Nas últimas décadas o interesse dos pesquisadores tem se despertado para contaminação ambiental, os quais oferecem risco a saúde humana, mesmo em baixas concentrações. Dentre estes contaminantes destacam-se micropoluentes como os disruptores endócrinos, caracterizados como substâncias químicas que promovem alterações no sistema endócrino humano. Estas substâncias são capazes de se acumular no solo e sedimentos dos rios, acumulando-se na cadeia alimentar, se ligando a receptores endócrinos e promovendo alterações na síntese, secreção, metabolismo e/ou ação hormonais. Estudos demonstram que a exposição de peixes ao 17?-estradiol (E2), induz a feminilização, produção de vitelogênese em machos, redução do índice gônado-somático e inibição do crescimento testicular. Diante disso, o presente estudo tem por objetivo avaliar os possíveis danos genéticos, utilizando a citogenética (testes de aberrações cromossômicas e do micronúcleo), bem como, análise de alterações morfológicas nucleares em espécimes de Astyanax altiparanae expostos ao hormônio E2. Os testes foram realizados em 20 espécimes de A. altiparanae, os quais, inicialmente, foram mantidos em aquários com água declorada, aeração e temperatura ambiente, por aproximadamente 10 dias. Após esse período, os peixes foram distribuídos em 4 grupos/aquários (10L de água), sendo colocados 05 indivíduos em cada um; grupo 1 foi estabelecido como controle negativo contendo somente água e nos demais (2,3 e 4) foram adicionadas soluções do hormônio 17?-estradiol obtido pelo fármaco Natifa® (estradiol 1mg). A concentração usada do hormônio foi 600 ng/L e os animais dos grupos 2,3 e 4 foram monitorados durante 24, 48 e 96h, respectivamente. Em um total de 45 metáfases obtidas encontrou-se três alterações cromossômicas, sendo 3 gaps cromatídicos, dois em um mesmo indivíduo do grupo 4 (96h) envolvendo cromossomos distintos e o outro do grupo 1 (controle). No teste de micronúcleo e da análise morfológica nuclear, foram contadas 2000 células/individuo totalizando 40.000 células analisadas. A frequência de micronúcleos deu-se em 0,085% do total das células visualizadas, com maior incidência no grupo 3, 48h (0,4%) as alterações morfológicas nucleares ocorreram com maior frequência no grupo 2 (0,165%). Esses dados sugerem a principio, um efeito genotóxico do hormônio E2, contudo as análises deverão ser complementadas com outras concentrações.
Loricariidae compreendem em sua maioria, peixes representantes de água doce, conhecidos vulgarmente como cascudos, sendo Hypostominae a maior e mais diversificada, tanto em aspecto morfológico quando citogenético, dentre as subfamílias dos loricarídeos. O objetivo do presente estudo foi caracterizar citogeneticamente duas espécies, Hypostomus aff. paulinus e H. albopunctatus provenientes do Ribeirão Andirá, localizado na cidade de Ourizona (PR), de forma que a descrição desses cariótipos adicionem informações na compreensão dos mecanismos envolvidos na diversificação cariotípica no gênero e sobre o papel da heterocromatina nesses processos. Foram analisados 4 exemplares de Hypostumus aff. paulinus e um exemplar de H. albopunctatus. As metáfases foram obtidas a partir de células renais e submetidas a coloração convencional com Giemsa, Ag-NOR, bandamento C e coloração com os fluorocromos base-específicos DAPI e CMA3. Os indivíduos da espécie Hypostomus aff. paulinus apresentaram um cariótipo com 2n=76 cromossomos, sendo 10m+14sm+24st+28a. O exemplar de H. albopunctatus apresentou um cariótipo com 2n=74 cromossomos, sendo 8m+14sm+16st+36a. O padrão de heterocromatina revelado pelo bandamento C consistiu em blocos heterocromáticos presentes em regiões pericentroméricas de vários cromossomos, com destaque para extensos blocos nos braços longos nos pares 25 e 29 para indivíduos de Hypostomus aff. paulinus. Esta heterocromatina revelou-se positiva para DAPI. Ainda nesta espécie um par de submetacêntricos e um dos homólogos de um par de acrocêntrico grande revelaram-se positivos para CMA3/Ag-NORs em posições terminais do braço curto e braço longo, respectivamente. Em H. albopunctatus, o exemplar apresentou blocos fortemente corados nas regiões pericentroméricas da maioria dos cromossomos; o par 25 apresentou forte marcação banda C em posição subterminal do braço longo. Ainda em H. albopunctatus um par submetacêntrico mostrou-se CMA3 positivo/Ag-NOR para região subterminal do braço curto com um evidente heteromorfismo de tamanho. Comparando os dados obtidos com estudos citogenéticos prévios realizados em outras populações de ambas espécies, sugere-se que os cromossomos que ficaram evidentemente marcados em Hypostomus. aff. paulinus e H. albopunctatus sejam possíveis marcadores para estas espécies.
A utilização de animais em pesquisas sempre foi controverso sendo a razão de diversas discussões em função do grande número necessário de espécimes e do sofrimento causado aos mesmo. Com os recentes avanços da citogenética de peixes reforça-se o interesse na análise cromossômica na área básica e aplicada da pesquisa. O protocolo mais empregado para obtenção de preparações cromossômicas em peixes neotropicais tem sido a técnica "air drying", que tem por base a injeção prévia de solução de colchicina nos exemplares vivos e a retirada da porção anterior do rim para a obtenção de uma suspensão celular. Embora esta técnica comumente empregada resulte em preparações cromossômicas de qualidade, tem a desvantagem da eutanásia dos animais. As questões éticas e morais que permeiam nossa sociedade contemporânea estabelecem alguns princípios orientadores na utilização de animais em pesquisas científicas. Entre estes princípios estão a substituição dos animais (replace) por outros métodos alternativos, tais como: testes in vitro, e a redução (reduce), que determina que os pesquisadores devem utilizar o mínimo de animais em um experimento. Deste modo, nosso estudo teve como objetivo experimentar e adequar uma metodologia alternativa (protocolo estabelecido por Völker e. Ráb, 2015) que utiliza nadadeiras regeneradas de peixes para a obtenção de metáfases mitóticas, com a vantagem de não provocar a morte dos espécimes. Foram testados um totade 24 de animais pertencentes a quatro espécies diferentes: Danio rerio, Astyanax altiparanae, Oreochromis niloticus e Otothyropsis biamnicus. Os resultados obtidos demonstraram que houve variação no tempo de regeneração das nadadeiras entre as espécies analisadas e, que o número médio de metáfases por lâmina foi igual a 4. Embora essa quantidade possa parecer pequena, o tempo para percorrer a lâmina à procura de metáfases é bem menor, uma vez que as células ficam concentradas em uma região especifica da lâmina. A qualidade das metáfases obtidas também foi satisfatória. Desta forma, a preparação cromossômica direta de nadadeiras regeneradas de peixes parece ser promissora na obtenção metáfases em quantidade e qualidade. Este fato é de extrema importância para as análises citogenéticas, pois a melhoria nas preparações cromossômicas obtidas diminui a necessidade de coletas subsequentes e o uso excessivo de exemplares.
Os agrotóxicos têm sido utilizados como um modelo de estudos em ecotoxicologia e quando atingem ambientes aquáticos podem ser altamente impactantes, reduzindo a biodiversidade. Diversos poluentes podem causar danos genotóxicos em peixes, resultando em aberrações cromossômicas (AC), como quebras cromossômicas ou cromatídicas, gaps, micronúcleos (MN) e, ainda alterações morfológicas nucleares em hemácias (AMN). Assim, o presente estudo tem por objetivo avaliar possíveis danos citogenéticos (aberrações cromossômicas e micronúcleos) e morfológicos (análise nuclear das hemácias) em espécimes de A. altiparanae (obtidos comercialmente), quando expostos ao agrotóxico comercial 2-4- Diclorofenoxiacético. Este composto atua como hormônio regulador do crescimento de ervas perenes e de folhas largas, em culturas de milho, cana-de-açúcar e pastagens sendo amplamente utilizado na agricultura. Os testes foram realizados em 24 espécimes (18 fêmeas e 06 machos) os quais, inicialmente, foram acondicionados e mantidos em aquários com água desclorada, aeração e temperatura ambiente, por aproximadamente 10 dias. Após esse período, os peixes foram distribuídos em 04 aquários (10L de água), sendo colocados 06 indivíduos em cada um; um aquário foi estabelecido como controle negativo contendo somente água e nos demais (3) foram adicionadas soluções de 2-4 D com as concentrações de 25, 70, 80 ?l/10L. Os peixes receberam estes tratamentos durante 24h para cada concentração. Dentre as 70 metáfases analisadas, no grupo controle não foram encontradas AC, contudo, nas concentrações de 25 e 70 ?l/10L, encontramos 04 quebras cromatídicas (5,71%) e 03 gaps cromatídicos (4,28%). Para análise do MN considerou-se a contagem de 2000 células/indivíduo, totalizando 48.000 células analisadas. A presença de micronúcleos foi muito baixa nestas células, sendo observada somente uma célula com 01 MN, tantos nos peixes do controle negativo, quanto naqueles tratados com a concentração de 80 ?l/10L, o que corresponde a (0,016%) para cada grupo. Por outro lado, foram encontrados diferentes tipos de AMN nos três tratamentos, totalizando 165 hemácias com alterações (0,33%) em relação ao total analisadas. Os resultados demonstraram a toxicidade do 2-4D ao nível cromossômico e nuclear sugerindo que esta substância ao penetrar no organismo vivo e em pouco tempo pode provocar danos genéticos significativos.Apoio: CNPQ – UEM / Programa de Pós Graduação em Biotecnologia Ambiental/ Universidade Estadual de Maringá (UEM).
Rineloricaria é caracterizado por uma ampla variabilidade cromossômica e relatos de polimorfismos numéricos e/ou estruturais são relativamente comuns para este gênero. A bacia do Rio Iguaçu, a qual pertence a população em estudo, caracteriza-se por uma ictiofauna endêmica tornando-a de grande interesse para estudos filogenéticos e evolutivos. Estudos prévios realizados para uma população do Rio Iguaçu no estado do Paraná, revelaram um expressivo polimorfismo cromossômico numérico e estrutural de 2n=64 a 68, com diversas fórmulas cariotípica em Rineloricaria aff. langei. Sendo assim, com a finalidade de investigar se há mais de uma espécie para a população em estudo, foi sequenciado gene mitocondrial Citocromo C Oxidase I (COI) de 12 indivíduos pertencentes a população de Rineloricaria aff. langei. O COI foi isolado através da amplificação com primers específicos gerando fragmentos de aproximadamente 500 pb. Analises filogenéticas utilizando os métodos de máxima verossimilhança e analise bayesiana evidenciaram a distinção de dois clados para esta população, com divergência genética média acima de 3%. O clado 1 agrupou 8 cariótipos prováveis: cariótipo A com 2n= 65, fórmula cariotípica 3m+62st/a e NF=68, B com 2n=65, 4m+61st/a e NF=69, C com 2n=66, 3m+63st/a e NF=69, D com 2n=67, 3m+64st/a e NF=70, E com 2n=65, 4m+61st/a e NF=69, F com 2n=67, 4m+63st/a e NF=71, G com 2n=67, 1m+66st/a e NF=68 e H com 2n=67, 2m+65st/a e NF=69. Enquanto, o clado 2 agrupou 4 cariótipos: cariótipo I com 2n=66, 2m+64st/a e NF=68, J com 2n=65, 3m+62st/a e NF=68, K com 2n=65, 6m+59st/a e NF=71, e L com 2n=67, 3m+64st/a e NF=70. Os dados moleculares, obtidos no presente estudo, suportam a hipótese de novas espécies, as quais podem estar envolvidas em um mecanismo de diferenciação de espécies decorrente de rearranjos cromossômicos tais como, translocação e/ou fusão e que, embora seja um polimorfismo extenso, a manutenção deste seja garantida pelos cruzamentos entre os indivíduos, com formação de gametas viáveis e que se perpetuam na população dado as características comportamentais sedentárias desta espécie.Suporte financeiro: CAPES – PBC/UEM
