was effective in reducing TNF alpha level, lesson area, and increasing prostaglandin E2 level in Wistar rat-model gastritis.
Diabetes mellitus is a disease or metabolic disorder that is increasing year by year, characterized by high blood sugar levels as a result of insulin function insufficiency. Diabetes mellitus, which is often referred to as diabetes, is a disease or metabolic disorder that is increasing year by year, characterized by high blood sugar levels as a result of insulin function insufficiency. Treatment is by using drugs. Currently, a treatment using herbal medicine is being developed, one of which is Kayu kuning, where in previous studies it was found that the roots of Kayu kuning can reduce blood glucose levels. This has triggered the Biochemistry and Medical Chemistry section of the Faculty of Medicine to introduce to the public the preparations of this herbal medicine with ready-to-use packaging and the right dosage. The process of making tea is carried out at the Chemical Basic Medicine Laboratory, Faculty of Medicine, Sriwijaya University. About 5.6 kg of yellow wood root, washed and cut into small pieces then dried and ground to obtain a coarse powder (simplicia) of about 2.5 kg. Yellow wood root simplicia is put into tea bags weighing 10 g per bag. One tea bag can be brewed with 200 mL of hot water and drunk once a day. In this service, 250 tea bags were produced which were packaged in 50 packages (5 tea bags/package). Penelitian ini menggunakan ekstrak akar kayu kuning (Arcangelisia flava) untuk melihat aktifitas sebagai antidiabetes yang dapat menurunkan kadar glukosa darah tikus putih jantan (Rattus novergicus) galur wistar yang diinduksi aloksan. Rancangan penelitian ialah studi eksperimental secara in vivo dengan pendekatan post test with control group design. Penelitian ini menggunakan 70 ekor tikus dan dibagi secara acak menjadi 14 kelompok dengan jumlah tikus lima ekor perkelompok yaitu kontrol negatif (NaCMC 0,5%), kontrol positif (acarbose 4,5 mg/kgBB), ekstrak heksana akar kayu kuning(dosis 250 mg/kgBB, 500 mg/kgBB dan 750 mg/kgBB), ekstrak etil asetat akar kayu kuning (dosis 250 mg/kgBB, 500 mg/kgBB dan 750 mg/kgBB), ekstrak etanol akar kayu kuning (dosis 250 mg/kgBB, 500 mg/kgBB dan 750 mg/kgBB), dan ekstrak aquades akar kayu kuning (dosis 250 mg/kgBB, 500 mg/kgBB dan 750 mg/kgBB). Seluruh kelompok diinduksi dengan aloksan 160 mg/kgBB sehingga mempunyai glukosa darah sewaktu ≥200 mg/dL (pretest). Setelah 15 hari diberi perlakuan, dilakukan pengukuran glukosa darahnya (posttest). Hasil penelitian menunjukkan bahwa ekstrak etanol dosis 750 mg/kgBB, ekstrak etil asetat (dosis 250 mg/kgBB, 500 mg/kgBB dan 750 mg/kgBB) dan ekstrak aquadest (dosis 250 mg/kgBB, 500 mg/kgBB dan 750 mg/kgBB) memiliki kemampuan menurunkan kadar glukosa darah dan hanya etil asetat dosis 750 mg/kgBB, aquadest dosis 500 mg/kgBB dan 750 mg/kgBB yang mampu menurunkan kadar glukosa darah hampir menyerupai kontrol positif (acarbose). Dari hasil uji fitokimia didapatkan adanya alkaloid dan flavonoid di dalam ekstrak etanol, etil asetat dan aquades, dan tannin hanya terdapat dalam ekstrak etanol dan etil asetat.
