This study reports an efficient inoculation protocol that allowed cytological analysis of the infection process of the rice false smut pathogen Ustilaginoidea virens. Examination of serial semithin and ultrathin sections of infected spikelets showed that the primary infection sites for the pathogen were the upper parts of the three stamen filaments located between the ovary and the lodicules. The stigma and lodicules were also occasionally infected to a limited extent. The pathogen infected the filaments intercellularly and extended intercellularly along the filament base. The host cells were degraded gradually. The pathogen did not penetrate host cell walls directly and did not form haustoria. In the balls the ovary remained alive and was never infected. This suggests that the pathogen is a biotrophic parasite that grows intercellularly in vivo.
Arbuscular mycorrhizal (AM) fungi in a chronosequence of 5-42-year-old Caragana korshinskii plantations in the semi-arid Loess Plateau region of northwestern China were investigated. AM fungi colonization, spore diversity and PCR-denatured gradient gel electrophoresis-based AM fungal SSU rRNA gene sequences were analyzed. AM fungi colonization [measured as the percent of root length (%RLC), vesicular (%VC) and arbuscular (%AC) colonization] and spore density were significantly correlated with sampling month, but not with plant age, except for %RLC. The percent of vesicular colonization was negatively correlated with soil total nitrogen and organic carbon, and spore density was negatively correlated with soil moisture and available phosphorus. Ten distinguishable AM fungal spore morphotypes, nine Glomus and one Scutellospora species, were found. Nine AM fungal Glomus phylotypes were identified by sequencing, but at each sampling time only four to six AM fungal phylotypes were detected. The AM fungal community was significantly seasonal, whereas the AM fungal species richness did not increase with plantation age. A significant change in AM fungal colonization and community composition over an annual cycle was observed in this study, and our results suggest that the changes of AM are the product of the interaction between host phenology, soil characteristics and habitat. Understanding these interactions is essential if habitat restoration is to be effective.
In order to study the adaptation ability of sweet sorghum (Sorghum bicolor L. Moench) in the Yellow River Delta, the sweet sorghum variety Mart was used in this study to determine the roles of different saline-alkaline ratio stress treatment during seed germination to seedling stage. The results showed that Na+ concentration had a significant impact on the seed germination, seedling growth, and plant survival of sweet sorghum. Increasing Na+ concentration led to a decline in germination rate, final germination percentage, survival percentage, plant height, and dry weight per plant, a prolonged mean time of germination, as well as loss of improvement effect of low-Na+ concentration. The interaction effect of Na+ concentration and pH on the mean time of germination and germination rate was not significant (p<0.05). However, under the condition of low-Na+ concentration (100 mM), high pH reduced the mean time of germination and increased the germination rate, without decline in final germination percentage and survival percentage. Therefore, at least in the duration of seed germination to the harvest period in the research, the sweet sorghum was resistant to the pH stress (≥9.04) when the Na+ concentration was below 100 mM. When suffered from the saline-alkaline stress, the seedling of sweet sorghum was characterized by ecological adaptive features, such as decreased stem ratio and chlorophyll b content in leaves and increased root ratio and chlorophyll a content, in order to maintain the uptakes of water and nutrient, and carbon assimilation. When the stress intensified, the lipid oxidation products, e.g., malondialdehyde (MDA), increased in sweet sorghum seedlings. However, the increasing of soluble protein content and antioxidant enzyme activity (superoxide dismutase (SOD), guaiacol peroxidase (POD), and gatalase (CAT)) was only founded in neutral low-Na+ concentration treatment (A1), which indicated that high-salt concentration and pH all elicited harmful effects and limited the self-healing ability of sweet sorghum seedlings. In all, in order to grow sweet sorghum in the saline-alkaline soils of the Yellow River Delta, the salt concentration and pH value of the soil must be taken into consideration, and seeding density should be increased and supported by appropriate irrigation measures to reduce saline-alkaline stress so as to ensure the survival and growth of sweet sorghum seedlings.
BackgroundNeural tube defects (NTDs) are congenital birth defects that occur in the central nervous system, and they have the highest incidence among all birth defects. Shanxi Province in China has the world’s highest rate of NTDs. Since the 1990s, China’s government has worked on many birth defect prevention programs to reduce the occurrence of NTDs, such as pregnancy planning, health education, genetic counseling, antenatal ultrasonography and serological screening. However, the rate of NTDs in Shanxi Province is still higher than the world’s average morbidity rate after intervention. In addition, Shanxi Province has abundant coal reserves, and is the largest coal production province in China. The objectives of this study are to determine the temporal and spatial variation of the NTD rate in rural areas of Shanxi Province, China, and identify geographical environmental factors that were associated with NTDs in the risk area.MethodsIn this study, Heshun County and Yuanping County in Shanxi Province, which have high incidence of NTDs, were selected as the study areas. Two paired sample T test was used to analyze the changes in the risk of NTDs from the time dimension. Ripley’s k function and spatial filtering were combined with geographic information system (GIS) software to study the changes in the risk of NTDs from the spatial dimension. In addition, geographical detectors were used to identify the risk geographical environmental factors of NTDs in the study areas, especially the areas close to the coal sites and main roads.ResultsIn both Heshun County and Yuanping County, the incidence of NTDs was significantly (P<0.05) reduced after intervention. The results from spatial analysis showed that significant spatial heterogeneity existed in both counties. NTD clusters were still identified in areas close to coal sites and main roads after interventions. This study also revealed that the elevation, fault and soil types always had a larger influence on the incidence of NTDs in our study areas. In addition, distance to the river was a risk factor of NTDs in areas close to the coal sites and main roads.ConclusionThe existing interventions may have played an important role to reduce the incidence of NTDs. However, there is still spatial heterogeneity in both counties after using the traditional intervention methods. The government needs to take more measures to strengthen the environmental restoration to prevent the occurrence of NTDs, especially those areas close to coal sites and main roads. The outcome of this research provides an important theoretical basis and technical support for the government to prevent the occurrence of NTDs.
Soil salinization is a serious problem that affects the seedling growth in many regions. A greenhouse experiment was carried to investigate the adaptation ability of seedlings ( Sorghum bicolor (L.) Moench.) in coastal saline alkaline environment. Seedlings of sorghum were treated by different salt and alkali stress (NaCl: Na 2 SO 4 : NaHCO 3 were 2:1:0, 2:1:1, 2:1:2). The treatments consisted of three levels of salinity (100, 200 and 300 mmol/L) and pH values were 7.08, 8.78 and 9.04. The results showed that the seedlings of sorghum have good adaptability to salt stress under low pH (pH ≤7.08). The plant height, the maximum leave areas of seedlings all dropped and root length first ascended and then descended with the increasing of salt and alkali stress. The contents of Chlorophyll b degraded significantly under salt and alkali stress. Salt and alkali stress stimulated the accumulation of organic solutes (proline and protein) and inorganic ions (Na + , Cl - , SO 4 2- ). Our results showed that salt and alkali stress have significant effect on growth indexes except root length and the interaction effect has significantly on physiology.
