Ancient DNA is highly degraded, resulting in very short sequences. Reads generated with modern high-throughput sequencing machines are generally longer than ancient DNA molecules, therefore the reads often contain some portion of the sequencing adaptors. It is crucial to remove those adaptors, as they can interfere with downstream analysis. Furthermore, overlapping portions when DNA has been read forward and backward (paired-end) can be merged to correct sequencing errors and improve read quality. Several tools have been developed for adapter trimming and read merging, however, no one has attempted to evaluate their accuracy and evaluate their potential impact on downstream analyses. Through the simulation of sequencing data, seven commonly used tools were analyzed in their ability to reconstruct ancient DNA sequences through read merging. The analyzed tools exhibit notable differences in their abilities to correct sequence errors and identify the correct read overlap, but the most substantial difference is observed in their ability to calculate quality scores for merged bases. Selecting the most appropriate tool for a given project depends on several factors, although some tools such as fastp have some shortcomings, whereas others like leeHom outperform the other tools in most aspects. While the choice of tool did not result in a measurable difference when analyzing population genetics using principal component analysis, it is important to note that downstream analyses that rely on quality scores can be significantly impacted by the choice of tool.
RESUMOObjetivo: Avaliar as diferenças nas práticas de evidenciação dos ativos intangíveis, conforme especificações das IFRS, em empresas brasileiras e australianas. Fundamento: Os ativos intangíveis possuem relativa subjetividade que dificulta a estimação dos seus futuros benefícios econômicos. Apesar da exigência de divulgação destas informações, existe baixa adesão ao CPC 04, o que pode ser investigado comparando a divulgação brasileira com paí-ses culturalmente distintos, como a Austrália. Esta perspectiva é amparada pela Teoria da Relevân-cia Cultural na Contabilidade, proposta por Gray (1988).
Historicamente, o ensino de artes marciais sempre esteve relacionado a dois aspectos fundamentais: defesa pessoal (parte física) e formação de valores subjetivos importantes para o desenvolvimento e aperfeiçoamento individual dos praticantes. O Kung Fu, arte marcial tradicional de origem chinesa, é pautado nesses aspectos e, portanto, sua prática desenvolve importantes conceitos, tais como liderança, motivação, sucesso, aprendizado, desenvolvimento, respeito, noções de hierarquia, entre outros. Este projeto de extensão irá proporcionar à comunidade da Universidade Federal do Paraná (UFPR) - Setor Palotina e à comunidade externa de Palotina e região a possibilidade de praticar Kung Fu, vivenciando conceitos filosóficos de um dos mais tradicionais estilos dessa arte marcial, o Hung Gar (ou estilo da Família Hung em livre tradução). O objetivo principal é formar um grupo de pessoas (alunos, servidores, membros da comunidade externa) que tenham interesse em praticar o Kung Fu mas não possuam condições financeiras de se matricular nas aulas disponíveis na cidade. Por meio deste projeto, os participantes terão a possibilidade de melhorar aptidões físicas, tais como alongamento, concentração, equilíbrio, coordenação motora, condicionamento e fortalecimento ósseo-muscular, bem como trabalhar conceitos subjetivos como motivação, perseverança, respeito e noções de hierarquia. Além disso, os envolvidos convivem com conceitos, filosofia e cultura chinesas, repassados tanto durante os treinos quanto em aulas teóricas. Por meio deste projeto, os alunos e demais participantes estão obtendo como resultado a consciência do seu papel na sociedade como cidadãos honestos e íntegros, que possuem autocontrole para enfrentar as adversidades que a vida apresenta no seu cotidiano.
