Siput laut gonggong asal Bintan merupakan salah satu gastropoda laut yang belum dimanfaatkan secara optimal. Siput ini merupakan makanan laut khas Bintan dan harganya sangat mahal. Secara empiris, siput ini dipercaya dapat meningkatkan stamina dan vitalitas. Tujuan penelitian ini adalah untuk melakukan penapisan senyawa bioaktif pada siput laut gonggong asal Bintan yaitu aktivitas antioksidan dan aktivitas antimikroba. Aktivitas antioksidan dianalisis menggunakan metode DPPH dan aktivitas antimikroba dianalisis menggunakan metode difusi sumur. Aktivitas antioksidan ekstrak gonggong bercangkang tipis dengan IC50 1.433,08±0,01 ppm lebih tinggi daripada gonggong bercangkang tebal dengan IC50= 2.051,55±0,10 ppm. Aktivitas antimikroba pada ekstrak gonggong lebih baik pada bakteri Gram positif daripada bakteri Gram negatif. Ekstrak gonggong rebus bercangkang tebal memiliki aktivitas antimikroba yang paling tinggi dengan nilai rata-rata diameter daya hambat (DDH) yaitu 25,53±0,12 mm.
ABSTRAKGonggong termasuk sejenis siput laut, biota endemik yang banyak hidup di pantai Pulau Bintan dan sekitarnya di Provinsi Kepulauan Riau. Gonggong merupakan ikon kota Tanjungpinang, Provinsi Kepulauan Riau. Sampai saat ini, penelitian gonggong masih sangat sedikit padahal siput ini merupakan spesies yang sangat potensial. Tujuan penelitian ini adalah untuk mengkarakterisasi protein histon dari siput gonggong Strombus sp. asal Bintan sebagai kandidat pangan fungsional. Karakterisasi profil protein menggunakan SDS-PAGE. Kadar protein pada gonggong dianalisis dengan menggunakan metode Bradford. Gonggong rebus bercangkang tebal diekstraksi dengan metode maserasi menggunakan pelarut etanol PA 95% dan uji aktivitas antimikroba menggunakan metode sumur. Asam amino dianalisis menggunakan HPLC. Hasil karakterisasi profil protein pada daging gonggong menunjukkan bahwa gonggong bercangkang tipis dan gonggong bercangkang tebal memiliki pita profil protein yang sama pada berat molekul 11-37 kDa, sedangkan profil protein pada lendir gonggong bercangkang tebal dan tipis memiliki pita protein yang sama pada berat molekul 37 kDa. Jenis protein pada spesies gonggong Bintan diprediksi merupakan protein histon karena hasil amplifikasi menggunakan primer protein histon H2A dan H2B didapatkan gen target pada 75 bp dan uji antimikroba pada bakteri S. aureus dan E. coli memiliki nilai DDH sebesar 25,65±0,02 mm dan 14,45±0,13 mm, sehingga diduga bahwa gonggong Bintan berpotensi sebagai kandidat pangan fungsional khas Kepulauan Riau. ABSTRACTGonggong is one of the sea snails, endemic species living on coastal waters of Bintan Island and surrounding islands of the Riau Islands Province. Sea snail gonggong is an icon of Tanjungpinang-Riau Islands Province. Until now, research this snail is the least, whereas it is potential species. The purpose of this study was to characterize histone protein from Bintan gonggong snail Strombus sp. as functional food candidate. Protein profiling used SDS-PAGE. Protein contents were analyzed by Bradford method. Boiled thick shelled gonggong were extracted by maseration method using ethanol PA 95% and antimicrobial activity tes using well method. Amino acid analized with HPLC. The result of characterization on protein profiles in meat gonggong showed that the thin-shelled and thick-shelled gonggong had the same band as protein profiles by 11-37 kDa and protein profiles in mucus gonggong were found the same band as protein profiles of 37 kDa. The type of protein in spesies Bintan gonggong had been predicted a histone protein because DNA identification using primer protein histone H2A and H2B had gen target of 75 bp. Antimicrobial activity test on S. aureus and E. coli bacteria had value DDH of 25.65±0.02 mm and 14.45±0.13 mm. In fact, gonggong snail was potentially as antimicrobial peptide, so it will make local functional food candidate from Riau Islands Province.
Gonggong snail is one of the famous and highly-priced local seafood in Bintan Island, Riau Islands Province. It is an icon of Tanjung Pinang city. Until now, general research on Bintan gonggong snail is scarce, even if it has the potential to be used for aquaculture, functional food ingredient, and medicine. Research of bioactive peptides are mostly performed using the group of molluscs (gastropods). The research on protein profiles and DNA isolation has never been studied in hemolymph Bintan gonggong snail. This research was aimed to identify protein profiles and DNA isolation in hemolymph Bintan gonggong snail (Strombus sp). Protein profiling used SDS-PAGE. DNA isolation used Qiagen DNeasy methods, while PCR used primer histon H2A. Identification on protein profiles displayed that thin-shelled and thick-shelled gonggong snails had a similiarity value of 37 kDa, whereas the other protein profiles were different. The types of protein of Bintan gonggong snails were predicted to be histone proteins. The DNA of hemolymph Bintan gonggong snails had a similiarity value of 75 bp fragment histon. It is predicted as protein histon H2A and will be developed potentially as antimicrobial peptides (AMPs).
Keberhasilan pemijahan menentukan ketersediaan benih siput gonggong. Tujuan penelitian ini adalah menganalisis efektivitas penggunaan jenis hormon berbeda yaitu ovaprim® dan hCG dalam menstimulasi pemijahan siput gonggong. Delapanpuluh empat pasang induk disuntik sesuai rancangan dengan ovaprim® atau hCG, atau kombinasi keduanya masing-masing dengan dosis 0,5 µL/g BB. Induk pascasuntik dipijahkan dalam akuarium dengan densitas 7 pasang per akuarium. Parameter yang diamati adalah: jumlah induk yang memijah, jumlah telur dikeluarkan oleh induk, waktu latensi, dan tampilan histologi gonad induk. Uji Kruskal Wallis digunakan untuk menganalisis data jumlah induk mijah, dan analysis of variance (ANOVA) untuk menganalisis jumlah telur dikeluarkan oleh induk siput. Hasil penelitian menunjukkan tidak ada perbedaan signifikan (P>0,05) jumlah induk mijah di antara perlakuan, namun pemberian hCG memberikan jumlah induk mijah paling tinggi (52,38%). Waktu latensi induk mijah umumnya mulai mijah pada hari ke-2 dan ke-3 setelah pemberian 0,5 µL/gBB ovaprim. Berdasarkan persentase induk mijah dan nilai seluruh parameter pengamatan disimpulkan bahwa pemberian suntikan hCG dosis 0,5 µL/g BB (P1) memberikan respons lebih baik dibanding perlakuan lain.
Sea snail gonggong is an icon of Tanjungpinang-Riau Islands Province. It is a favorite seafood item in Riau Islands Province, and is high economic value but not known widely yet. Until now, sea snail gonggong has been highly exploited but the research on this snail is very limited. The aim of this study was to morphology and molecular characterization of Bintan gonggong snail (Strombus sp.) as a species validation. Bintan gonggong snail included thick-shelled gonggong and thin-shelled gonggong. Morphology identifcation of species Bintan gonggong snail was based on morphometric variability. Molecular identifcation used partial Histone-H3, MEGA version 6.06, and bioinformatics analysis. The result showed that the morphological identifcation of thick-shelled and thin-shelled gonggong based on shell width, the lip thickness, and total weight signifcantly different, but other variables (i.e shell length, shell depth, aperture length, and gonggong weight) were not signifcantly different (p<0.05). Resulted of a molecular identifcation with phylogenetic analysis that the thin-shelled and thick-shelled Bintan gonggong snails were 1 species and a genetic distance of 1%. They were not species Strombus canarium, Strombus vitatus, and Strombus epidromis. Bintan gonggong snails were Strombus turturella (Leavistrombus turturella). DNA sequences of Bintan gonggong have been registered in Gen-Bank with registration numbers MH348131 (thinshelled gonggong) and MH348132 (thick-shelled gonggong).
Produk tradisional khas dari Kepulauan Riau salah satunya adalah ikan tongkol asap. Produk tersebut tersebar di beberapa kota dan kabupaten di Provinsi Kepulauan Riau. Beberapa daerah yang terkenal memproduksi ikan tongkol asap secara tradisional adalah Kabupaten Natuna, Kabupaten Bintan dan Kota Tanjungpinang. Penelitian ini bertujuan untuk mengetahui karakteristik produk ikan tongkol asap berdasarkan bahan baku jenis ikan yang digunakan, komposisi kimia serta penilaian organoleptiknya. Penelitian dilakukan secara deskriptif dengan mengambil sampel produk ikan tongkol asap dari beberapa pengusaha di Pulau Bintan (Bintan dan Tanjungpinang) dan Pulau Bunguran (Natuna). Tahapan penelitian dilakukan dengan cara mengidentifikasi bahan baku ikan yang digunakan untuk produk ikan tongkol asap, analisis morfometrik, analisis organoleptik/sensori, serta analisis proksimat produk. Pada penelitian ini didapatkan bahwa bahan baku yang digunakan untuk ikan asap adalah jenis ikan Thunnus tonggol, Auxis thazard, Euthynnus affinis dan Katsuwonus pelamis. Secara morfometrik ikan yang digunakan adalah dengan panjang 43,50-46,25 cm dan bobot 1,25-1,85 kg. Pada analisis sensori dengan parameter kenampakan, aroma, rasa dan tekstur, keseluruhan produk yang berasal dari tiga daerah tersebut tidak berbeda nyata. Berdasarkan analisis proksimat, setiap produk ikan asap memiliki nilai tersendiri dari setiap daerah dengan nilai protein kasar berkisar antara 34,04-45,28%. Hasil analisis sensori dan proksimat yang dilakukan diketahui masih sesuai dengan SNI 2723:2013, kecuali pada kadar air jenis ikan asap Euthynus affinis dari Kota Tanjungpinang dan ikan asap Thunnus tonggol dari Kabupaten Bintan.
In the pasca of the COVID-19 pandemic, many researchers have focused on finding drugs for the COVID-19. Peptide research is predicted to have a physiological mechanism of action and it does not cause side effects, therefore it is necessary to search for peptide sources. The aim of this study was to characterize peptides extracted from tamban fish (Sardinella fimbriata) and fermented products (Cincalok of Tanjungpinang). Peptide extract was obtained by maceration extraction using 95% PA (1:5) ethanol solvent, and then it filtered using MWCO 3 kDa and 10 kDa. The amino acid characteristics of the peptides was obtained using HPLC. Peptides of tamban fish had the pH of 7.6 and they are rich of amino acid histidine (890.18 mg/L), in addition to the amino acids glutamate, tyrosine, glycine, leucine, alanine, and aspartic acid. But the peptides of the fermented cincalok product had the pH of 6.4 and they are rich of amino acid leucine (1066.83 mg/L), in addition to the amino acids glutamate, valine, isoleucine, and alanine. Each of these peptides had the molecular weight of 10 kDa. The results of this study can be used as preliminary information to the pharmaceutical development of a new source of peptides.
