Introdução: A Giardia lamblia, causadora da giardíase em humanos, é um dos protozoários intestinais mais comuns no mundo. A infecção humana pode variar de cistos de giardíase assintomáticos a giardíase sintomática. Atualmente, as opções de tratamento incluem derivados do nitroimidazol; o metronidazol, em particular, tem sido um dos pilares do tratamento por décadas e ainda é amplamente utilizado hoje. O número crescente de relatos de casos de falha na resposta a esse grupo de medicamentos e a outros medicamentos antigiárdicos levantou preocupações e levou à busca de compostos adicionais, alguns dos quais surgiram como resultado da introdução de medicamentos originalmente prescritos para outros doenças. Os princípios ativos de plantas medicinais tem um papel essencial na produção de fitoterápicos e podem ser solução atrativa para falha de tratamento alopático. Objetivo: Verificar plantas com efeito antiparasitário especialmente com atividade giardicida. Materiais e Métodos: A metodologia empregada consiste em revisão de literatura sistematizada, utilizando os Descritores em Ciências da Saúde Giardia, Plants e Medicinal, na base de trabalhos PubMed. Utilizou-se um intervalo de cinco, assim sendo, trabalhos que publicados entre os anos 2018 a 2023. Um total de 55 trabalhos foi encontrado, e após análise dos resumos 15 trabalhos foram incluídos na pesquisa. Resultados: Diversas espécies e formulações são citadas na literatura, sendo que a gênese das pesquisas com plantas é, comumente, etnofarmacologia. Diversas partes das espécies Syzygium aromaticum, Achillea santolina, Allium sativum, Morinda royoc, Cymbagogon citratus, Adenophyllum aurantium, Chenopodium botrys, Rhamnus cathartica, Artemisia annua, Foeniculum vulgare, Ageratum conyzoides e Glycosmis spp., foram testas em ensaios in vitro e demonstraram atividade contra G. lamblia. Estudos in vitro são menos efetivos na medida real da aplicabilidade de plantas como um medicamente, contudo esses ensaios são o ponto de partida para descobertas de compostos inovadores. Conclusão: A etnofarmacologia continua como uma forte aliada na busca de novos compostos bioativos. Os ensaios in vitro têm sido forte aliados na desmistificação de várias plantas utilizadas comumente na farmacopeia popular. Pode-se concluir que algumas espécies vegetais podem conter compostos com potencial terapêutico, podendo se tornar tratamentos alternativos para pacientes que foram refratários aos tratamentos convencionais.
GUILHERME GONÇALVES FERNANDES; MERIANE GONÇALVES RESENDE; CINTIA WILLIANY ALVES SANTOS ROCHA INTRODUÇÃO: O gênero Salmonella incluem numerosos sorovares patogênicos para humanos e uma grande variedade de animais. As salmoneloses, são doenças transmitidas por alimentos mais frequente em humanos, geralmente resulta de água e alimentos contaminados. Essas infecções são responsáveis ??por significativa morbidade e mortalidade em todo o mundo, fazendo-se necessária o desenvolvimento de vacinas eficazes com esses microrganismos. As Ilhas de Patogenicidade de Salmonella (SPIs -"Salmonella Pathogenicity Islands"), SPI-1 e SPI-2, desempenham um papel crucial na interação entre as espécies de Salmonella e células hospedeiras. A SPI-1 é onipresente e altamente conservada nesse gênero, demonstrando o seu papel ativo no processo de invasão nas células. A SPI-1 apresenta cinco proteínas, a saber; SpaO, OrgA, OrgB, InvI e a ATPase hexamérica InvC, que juntamente a outras proteínas garante a internalização das bactérias. OBJETIVOS: Verificar o potencial antigênico da proteína InvI e o potencial desses antígenos para um modelo de vacina anti-Salmonella. METODOLOGIA: Consiste em utilização de ferramentas de bioinformáticas para analise de proteínas e potencial antigênico e vacinal. Utilizou-se uma sequência proteica no site UniProt (B8Y8H6), a ancoragem em MCH-I e identificação regiões imunodominantes foram realizadas utilizando recursos do IEDB Analysis Resource; e a previsão de antígenos protetores e vacinas de subunidades via VaxiJen v2.0. RESULTADOS: A proteína apresentou quatro sequências de aminoácidos em potencial; duas sequências de altos escores para ancoragem em MHC-I ("EEFQEKSKY"; "EEFQEKSKYW") e duas sequências como regiões imunodominantes ("EEIYALLRKQSIVRR"; "AIVEQIAGLKLLLDT"). Contudo nenhuma das sequências apresentou escores significativos para antígenos protetores com potencial vacinal. CONCLUSÃO: A proteína InvI não apresentou potencial antigênico/vacinal através das ferramentas utilizadas. Outros recursos e algoritmos podem ser utilizados visando melhor elucidação da proteína. Vale frisar que a conjunção dos fragmentos proteícos a moléculas imunogênicas ainda podem viabilizar a utilização das sequências analisadas como possíveis vacinas.
Introdução: Escherichia albertii é um enteropatógeno emergente de humanos, que apresenta considerável similaridade a Escherichia coli. Surtos de origem alimentar causados por E. albertii foram relatados recentemente no Japão. Uma vez que essa bactéria possui o gene eae, comumente é identificada erroneamente como E. coli enteropatogênica (EPEC) ou enterohemorrágica (EHEC), devido à sua similitude fenotípicas e genéticas. Objetivo: Almeja-se verificar as principais características da E. albertii e relatar sobre os desafios relacionados a identificação da espécies. Metodologia: Fez-se revisão de literatura especializada consultando o PubMed®, sendo os descritores utilizados Escherichia albertii e E. albertii. Utilizou-se recorte temporal entre 2017 a 2022. Foram incluídos no trabalho um total de 15 artigos. Resultados: Bioquimicamente, essa bactéria apresentam como reações positiva: Vermelho de Metila, Lisina e Ornitina descarboxilase, fermentação do manitol e arabinose, apresentam DNASE à 25ºC, além de produzirem ácidos e gás ao metabolizar a glicose. Cepas de E. albertii são frequentemente identificadas de forma errônea como H. alvei, S. boydii, E. coli ou Y. ruckeri por apresentar provas bioquímicas semelhantes. Um prova bioquímica extremamente relevante seria a fermentação na ramnose, uma vez que a maior parte das cepas de E. coli são capazes de utilizar esse carboidrato e o contrario é observado em cepas de E. albertii. Trabalhos utilizando de biologia molecular têm demonstrado algumas características genéticas únicas ou notáveis de E. albertii, incluindo marcadores responsáveis por características bioquímicas conhecidas e fatores de virulência apresentados por esse microrganismo. Além disso, diversas linhas de pesquisas tem avaliado a possibilidade da identificação através dos antígenos O e H. Conclusão: Pôde-se inferir que a E. albertii, permanece grosseiramente subcaracterizado, apesar de fortes evidências que implicam a bactéria em múltiplos surtos globalmente. Trabalhos visando criar ensaios diagnósticos sejam microbiológicos (provas bioquímicas ou meios cromogênicos), genotípicos ou sorológicos, são necessários frente o avanço desse microrganismo como patógeno emergente.
Introdução: Comumente aquisição de novos equipamentos em laboratório de análises clínicas leva a três possíveis resultados, a saber; venda do aparelho antigo, utilização do aparelho como backup, ou sucateamento do maquinário. Os avanços na tecnologia que desenvolveram equipamentos de laboratórios mecanizados em interface com equipamentos e programas computadorizados providos de vários tipos suporte, dá-se o nome de automação. O mundo da ciência laboratorial clínica sofre constantes mudanças, trazendo consigo diversos desafios. Objetivo: Tem-se como objetivo elucidar sobre a importância de automação utilizada como backup, contrapondo com tecnologias obsoletas. Metodologia: Fez-se revisão de literatura especializada consultando sites de empresas de tecnologia de automação em análises clínicas bem como artigos de que abordagem a temática. Utilizou-se recorte temporal entre 2017 a 2022, sendo os descritores utilizados foram tecnologia, backup, automação, equipamento e laboratório. Resultados: As manutenções preventivas e corretivas dos equipamentos em laboratórios são motivo de preocupação entre os gestores e donos de laboratórios. As manutenções embora sejam de extrema importância para manter a qualidade dos exames, bem como garantir qualidade e longevidade da maquina, podem gerar impactos nos prazos de entregas dos resultados. Fica clara a necessidade de maquinas que assumam a rotina enquanto manutenções e reparos são realizados. Contadores hematológicos de 5 partes podem ter como backup maquinas de mesma tecnologia ou contar como suporte maquinas de 3 partes com análise de lâmina. Aparelhos de bioquímica contam com analisadores menos robustos e até mesmo semi-automatizados. Vale frisar que o fluxo e rotina, bem como a quantidade de amostras norteiam a (in)viabilidade de utilizar uma maquina, dita inferior, como backup. Conclusão: Embora diversas metodologias possam ser tidas como obsoletas e até mesmo arcaicas, fica clara que uma vez garantida a qualidade do ensaio e a compatibilidade com a rotina do laboratório, há algumas tecnologias consideravelmente em conta úteis como backup em laboratórios de análises clínicas.
INTRODUÇÃO:A hanseníase é uma doença infectocontagiosa crônica, tendo como agente etiológico o Mycobacterium leprae, que acomete pele e/ou os nervos periféricos. Qualquer órgão que apresente macrófagos, excetuando o sistema nervoso central, podendo ser acometido, embora não seja tão frequente.
