Gastrointestinal (GI) parasitic infection is a serious issue in cattle management. The effects of GI parasites may vary with age, sex of cattle, nutritional condition, and severity of infection. Prevalence of GI parasites among cattle population in Gampaha District has not been studied and there is no published study available. A total of 45 farms rearing cattle were selected randomly in three areas, namely, Kelaniya, Ganemulla, and Welisara, under three Veterinary Surgeon Divisions (VSD) in Gampaha District (Mahara, Gampaha, and Welisara). Freshly voided cattle fecal samples were collected randomly from the selected farms during March 2017–December 2017. Out of 163 cattle and buffaloes examined, 13.39% (n=22) were positive for eggs of one or more species of GI parasites. The prevalence of parasitic infection was higher in buffaloes (31.25%, 5/16) as compared to that of cows (11.56%, 21/147), but the difference was not significant (P >0.05). Hookworms (Bunostomum spp.), whipworms (Trichuris spp.), digenetic trematodes (Paramphistomum spp.), cestodes (Moniezia spp.), and oocysts of protozoans (coccidians) were found during the study. The nontreated animals indicated the highest percentage of parasitic infections accounting for 46.67% (n= 14), followed by partially treated individuals (15.15%, n= 5). GI parasite prevalence in males was higher when compared to that of females, but the difference was nonsignificant (P >0.05). General Linear Modelling (GLM) revealed that the effect of treatment status was significantly associated with the prevalence of GI parasites. The calves and yearlings had the highest rate of GI parasitic infections. The highest infection rate was observed at Kelaniya, followed by Welisara. Future investigations are necessary to evaluate the economic impact of GI parasites in the study areas.

Entomopathogenic nematodes (Heterorhabditidae and Steinernematidae) were isolated from Sri Lankan soils using the Galleria baiting technique. Samples collected from different agro-ecological regions and different habitat types revealed that the presence of nematodes was restricted to sandy coastal soils. Two heterorhabditid and three steinernematid RFLP types were discovered within 5–100 m of the sea. Members of both families were equally prevalent. However, no parasitized insects were found although insect species were encountered which are potentially natural hosts. These nematodes were present at high temperatures (some above 30°C) and neither the monsoon rain nor the drought had any effect on their prevalence.

Experiments were conducted to (i) evaluate the efficacy of propanil formulations available in Sri Lanka in controlling Echinochloa crus‐galli; (ii) study the seedling growth of propanil‐resistant (R) and ‐susceptible (S) biotypes of the weed under different temperatures; (iii) quantify the level of resistance in R biotypes and; (iv) to suggest alternative control measures for R biotypes. Field studies showed that retail propanil formulations (36% a.i., EC) applied at 2.7 kg a.i. ha−1 gave less than 30% control of E. crus‐galli collected from several locations of the north dry zone of Sri Lanka. Chemical analysis revealed that there was no adulteration of propanil formulations at the retailer level. Growth studies conducted in controlled environments indicated that per cent germination and seedling growth of R and S biotypes were similar at the day/night temperature regimes imposed. However, per cent germination for plants grown under a 34/31°C (day/night) regime was 27–29% higher compared to those grown at 28/24°C. At the higher temperature regime, R and S biotypes reached the 2–3 leaf stage five days earlier, and the 4–5 leaf stage seven days earlier. The ED50 values from the dose–response experiments indicated that the R biotype was four times more resistant to propanil than susceptible ones. The resistance index (RI) did not vary significantly under different temperature regimes. Quinclorac (25% a.i., SC) applied at 200 g a.i. ha−1 and bispyribac‐sodium (10% a.i., SC) applied at 30 g a.i. ha−1 (recommended dosages) successfully controlled propanil‐resistant biotypes of E. crus‐galli. Conversely, oxadiazon and propanil (8% and 23% a.i., EC, respectively) applied at 280 + 805 g a.i. ha−1 did not result in satisfactory control.

Larval diet quality and rearing conditions have a direct and irreversible effect on adult traits. Therefore, the current study was carried out to optimize the larval diet for mass rearing of Aedes aegypti, for Sterile Insect Technique (SIT)-based applications in Sri Lanka. Five batches of 750 first instar larvae (L1) of Ae. aegypti were exposed to five different concentrations (2-10%) of International Atomic Energy Agency (IAEA) recommended the larval diet. Morphological development parameters of larva, pupa, and adult were detected at 24 h intervals along with selected growth parameters. Each experiment was replicated five times. General Linear Modeling along with Pearson's correlation analysis were used for statistical treatments. Significant differences (P < 0.05) among the larvae treated with different concentrations were found using General Linear Modeling in all the stages namely: total body length and the thoracic length of larvae; cephalothoracic length and width of pupae; thoracic length, thoracic width, abdominal length and the wing length of adults; along with pupation rate and success, sex ratio, adult success, fecundity and hatching rate of Ae. aegypti. The best quality adults can be produced at larval diet concentration of 10%. However, the 8% larval diet concentration was most suitable for adult male survival.

The present study reports mosquito larvicidal potential of green synthesized silver nanoparticles by using Annona glabra leaves (An-AgNPs). Synthesized An-AgNPs were characterized by Ultraviolet-Visible spectroscopy (UV-VIS), Scanning Electron Microscopy (SEM), Dynamic Light Scattering (DLS) technique and Fourier transform infrared spectroscopy (FTIR). Colur change from pale yellow to brick red of the plant extract and AgNO 3 solution indicated the formation of An-AgNPs initially. Surface Plasmon Resonance (SPR) band at 435 nm in the UV-Vis confirmed the formation of An-AgNPs. SEM images showed that An-AgNPs were spherical in shape. FTIR proved that An-AgNPs were functionalized with biomolecules in A. glabra leaves. Based on DLS analysis the average size range of synthesized An-AgNPs was determine to be 10-100 nm and 100-1000 nm.Third instar larvae of dengue vector mosquitoes, Aedes aegypti and Aedes albopictus were subjected to larvicidal bioassays in a range of concentrations of An-AgNPs and A. glabra crude aqueous leaf extract (2-10 mg/L). An-AgNPs exhibited very high larvicidal activity against dengue vector mosquito larvae; LC 50 value for Ae. aegypti at 24 h exposure to An-AgNPs (Plant extract: AgNO 3 1 : 10) 5.29 mg/L; An-AgNPs (Plant extract: AgNO 3 2 : 10) 2.43 mg/L while LC 50 value for Ae. albopictus at 24 h exposure to An-AgNPs (Plant extract: AgNO 3 1:10) 3.02 mg/ L; An-AgNPs (Plant extract: AgNO 3 2:10) 2.51 mg/L. LC 50 values obtained for A. glabra leaf extract tested against Ae. aegypti and Ae. albopictus are 5.94 mg/L and 5.00 mg/L respectively at 24-hour exposure. This study further revealed that Ae. albopictus is more susceptible than to Ae. aegypti to a given concentration of An-AgNPs and to crude aqueous leaf extract of A. glabra. Larvicidal effect of An-AgNPs is superior to the crude aqueous leaf extract of A. glabra. An-AgNPs is a potent larvicide for dengue vector control.

Immature mosquitoes are aquatic, and their distribution, abundance, and individual fitness in a particular breeding habitat are known to be dependent on mainly three factors: biotic factors, abiotic factors, and their interaction between each other and with other associated taxa. Mosquito breeding habitats harbor a diversified naturally occurring microbiota assemblage, and the biota have different types of interactions with mosquito larvae in those habitats. Those interactions may include parasitism, pathogenism, predation, and competition which cause the mortality of larvae, natural reduction of larval abundance, or alterations in their growth. Many microbiota species serve as food items for mosquito larvae, and there are also some indigestible or toxic phytoplanktons to larvae. However, when there is coexistence or mutualism of different mosquito species along with associated microbiota, they form a community sharing the habitat requirements. With the available literature, it is evident that the abundance of mosquito larvae is related to the densities of associated microbiota and their composition in that particular breeding habitat. Potential antagonist microbiota which are naturally occurring in mosquito breeding habitats could be used in integrated vector control approaches, and this method rises as an ecofriendly approach in controlling larvae in natural habitats themselves. To date, this aspect has received less attention; only a limited number of species of microbiota inhabiting mosquito breeding habitats have been recorded, and detailed studies on microbiota assemblage in relation to diverse vector mosquito breeding habitats and their association with mosquito larvae are few. Therefore, future studies on this important ecological aspect are encouraged. Such studies may help to identify field characteristic agents that can serve as mosquito controlling candidates in their natural habitats themselves.

Background The lack of information on behavioural patterns of Aedes aegypti and Aedes albopictus has become a significant limitation in vector control and disease management programmes. Therefore, the current study was focused on determining some bionomics aspects: breeding, resting, host-seeking and feeding preferences of Ae. aegypti and Ae. albopictus in Sri Lanka. Methods Larval and adult surveys were conducted from April 2017 to April 2019 monthly in six selected Medical Officer of Health (MOH) areas in Gampaha Distinct, Western province, Sri Lanka, representing urban, suburban and rural settings. Resting preferences of adult mosquitoes were observed from indoor and outdoor places using a Prockopack aspirator. The information on resting height, surface, material and locality was recorded. Human-baited double-net traps were used to determine the host-seeking time of Aedes mosquitoes. Statistical differences in the spatial distribution of mosquitoes in selected MOH areas and prevalence of vectors were analysed using general linear model (GLM). A chi-square test was used to analyse the resting behaviour. Results Total of 19,835 potential breeding sites were examined at 13,563 premises, and 18.5% (n = 1856) were positive for Aedes larvae. Distribution of Ae. aegypti and Ae. albopictus was statistically significant at species level (df = 1; F = 137.134; P < 0.05 GLM) and study setting (df = 2; F = 8.125; P < 0.05). Aedes aegypti breeding was found mainly in temporary removals (18.8%; n = 34), discarded non-degradables (12.15%; n = 22) and tyres (9.95%; n = 18). Natural (14.7%; n = 246) and temporary removals (13.6%; n = 227) and discarded non-reusable items were the key ovipositing sites for Ae. albopictus. In the adult mosquito survey, the majority was comprised of Ae. albopictus (54.5%; n = 999), which denoted exophilic nature (90.8%; n = 758), and 45.5% (n = 835) represented by Ae. aegypti mosquitoes who were mainly endophilic (84.3%; n = 842). Aedes aegypti rested on cloth hangings and curtains, followed by the furniture, while Aedes albopictus was predominant in outdoor vegetation. In both vectors, biting patterns denoted a typical diurnal pattern with two peaks of host-seeking and biting activity in the morning and afternoon. Conclusions The majority (80%) of the larval habitats were artificial containers. The use of larvicides for vector control as the prominent measure is questionable since applying these chemicals may target only 20% of the total breeding grounds, which are permanent. The resting places of adult mosquitoes are mainly indoors. Therefore, using thermal space spraying of insecticide may not be appropriate, and indoor residual spraying is recommended as a suitable intervention to target adult mosquitoes. This study warrants a holistic vector control approach for all medically important mosquitoes and insects, ensuring the rational use of finance and resources. Graphical Abstract
