Phosphorescence has rarely been observed in pure organic chromophore systems at room temperature. We herein report efficient phosphorescence from the crystals of benzophenone and its derivatives with a general formula of (X-C 6 H 4 ) 2 CdO (X ) F, Cl, Br) as well as methyl 4-bromobenzoate and 4,4′-dibromobiphenyl under ambient conditions. These luminogens are all nonemissive when they are dissolved in good solvents, adsorbed on TLC plates, and doped into polymer films, because active intramolecular motions such as rotations and vibrations under these conditions effectively annihilate their triplet excitons via nonradiative relaxation channels. In the crystalline state, the intramolecular motions are restricted by the crystal lattices and intermolecular interactions, particularly C-H · · · O, N-H · · · O, C-H · · · X (X ) F, Cl, Br), C-Br · · · Br-C, and C-H · · · π hydrogen bonding. The physical constraints and multiple intermolecular interactions collectively lock the conformations of the luminogen molecules. This structural rigidification effect makes the luminogens highly phosphorescent in the crystalline state at room temperature.
A conceptually new "light-up" biosensor with a high specificity for d-glucose (Glu) in aqueous media has been developed. The emission from a tetraphenylethene (TPE)-cored diboronic acid (1) was greatly boosted when the fluorogen was oligomerized with Glu because of restriction of the intramolecular rotations of the aryl rotors of TPE by formation of the oligomer. Little change in the light emission was observed when 1 was mixed with D-fructose, D-galactose, or D-mannose, as these saccharides are unable to oligomerize with the fluorogen.
The phytohormone cytokinin (CK) positively regulates the activity and function of the shoot apical meristem (SAM), which is a major parameter determining seed production. The rice (Oryza sativa L.) Gn1a/OsCKX2 (Grain number 1a/Cytokinin oxidase 2) gene, which encodes a cytokinin oxidase, has been identified as a major quantitative trait locus contributing to grain number improvement in rice breeding practice. However, the molecular mechanism of how the expression of OsCKX2 is regulated in planta remains elusive. Here, we report that the zinc finger transcription factor DROUGHT AND SALT TOLERANCE (DST) directly regulates OsCKX2 expression in the reproductive meristem. DST-directed expression of OsCKX2 regulates CK accumulation in the SAM and, therefore, controls the number of the reproductive organs. We identify that DST(reg1), a semidominant allele of the DST gene, perturbs DST-directed regulation of OsCKX2 expression and elevates CK levels in the reproductive SAM, leading to increased meristem activity, enhanced panicle branching, and a consequent increase of grain number. Importantly, the DST(reg1) allele provides an approach to pyramid the Gn1a-dependent and Gn1a-independent effects on grain production. Our study reveals that, as a unique regulator of reproductive meristem activity, DST may be explored to facilitate the genetic enhancement of grain production in rice and other small grain cereals.
Rice grain with excessive cadmium (Cd) is a major source of dietary Cd intake and a serious threat to health for people who consume rice as a staple food. The development of elite rice cultivars with consistently low Cd content is challenging for conventional breeding approaches, and new strategies urgently need to be developed. Here, we report the development of new indica rice lines with low Cd accumulation and no transgenes by knocking out the metal transporter gene OsNramp5 using CRISPR/Cas9 system. Hydroponic culture showed that Cd concentrations in shoots and roots of osnramp5 mutants were dramatically decreased, resulting in rescue of impaired growth in high Cd condition. Cd-contaminated paddy field trials demonstrated that Cd concentration in osnramp5 grains was consistently less than 0.05 mg/kg, in contrast to high Cd concentrations from 0.33 mg/kg to 2.90 mg/kg in grains of Huazhan (the wild-type indica rice). In particular, the plant yield was not significantly affected in osnramp5 mutants. Furthermore, we developed promising hybrid rice lines with extremely low Cd content in grains. Our work supplies a practical approach to developing Cd pollution-safe indica rice cultivars that minimizes Cd contamination risk in grains.
Poly(N-isopropylacrylamide) was labelled using a fluorogen with an aggregation-induced emission feature by direct polymerization; the label served as a fluorogenic probe that reveals fine details in the thermal transitions in the aqueous solution of the polymer; the working mode was readily tuned between non-monotonic and monotonic by changing the labelling degree of the polymer.
Heat stress due to high environmental temperature negatively influences animal performances. To better understand the biological impact of heat stress, laying broiler breeder chickens were subjected either to acute (step-wisely increasing temperature from 21 to 35°C within 24 hours) or chronic (32°C for 8 weeks) high temperature exposure. High temperature challenges significantly elevated body temperature of experimental birds (P<0.05). However, oxidation status of lipid and protein and expression of heat shock transcription factors (HSFs) and heat shock proteins (HSPs) 70 and 90 were differently affected by acute and chronic treatment. Tissue-specific responses to thermal challenge were also found among heart, liver and muscle. In the heart, acute heat challenge affected lipid oxidation (P = 0.05) and gene expression of all 4 HSF gene expression was upregulated (P<0.05). During chronic heat treatment, the HSP 70 mRNA level was increased (P<0.05) and HSP 90 mRNA (P<0.05) was decreased. In the liver, oxidation of protein was alleviated during acute heat challenge (P<0.05), however, gene expression HSF2, 3 and 4 and HSP 70 were highly induced (P<0.05). HSP90 expression was increased by chronic thermal treatment (P<0.05). In the muscle, both types of heat stress increased protein oxidation, but HSFs and HSPs gene expression remained unaltered. Only tendencies to increase were observed in HSP 70 (P = 0.052) and 90 (P = 0.054) gene expression after acute heat stress. The differential expressions of HSF and HSP genes in different tissues of laying broiler breeder chickens suggested that anti-heat stress mechanisms might be provoked more profoundly in the heart, by which the muscle was least protected during heat stress. In addition to HSP, HSFs gene expression could be used as a marker during acute heat stress.
Regioselective 1,3‐dipolar polycycloadditions of tetraphenylethene (TPE)‐containing diazides 1–3 and bis(aroylacetylene) 4 are initiated by simple heating, affording poly(aroyltriazole)s (PATAs) PI–PIII with high molecular weights in high yields. The PATAs are completely soluble in common organic solvents and stable at temperatures up to 358 °C. Thanks to their TPE units, the polymers show aggregation‐induced emission and work as explosive sensors with high sensitivity. The PATAs are optically transparent in the whole visible spectral region. Their refractive indexes can be tuned to a great extent (Δn ≈ 0.08) by simply changing their alkyl spacer lengths. The modified Abbé numbers of the PATAs are very high (up to 273), indicative of very low optical dispersions in the telecommunication‐important wavelength region. UV irradiation through a photomask quenches the light emissions of the polymers, enabling the generation of two‐dimensional fluorescent images without development. The polymers can be readily photo‐crosslinked, yielding three‐dimensional patterns with high resolutions.
