Hematology Profile of Sprague-Dawley Male Rats (Rattus norvegicus) Aged 7 and 10 WeeksCurrently, the normal hematology profile has not been yet available for outbred rats in Indonesia. Most of the researchers use the data of hematology profile from references outside Indonesia in which the breeding environment is different. This study aimed to obtain a hematology profile of Sprague-Dawley male rats aged 7 and 10 weeks. The animals were obtained from the Laboratory of Animal Experiment, NADFC. Blood was drawn from the retro-orbital sinus and then analyzed to obtain the hematology profile including erythrocytes, hemoglobin, hematocrit, MCV, MCH, MCHC, leukocytes. Statistically significant differences for erythrocytes, hemoglobin, MCV, MCHC, thrombocyte, and leukocytes parameters were observed between rats aged 7 and 10 weeks. However, there were no statistically significant differences observed on MCH, thrombocyte, and body weight parameters. Data presented here would be useful as a reference for hematology research as well as in evaluating alterations in blood parameters.Keywords: erythrocyte, hematology, leukocyte, Sprague-Dawley, thrombocyteABSTRAKSaat ini belum banyak tersedia informasi profil hematologi untuk hewan coba tikus yang dikembangbiakkan di Indonesia. Kebanyakan peneliti menggunakan data profil hematologi dari referensi luar Indonesia. Penelitian ini bertujuan untuk mendapatkan data hematologi tikus jantan usia 7 dan 10 minggu. Hewan coba yang digunakan adalah tikus jantan galur Sprague-Dawley yang diperoleh dari Laboratorium Hewan Percobaan-BPOM. Sebanyak 60 ekor tikus jantan yang terdiri dari 30 ekor usia 7 minggu dan 30 ekor usia 10 minggu diambil darahnya melalui sinus orbitalis. Selanjutnya, dianalisa jumlah sel eritrosit, hemoglobin, hematokrit, MCV, MCH, MCHC, leukosit. Hasil analisis statistik menunjukkan terdapat perbedaan signifikan antara tikus jantan usia 7 dan 10 minggu untuk parameter jumlah eritrosit, hemoglobin, hematokrit, MCV, MCHC dan leukosit. Tetapi tidak terdapat perbedaan signifikan untuk jumlah MCH, trombosit dan berat badan tikus. Data yang diperoleh dapat digunakan sebagai referensi penelitian yang berhubungan erat dengan profil hematologi dan penyakit kelainan darah lainnya.
Salmonella typhimurium, that cause typhoid fever in mice was used as a model to study typhoid fever in humans. This study is aimed to determine the immunogenicity of recombinant protein of Fim-C-S. typhimurium and specificity of antibody antirecombinant protein of Fim-C-S. typhimurium in ddY mice. The immunogenicity studies by ELISA indicated that recombinant protein of Fim-C-S. typhimurium evokes strong immune response in ddY mice, particularly in experiment group. The specificity evaluated by Western Immunobloting technique indicated that the antibody anti-recombinant protein of Fim-C-S. typhimurium can detect the 31 kDa of recombinant protein of Fim-C-S. typhimurium as an antigen. This data reported that recombinant protein of Fim-C-S. typhimurium is immunogenic and the antibody anti-recombinant protein of Fim-C-S. typhimurium is specific. The results of this study can be used as a model to develop recombinant vaccine candidate against typhoid.
Labu siam (Sechium edule (Jacq.) Sw.) merupakan tanaman sayur yang dapat digunakan untuk obat tradisional dan memiliki aktivitas sebagai antikolesterol, antihipertensi, antidiabetes maupun antioksidan. Penelitian ini bertujuan untuk menetapkan mutu ekstrak buah labu siam sebagai bahan baku obat yang berasal dari bahan alam. Standardisasi ekstrak buah labu siam dilakukan terhadap parameter spesifik dan non-spesifik menurut metode standardisasi yang dipersyaratkan. Hasil parameter spesifik ekstrak buah labu siam menunjukkan kadar sari larut air 2,74 ± 0,07%, kadar sari larut etanol 2,75 ± 0,05%, kadar senyawa fenolik 0,36 ± 0,04 mg EAG/g ekstrak dan kadar pektin (bobot equivalen 4.083,51 ± 635,30 mg/eki, kadar metoksi 0,35 ± 0,03% dan kadar galakturonat 2,00 ± 0,19%). Parameter non-spesifik ekstrak buah labu siam menunjukkan kadar air 7,62 ± 0,53%, susut pengeringan 4,49 ± 0,42%, bobot jenis 1,408 ± 0,001 g/ml, abu total 6,33 ± 0,15%, abu tidak larut asam 0,76 ± 0,14%, kadar logam berat As, Cd, Hg dan Pb tidak terdeteksi. Total cemaran mikroba ektrak 7,5 x 103 koloni/g dan angka kapang khamir 20* koloni/g. Berdasarkan data diatas disimpulkan ekstrak etanol 70% buah labu siam memenuhi persyaratan secara umum untuk ekstrak tumbuhan obat.
The member of Zingiberaceae, Curcuma aeruginosa Roxb. (C. aeruginosa) has been recognized as medicine for cancer. However, not enough effort has been invested on this herb as chemoprevention agent on cancer development. In this current study, we investigated the influence of C. aeruginosa extracts as a chemo-preventive agent in Wistar rat induced by 7,12-Dimethylbenz[a]anthracene (DMBA). As methods, female Wistar rats were treated with three C. aeruginosa extract doses (CA1: 40mg/ 200g BW; CA2: 80mg/ 200g BW; CA3: 120mg/ 200g BW) in the entire course of the experiment, and were induced with DMBA after one week of administering the extract. As comparison, commercial drugs containing Phyllanthus niruri L. were being used as positive control group, while the group given only DMBA represents the negative control group. Based on experiment, we found that the administration of C. aeruginosa during experiment promotes an increasing in TNF-α, IL-2, and IL-12 levels. The dose of 80mg/ 200g BW (CA2) appeared to be best potential dose as chemo-preventive agent represented the smallest tumor incidence and tumor multiplicity at the end of the experiment. Moreover, this plant also efficient to eliminate the cancer cells at the beginning of the tumor until the metastatic phase.
