Реферат: Разработка безотмывочных методов криоконсервирования фракции ядросодержащих клеток кордовой крови, в состав которой входит популяция гемопоэтических стволовых клеток-предшественников, является актуальной задачей для их долгосрочного хранения. В работе использована технология криоконсервирования ядросодержащих клеток кордовой крови, включающая выделение популяции клеток оригинальным методом двухэтапного центрифугирования, обработку непроникающим криопротектором полиэтиленоксидом с м. м. 1500 (ПЭО-1500) и замораживание по двухэтапной программе. Установлено, что данная технология позволяет сохранить более 72% CD45 + -и 75% CD34 + -клеток с показателями жизнеспособности более 83 и 91% соответственно. При этом повышенное содержание активных форм кислорода (в (42,7 ± 6,3)% клеток) на фоне высоких показателей количества клеток и их жизнеспособности может рассматриваться как физиологичный ответ клеток на воздействие факторов криоконсервирования.Ключевые слова: гемопоэтические стволовые клетки, ядросодержащие клетки, кордовая кровь, криоконсервирование, непроникающий криопротектор, ПЭО-1500.Реферат: Розробка безвідмивних методів кріоконсервування фракції ядровмісних клітин кордової крові, до складу якої входить популяція гемопоетичних стовбурових клітин-попередників, є актуальним завданням для їх довгострокового зберігання. У роботі використовується технологія кріоконсервування ядровмісних клітин кордової крові, яка включає виділення популяції клітин оригінальним методом двоетапного центрифугування, обробку непроникаючим кріопротектором поліетиленоксидом із м. м. 1500 (ПЕО-1500) і заморожування за двоетапною програмою. Встановлено, що представлена технологія дозволяє зберегти більше 72% CD45 + -та 75% CD34 + -клітин із показниками життєздатності більше 83 і 91% відповідно. При цьому підвищений вміст активних форм кисню (в (42,7 ± 6,3)% клітин) при високих показниках кількості клітин та їх життєздатності може розглядатися як фізіологічна відповідь клітин на вплив факторів кріоконсервування.Ключові слова: гемопоетичні стовбурові клітини, ядровмісні клітини, кордова кров, кріоконсервування, непроникаючий кріопротектор, ПЕО-1500.Abstract: Development of wash-free cryopreservation methods for cord blood nucleated cell fraction, including the population of hematopoietic stem/progenitor cells, is an actual task for their long-term storage. Present investigation involved the technique of cord blood nucleated cells cryopreservation, comprising the isolation of cell population by the original two-step centrifugation method, the treatment with non-penetrating cryoprotectant polyethylene oxide with molecular weight of 1500 (PEO-1500) and freezing by the two-step program. Реферат: В работе изучали влияние полиэтиленгликоля с м. м. 1500 (ПЭГ-1500) и сахарозы на изменение геометрических параметров и осмотической хрупкости эритроцитов человека. При равных массовых долях двух веществ сахароза вызывала более выраженные изменения в распределении эритроцитов по показателям прямого светорассеивания (FSC) в сравнении c ПЭГ. Тем не менее, ПЭГ индуцировал сдвиг распределения эритроцитов по индексу сферичности в нап-равлении сферических типов клеток, а также повышал осмотическую хрупкость эритроцитов относительно интактных и инкубированных в присутствии сахарозы клеток. Превалирующей направленностью модификации формы в присутствии ПЭГ был стоматоцитоз, в то время как влияние сахарозы приводило к доминированию дегидратированных уплощенных кле-ток. Различия внешних проявлений ответа эритроцитов на растворы ПЭГ и сахарозы, сопровождаемые неодинаковыми изменениями осмотической хрупкости, свидетельствуют о роли структурных модификаций мембран, индуцированных крио-протекторами, в стабилизации клеток при последующем замораживании.Ключевые слова: эритроцит, геометрические параметры, криопротекторы, полиэтиленгликоль, сахароза, осмотическая хрупкость.Реферат: У роботі вивчали вплив поліетиленгліколя м. м. 1500 (ПЕГ-1500) і сахарози на зміну геометричних параметрів та осмотичної крихкості еритроцитів людини. При рівних масових частках двох речовин сахароза викликала більш виражені зміни в розподілі еритроцитів за показниками прямого світлорозсіювання (FSC) в порівнянні з ПЕГ. Проте ПЕГ індукував зсув розподілу еритроцитів за індексом сферичності в напрямку сферичних типів клітин, а також підвищував осмотичну крихкість еритроцитів щодо інтактних і інкубованих у присутності сахарози клітин. Переважаючою спрямованістю модифікації форми в присутності ПЕГ був стоматоцитоз, у той час як вплив сахарози призводив до домінування дегідратованих сплощених клітин. Відмінності зовнішніх проявів відповіді еритроцитів на розчини ПЕГ і сахарози супроводжуються неоднаковими змінами осмотичної крихкості, свідчать про роль структурних модифікацій мембран, індукованих кріопротекторами, у стабі-лізації клітин при подальшому заморожуванні.Ключові слова: еритроцит, геометричні параметри, кріопротектори, поліетиленоксид, сахароза, осмотична крихкість. Abstract:The effect of the polyethylene glycol with a molecular weight of 1500 (PEG-1500) and sucrose on the changes in geometric parameters and osmotic fragility of human erythrocytes was studied. At equal mass fractions of two substances, sucrose caused more pronounced changes in the distribution of erythrocytes according to direct light scattering (FSC) in comparison with PEG. Nevertheless, PEG induced a shift in the distribution of erythrocytes according to the sphericity index in the direction of the spherical cell types, and also increased the osmotic fragility of the erythrocytes relative to the intact and incubated in the presence of sucrose cells.
