Although cows with subclinical mastitis have no difference in the appearance of their milk, milk composition and milk quality are altered because of the inflammation. To know the changes in milk quality with different somatic cell count (SCC) levels, 5 pooled bovine milk samples with SCC from 10(5) to 10(6) cells/mL were analyzed qualitatively and quantitatively using both one-dimension sodium dodecyl sulfate PAGE and filter-aided sample preparation coupled with dimethyl labeling, both followed by liquid chromatography tandem mass spectrometry. Minor differences were found on the qualitative level in the proteome from milk with different SCC levels, whereas the concentration of milk proteins showed remarkable changes. Not only immune-related proteins (cathelicidins, IGK protein, CD59 molecule, complement regulatory protein, lactadherin), but also proteins with other biological functions (e.g., lipid metabolism: platelet glycoprotein 4, butyrophilin subfamily 1 member A1, perilipin-2) were significantly different in milk from cows with high SCC level compared with low SCC level. The increased concentration of protease inhibitors in the milk with higher SCC levels may suggest a protective role in the mammary gland against protease activity. Prostaglandin-H2 D-isomerase showed a linear relation with SCC, which was confirmed with an ELISA. However, the correlation coefficient was lower in individual cows compared with bulk milk. These results indicate that prostaglandin-H2 D-isomerase may be used as an indicator to evaluate bulk milk quality and thereby reduce the economic loss in the dairy industry. The results from this study reflect the biological phenomena occurring during subclinical mastitis and in addition provide a potential indicator for the detection of bulk milk with high SCC.
A model forthe development, growth and potential production of cotton (SUCROS-Cotton) was developed.Particular attention was given to the phenological development of the plant and the plasticity of fruit growth in response to temperature, radiation, daylength, variety traits, and management. The model is characterized by a comparatively simple code and transparent algorithms. The model was parameterized for Chinese cotton varieties and validated with extensive independent data sets on cotton growth and production from the Yellow River region and Xinjiang Province. The model validation showed that the phenology, growth and yield were simulated satisfactorily. The root mean square error (RMSE) for date of emergence, date of flowering, date of open boll stage and duration from sowing to boll opening was less than four calendar days, both for cotton grown in monoculture and cotton grown in a relay intercropping system with wheat. The RMSE of predicted total dry matter compared with observations was at most 6.6%, oflint yield 6.6%, and for number of harvestable bolls IO.O%. SUCROS-Cotton provides a tool to (I) assess production opportunities of cotton in various ecological zones in response to temperature, incoming radiation and management, (2) identify optimal cotton ideotypes for different agro-ecological conditions and for guiding breeding efforts, and (3) explore resource-use-efficient cropping systems, including intercropping options, and crop management practices such as plastic film mulching and sowing date.
Net primary productivity (NPP) of grasslands is a key variable for characterising carbon cycles in grassland ecosystems. The prediction of NPP in Inner Mongolia is important for adaptation to future climate change, food security and sustainable use of the grassland resources. The output from two models, potentially suitable for simulating NPP in response to climate change, was tested against observed aboveground forage mass of dry matter at eight sites in Inner Mongolia from 1995 to 2005. The Classification Indices-Based Model (CIBM) showed an acceptable agreement with field measurements. The impact of climate change on the NPP of grasslands was subsequently analysed by CIBM using future climate projections from a Global Circulation Model based on three greenhouse gas emission scenarios: A2 (medium-high emission), A1B (medium emission) and B2 (medium-low emission) differing in assumptions about patterns of global social and economic development. Generally, significant increases in NPP, compared with the baseline NPP of 3.6 tonnes ha–1 for 1961–90, were predicted. The magnitude of the increase in NPP depended on the emission scenario, as well as on the time frame and region considered. Overall the predicted NPP stimulation increased with the level of emissions assumed, being 4.8 tonnes ha–1 in the A2 scenario, 4.3 tonnes ha–1 in the B2 scenario and 4.5 tonnes ha–1 in the A1B scenario in the 2080s (2071–2100). The increase in NPP in response to climate change differed between regions and there was an interaction with emission scenario. For the A2 and the B2 emission scenarios, the western region of Inner Mongolia was predicted to exhibit the strongest NPP increases, but, under the A1B scenario for the 2050s, the south-eastern region exhibited the greatest increase in NPP. It is concluded that the productivity of grassland in Inner Mongolia is likely to increase in response to climate change but these predicted effects are sensitive to emission scenarios and differ regionally. This will provide opportunities but also challenges for herders and policy makers in adapting to this change.
Porcine vertebral and teat numbers are variable and important economic traits in pig production. However, the quantitative trait loci (QTL) and candidate genes for both of these traits in the Beijing Black Pig are not yet known. In the present study, number of vertebrae and number of teats were obtained for 891 individuals of the Beijing Black Pig and genotyped using the Illumina Porcine 50 K BeadChip. Genome scanning was performed to detect associated variants and candidate genes for both traits using a genome-wide association study by TASSEL software. For vertebral number, 15 significant SNPs were located on SSC7. According to linkage disequilibrium analysis on SSC7, a haplotype block of 221 kb from 97.4 to 97.6 Mb was shown to contain a good candidate gene ABCD4. Interestingly, on SSC12, we recorded a novel QTL containing three significant SNPs and 34 annotated genes from 24.0 to 25.7 Mb for vertebral number. Of the 34 genes, nine Hox family genes (HOXB 1-7, 9, and 13) were found to be good candidate genes. Using the 34 genes, a gene ontology analysis was performed to detect enrichment of anterior/posterior pattern specification. For teat number, a novel chromosome-wide significant QTL was identified on SSC10. In this QTL region, one significant SNP was identified. The nearest gene, NTRK2, was regarded as a candidate gene. The present results expanded the QTL for vertebral and teat numbers and provided useful molecular markers for breeding in the Beijing Black Pig population.
Ticks act as important vectors of infectious agents, and several emerging tick-borne viruses have recently been identified to be associated with human diseases in northeastern China. However, little is known about the tick virome in northeastern China. We used metatranscriptomics to investigate the virome diversity in Haemaphysalis japonica, H. concinna, Dermacentor silvarum, and Ixodes persulcatus ticks collected from northeastern China. In total, 22 RNA viruses were identified, including four each in the Nairoviridae and Phenuiviridae families, three each in the Flaviviridae, Rhabdoviridae, and Solemoviridae families, two in the Chuviridae family, and one each in the Partitiviridae, Tombusviridae families and an unclassified virus. Of these, eight viruses were novel species, belonging to the Nairoviridae (Ji’an nairovirus and Yichun nairovirus), Phenuiviridae (Mudanjiang phlebovirus), Rhabdoviridae (Tahe rhabdovirus 1-3), Chuviridae (Yichun mivirus), and Tombusviridae (Yichun tombus-like virus) families, and five members were established human pathogens, including Alongshan virus, tick-borne encephalitis virus, Songling virus, Beiji nairovirus, and Nuomin virus. I. persulcatus ticks had significant higher viral species than H. japonica, H. concinna, and D. silvarum ticks. Significant differences in tick viromes were observed among Daxing’an, Xiaoxing’an and Changbai mountains. These findings showed an extensive diversity of RNA viruses in ticks in northeastern China, revealing potential public health threats from the emerging tick-borne viruses. Further studies are needed to explain the natural circulation and pathogenicity of these viruses.
