Juni 1932.)Die Eigenschaften der Proteine variieren mit der Art und der Reihenfolge der miteinander verkniipften Amino-sauren. Das Studium des gegenseitigen Einflusses der Amino-saure-Reste innerhalb der Peptid-Kette war bisher gehemmt durch die Unvollkommenheit unserer Verfahren zur S yn these von Peptiden. Von den klassischen Peptid-Synthesen Emil Fischers hat jene noch den weitesten Anwendungsbereich, die sich der Halogenacyl-halogenide bedientl). Aber auch sie ist beschriinkt auf die wenigen Amino-sauren, die aus Halogen-acylhalogeniden erhiiltlich sind. Wir suchten deshalb nach einem vielseitigeren Verfahren, das uns schon aus dem Grunde erwiinscht war, weil die moderne Ferment-Chemie fiir ihre UnteBuchungen uber Wirkungsmechanismus und Spezifitat der Peptidasen moglichst viele verschiedenartige Peptid-Kombinationen zur Verfiigung haben m a .Alle neueren Versuche zur Peptid-Synthese arbeiten nach folgendem Schema: Die Aminogruppe der ersten Amino-saure wird zuerst durch Einfiihrung einer schiitzenden Atomgruppe R stabilisiert, dann wird das Carboxyl der Amino-saure so verandert, dal3 es zur Kuppelung mit einer zweiten Aminosaure befihigt wird, die Kuppelung wird vollzogen und schlieBlich die Atomgruppe R wieder abgespalten ; damit liegt das fertige Peptid vor. Die Schwierigkeit liegt nun darin, eine geeignete Atomgruppe R zu finden, die sich am SchluS der Synthese so leicht wieder abspalten laat, dal3 die Peptid-Bindung dabei nicht in erheblichem M&e angegriffen wird. Als Schutzgruppe R hat man Acetyl 2, oder Toluolsulfony13) vorgeschlagen ; aber ihre Anwendung hat bei der Peptid-Synthese nur zu vereinzelten Erfolgen gefiihrt.Vie1 allgemeinerer Anwendung fahig ist, wie wir im folgenden zeigen. der Rest C,H,.CH,.O.CO d e r Benzylester-kohlensaure, kurz Carbobenzoxy-Rest (Cbzo) genannt; denn er lafit sich mit Hilfe des leicht zugaglichen Chlorids C,H,.CH,.O.CO.Cl unschwer in Amino-sauren der verschiedensten Art einfiihren (Glykokoll-, d-und d, 1-Alanin, 1-Asparaginsiiure, 1-Asparagin, d-Glutaminsaure, &Glutamin, d, 1-Serin, 1-Cystin, d, 1-Phenyl-alanin, I-Tyrosin. d-Arginin, I-Histidin, ferner Glpcyl-glycin) und was das Wesentliche istdurch einfache katalytische HydrierungP) im offenen GefaB in Form von Toluol und Kohlendioxyd wieder abspalten. Intermediar werden wohl C a r b a mi n s a u r e n auftreten : C,H, . CH, . 0 . CO .NH.CH(R)COOH -+ C,H,.CH, $-HoCO.NH.CH(R).COOH + CO, + NH,.CH(R) .COOH. Tricarbobenzoxy-L-arginine, as its sodium salt, was prepared in strongly alkaline medium via the carbobenzoxylation of L-arginine. This sodium salt, which was in fact a mixture of at least two different isomeric forms, was converted into a mixture of jV"V,"-dicarbobenzoxy-L-arginine and a single pure isomer of sodium tricarbobenzoxy-L-argininate upon treatment with boiling ethanol. Fractionation of these compounds, followed by acidification of the latter material, yielded pure tricarbobenzoxy-L-arginine wherein the basic properties of the guanido group were completely masked. The utility of this compound in the preparation of TV-terminal arginine peptides was demonstrated upon its condensation with amino acid benzyl esters, via the mixed carbonic-carboxylic acid anhydride procedure, followed by catalytic hydrogenolysis of the condensation product. In addition, the V-carboxyanhydride procedure was utilized to effect the transformation of tricarbobenzoxy-L-arginine into Ar"Ar"-dicarbobenzoxy-L-arginine, and of V".V"-dicarbobenzoxy-i.-arginine into V"-carbobenzoxy-L-arginine, as well as the benzyl and methyl ester derivatives of the latter. All of these products are of potential value in the synthesis of C-terminal arginine peptides. In this connection, the esters of V"-carbobenzoxy-L-arginine were employed in the synthesis of the benzyl and methyl esters of tricarbobenzoxy-L-arginyl-V"-carbobenzoxy-L-arginine.
A method of peptide synthesis is described in which the trityl group is used to protect the amino group of amino acids and peptides. These trityl derivatives are coupled with other amino acids by the usual methods. The trityl group is removed under mild conditions, e.g., by acetic acid, by one equivalent of hydrochloric acid or by catalytic hydrogenation.
